Vladimíra Řezáčová, Ph.D. Název práce: Monitorování průběhu sublingvální specifické alergenové imunoterapie pomocí sledování hladin vybraných cytokinů Klíčová slova: alergenová imunoterapie, cytokiny, ELISA technika, alergie na pyly trav Cíl práce: Cílem této diplomové práce bylo zjistit, zda dochází v průběhu alergenové imunoterapie k detekovatelným změnám hladin IFNγ, IL-4 a IL-5 a zda by tak tyto cytokiny mohly sloužit jako biomarkery účinnosti této léčby. Metody: Vybrané cytokiny byly stanoveny pomocí metod Quantikine® HS ELISA Human IFN-γ Immunoassay, Quantikine® HS ELISA Human IL-4 Immunoassay a Quantikine® ELISA Human IL-5 Immunoassay. Sledovaný soubor se skládal ze 32 polinotiků, přičemž 19 z nich byla podávána alergenová imunoterapie a 13 z nich tvořilo kontrolní soubor. V rámci sledování také obě skupiny průběžně zaznamenávaly tíži svých symptomů pomocí vizuální analogové škály. Výsledky: U skupiny léčených pacientů byl v průběhu léčby pozorován pokles koncentrací IL-4, IL-5 i IFNγ, avšak mezi pacienty léčenými přípravkem Oralair a kontrolními jedinci nebyly v průběhu léčby nalezeny významné rozdíly hodnot těchto cytokinů. Vladimíra Řezáčová, Ph.D. Title of thesis: Monitoring of sublingual allergen immunotherapy using serum cytokine levels Keywords: allergen immunotherapy, cytokine, ELISA, grass pollen allergy Background: The aim of this thesis was to find out whether there are detectable changes in the levels of IFNγ, IL-4 and IL-5 during allergen immunotherapy and whether these cytokines could thus serve as biomarkers of the efficiency of this treatment. Methods: Selected cytokines were determined using Quantikine® HS ELISA Human IFN-γ Immunoassay, Quantikine® HS ELISA Human IL-4 Immunoassay and Quantikine® ELISA Human IL-5 Immunoassay. The monitored population consisted of 32 polinotics, 19 of which were administered allergen immunotherapy and 13 of which constituted the control group. As part of the follow-up, both groups also continuously recorded the severity of their symptoms using a visual analog scale.
