Jelinek, R. Monitoring of Company Infrastructure using Zabbix Tool with Integra-tion of ELISA Log Manager. Master's Thesis. Brno: Mendel University in Brno, 2023. This thesis deals with the creation of a monitoring system for a company's infrastructure. The monitoring system is implemented using the Zabbix monitoring tool, configured according to the organization's requirements. Furthermore, log files are analyzed using the ELISA log manager, and the logs are subsequently evaluated based on created rules. As part of this thesis, the integration of these two systems was configured to enable monitor alerts sent from the ELISA system within the Zabbix interface.
Expression and binding properties of human galectin-4

Galectin-4, a member of the galectin family known for its affinity to β-galactosides, belongs to tandem-repeat galectins and consists of two distinct subunits connected with a peptide linker. Galectins play a key role in various biological processes and are involved in the pathogenesis of many diseases. Colorectal cancer (CRC) tissue shows a lower expression of galectin-4 compared to healthy colon tissues, and the lower expression of galectin-4 promotes tumor progression and transport. Galectin-4 was found to inhibit tumorigenesis of CRC cells via Wnt/β-catenin and IL-6/NF-κB/STAT3 signaling pathways, which play a key role in CRC development. In inflammatory bowel diseases such as Crohn's disease and ulcerative colitis, galectin-4 interacts with CD4+ T lymphocytes, influencing their behavior in the immunological synapsis. Furthermore, galectin-4, primarily expressed in the gastrointestinal tract, participates in the apical transport in epithelial cells, promotes the healing of intestinal epithelial wounds, participates in the adaptive immune response and has multiple roles in the central nervous system. Methods: Selected cytokines were determined using Quantikine® HS ELISA Human IFN-γ Immunoassay, Quantikine® HS ELISA Human IL-4 Immunoassay and Quantikine® ELISA Human IL-5 Immunoassay. The monitored population consisted of 32 polinotics, 19 of which were administered allergen immunotherapy and 13 of which constituted the control group. Preparation and characterization of chicken antibodies against pathogenic yeast Candida albicans

The increase of infections caused by yeast pathogens is an alarming global public health problem. People with compromised immune systems, such as after infection with human immunodeficiency virus or diabetes mellitus, are most at risk. Diagnosis is often inadequate, and treatment relies on a limited selection of antifungal drugs, to which resistance is also increasing. These aspects represent a major problem, as second-choice drugs, which may have toxic side effects (e.g., nephrotoxicity in the case of amphotericin B), must then be applied to resistant pathogens. Given the severity of yeast infections, scientists are trying to find new ways of protection against these pathogens. One option is passive immunotherapy. In this bachelor thesis, antibodies from hen's eggs were chosen for this purpose, as they have the potential for wide therapeutic use. The preparation of hen's egg yolk IgY antibodies is both ethically and economically acceptable. Moreover, egg yolk antibodies show relatively high stability. Polyclonal antibodies were isolated from eggs of hens immunized with either cell wall or whole C. albicans cells in a medium simulating the vaginal environment (VSF). Monitoring of gluten content in barley varieties recommended for the production of Czech Beer

Sandwich enzyme-linked immunosorbent assay ELISA was used for determination of gluten in barley varieties recommended for the production of Czech beer. Currently, it is the most widely used method for determining gluten in the food industry. Monitoring the gluten content in raw materials and foods made from them is especially important for people suffering from celiac disease. Gluten intolerance is an autoimmune disorder that causes morphological and functional changes in the small intestine. Currently, the most effective treatment is following a gluten-free diet. In this bachelor thesis, content of gluten in 14 barley samples was monitored. The RIDASCREEN® Gliadin set was used for the measurement. Results were evaluated with RIDA SOFT Win.NET software. Content of gluten in the barley samples varied from 36,3 g/kg to 82,5 g/kg.
Chicken antibodies against the pathogenic yeast Candida albicans

5 Abstract Candida albicans belongs to the most widespread pathogenic yeasts. As an opportunistic pathogen, it mainly colonises the mucous membranes of the host and causes infection, especially in individuals with a compromised immune system. The most common infection caused by C. albicans is vaginal mycosis, which at least 3 out of 4 women will experience during their lifetime. Antifungal drugs are used to treat this condition, but the yeasts are becoming less susceptible. For this reason, there is an effort to develop new treatment options for (not only) infections caused by C. albicans. Although vaginal mycosis is not a severe condition in most cases, it is a widespread infection that occurs repeatedly in many women, sometimes several times a year. Since vaginal mycosis reduces the quality of life in many women, we decided to focus on this disease in this paper. This project used hen antibodies, which have a number of advantages over mammalian antibodies. In oviparous animals, antibodies are not only found in the blood serum, as seen in mammals but are also secreted into their eggs. This makes the isolation of hen antibodies non-invasive and ethical. Karolína Jankovičová, Ph.D. Title of diploma thesis: Monitoring of sublingual allergen immunotherapy using serum levels of allergen specific IgG antibodies The first aim of the thesis was to evaluate the clinical course of sublingual allergen immunotherapy (SLIT). Another aim was to assess the changes in antibody response over time and to compare the results of the Oralair treated group with the control group. Last but not least, the aim of the thesis was to evaluate the changes in the levels of specific IgG antibodies in relation to the clinical course of SLIT. The visual analogue scale (VAS) was used for the clinical assessment of allergic symptoms. A commercially available ELISA method was used to determine sIgG4 against grass pollen allergens. sIgG1, sIgG2 and sIgG3 were determined by an in-house developed and validated ELISA. Based on the results, statistical analysis was performed. It was found that Oralair therapy was clinically effective from the first season of the treatment. Porovnání metod stanovení obsahu mykotoxinů v obilninách

In the Bachelor thesis I have dealt with a comparison of methods for determination of mycotoxins in cereals. The first part is a recent survey, in which I described the distribution of mycotoxins, I described the toxigenic fungi and produced mycotoxins. I noted the characteristics of the best-known mycotoxin and described some of the analytical method for the determination of mycotoxins. In the second part I focused on analysis of cereal samples. A total of 12 samples were analyzed by parallel analysis, that should determine the accuracy of the measurement. Analysis was performed using three methods of analysis, and ROSA for ZEA and DON, Reveal Q + for DON DON DON RAPID, Veratox for ZEA and the reference method was HPLC method. Difference was observed in the parallel measurements, the results of mycotoxins analyzed by various methods differed. We can not say which method is more accurate.

This thesis deals with the issue of mycotoxins, which are important pathogens of many agricultural crops and may pose a high health risk to humans and livestock. It contains an overview of the most important mycotoxins their producers and mycotoxicoses. The main focus of this thesis is the occurrence of mycotoxins in cereals and the effect of processing it on their content. The experimental part focuses on monitoring the content and the penetration of deoxynivalenol and zearalenone in winter wheat from grain to bread. We examined a total of 10 samples of treated and 10 samples of untreated variants of winter wheat. For each sample grain, flour and bread were examined for DON and ZEA content and all tested samples were positive for DON and ZEA. None of the examined samples exceeded or even approached the legislative limits.
