Study of biological activity of alkaloids isolated from Fumaria officinalis L. (Fumariaceae) II.

Diploma Thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, Hradec Králové 2014, 49 pp. Obtained diethylether extract of Fumaria officinalis L. was separated to fractions in column chromatography with petrol, chloroform and ethanol. Preparative TLC and crystalisation led to isolation of five alkaloids from fraction. Alkaloids were identified by GC-MS and NMR specters, optical rotation and melting point as protopine, cryptopine, (-)-fumaricine, (+)-fumariline and (+)-parfumidine. Isolated alkaloids were tested for their inhibition activity towards acetyl- and butyrylcholinesterase and towards prolyloligopeptidase. Activities were compared with standards. Natural inhibitor galanthamine showed IC50 AChE 1.710 ± 0.065 µM, IC50 BuChE 42.30 ± 1.30 µM. Best inhibition activity showed protopine (IC50 AChE 345.4 ± 24 µM, IC50 BuChE 239.6 ± 22.3 µM) and cryptopine (IC50 AChE 477.71 ± 47.33 µM, IC50 BuChE 270.82 ± 39.12 µM). The highest prolyloligopeptidase inhibition activity showed (+)-parfumidine with IC50 POP 99.2 µM, which was more active than used natural inhibitor baicaline (IC50 POP 605.9 ± 0.021 µM). Vítovcová, A.: Biological activity of secondary plants metabolites IV. Alkaloids of Vinca minor L. Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, Hradec Králové 2016. Extract of Vinca minor L. was separated to fractions in column chromatography. Flash chromatography, preparative TLC and crystalisation led to isolation of two alkaloids from fraction number two and number five. Alkaloids were identified by GC/MS as (+)-vincaminorine and vincorine. These alkaloids were tested for their inhibition activity towards cholinesterase (AChE and BuChe). Obtained activities IC50 were compared with standards (galanthamine, huperzine A). The most interesting activity against galanthamine (AChE 1,710 ± 0,065 µM, BuChE 42,30 ± 1,30 µM) and huperzine A (AChE 0,033 ± 0,001 µM, BuChE > 1000 µM) showed vincorine (AChE > 1000 µM, BuChE 9,75 ± 0,45 µM). His activity toward BuChE is higher than the activity of both standards. Inhibition activities of (+)-vincaminorine (AChE 746,5 ± 84,13 µM, BuChE 684,32 ± 70,66 µM) are negligible. Validation of an adventure programme with cooperative elements in school P.E. lessons Summary of the thesis The thesis deals with validating an innovative motor program with adventure and cooperative elements at a Czech elementary school. The aims of the thesis was to observe possible changes under the influence of the programme in social acceptance of children in class, in their self-concept and the amount of physical activities in their leisure time. The study was a quasi-experiment, concretely a cross classification with repeated testing. It was realized in classes of 6th , 7th and 8th grade (12-14 year old boys) (N=154) in physical education lessons. The research was realized during 6 month, intervention time was 3 months and data were gained in three time intervals. We have used the Social acceptance questionnaire and Self-concept and quality of life questionnaire as main research methods. Results brought two statistically significant values, however, in practical significance they explain a small number of results. Values in the sub-scale Pupil-classmates relationship were statistically significant (p=0,031), which explained 3% of results, and values in the item Evaluation of satisfaction with factors effecting self-concept had the statistical significance p=0,002 explaining 6,5% of results. Determination of Copper in Food Supplements by AAS

EN This bachelor thesis deals with the determination of copper in the form of copper ions in selected food supplements by atomic absorption spectrometry. The first step of the experiment was to find the optimal measurement conditions for the parameters: beam height above the burner edge, burner horizontal position, fuel flow rate, burner angle and spectral interval width. Subsequently, the calibration dependence was determined and the basic measurement characteristics were calculated: repeatability, sensitivity, limit of detection (0.019 mg dm−3 ) and limit of quantification (0.065 mg dm−3 ) at the burner position 0 ř. Interference studies were performed for the presence of iron, manganese and sodium elements in selected food supplements because of theirs increased concentrations. These selected food supplements were analysed: Spektrum 50, Supradyn Energy Complex, Center from A to iron, GS Extra Strong Multivitamin, Caltrate Plus, DAS Gesunde Plus, A-Z Multispektrum and Calibrum 50 Plus. A comprehensive approach of emergency medical services providers to a transport of a patient suffering from a highly contagious disease

In the thesis "Comprehensive approach of emergency medical services provider to a highly-contagious patient transportation" we concentrated on nursing care and aspects of transportation administered by operating paramedics, all in cases that public health authorities suspect to be related to a highly - contagious disease and transportation in isolated transportation vehicle is mandated. The theoretical part of the thesis defines the key concepts of highly - contagious disease, it describes individual causal agents of contagious diseases and their ways of transmission to humans. For description of contagious diseases were chosen those with high morbidity, lethality and interpersonal transmission. We mention the danger of biological agents misuse, ways of protection against contagious diseases and types of decontamination procedures. We describe different means of personal protection used by biohazard team members and even environmental factors affecting the work of paramedics using personal protective agents. Further on, we bring to attention legal aspects and inner policies related to aforementioned issues including analysis of procedure logistics of emergency services teams dealing with emergencies with suspicion of highly - contagious disease. Research in practical part of the thesis concentrates on aims related to optimizing nursing care for patients suspected of suffering from a highly - contagious disease, when securing them in the transportation vehicle, furthermore, we examine the impact of personal protective agents used by paramedics when transporting the patient, and we also analyze environmental factors inside the ambulance vehicle and isolated personal transportation vehicle. All of these aims were researched with the help of planned experiments and the resulting findings are based on the outcomes of these experiments.
Voltammetric Determination of Tumor Biomarkers using Flow Injection Analysis with Amperometric Detection

Three tumor biomarkers (homovanillic acid, vanillylmandelic acid, and 5-hydroxyindole-3-acetic acid) have been determined by flow injection analysis with amperometric detection at screen-printed carbon electrodes in optimum medium of Britton-Robinson buffer (0.04 mol l(-1), pH = 2.0). Dependences of the peaks current on the concentration of biomarkers were linear in the tested concentration region from 0.05 to 100 mu mol l(-1), with the limits of detection of 0.065 mu mol l(-1) for homovanillic acid, 0.053 mu mol l(-1) for vanillylmandelic acid, and 0.033 mu mol l(-1) for 5-hydroxyindole-3-acetic acid (calculated from heights), and 0.024 mu mol l(-1) for homovanillic acid, 0.020 mu mol l(-1) for vanillylmandelic acid, and 0.012 mu mol l(-1) for 5-hydroxyindole-3-acetic acid (calculated fromareas), respectively.
Alkaloids of Papaver rhoeas L. (Papaveraceae) and their biological activity related to Alzheimerʼs disease II.

Čakurdová, M.: Alkaloids of Papaver rhoeas L. (Papaveraceae) and their biological activity related to Alzheimer's disease II. Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, Hradec Králové, 2017. The aim of this work was focused on isolation of alkaloids from the fraction 1 (joined fractions 20-27), which was obtained from the summary alkaloid extract of Papaver rhoeas L. (Papaveraceae). The alkaloids were identified by structural analysis NMR, GC-MS, optical activity and melting point. Two alkaloids were identified as (-)-stylopine and (+)-rhoeadine. Isolated substances were tested on ability to inhibit the enzymes acetylcholinesterase, butyrylcholinesterase and prolyloligopeptidase. Obtained data were expressed as IC50 values: (-)-stylopine (IC50 AChE = 522 ± 67 µM, IC50 BuChE = >1000 µM, IC50 POP = >790 µM); (+)-rhoeadine (IC50 AChE = 915 ± 64 µM, IC50 BuChE = >1000 µM, IC50 POP = >790 µM). None of the isolated substances showed so potent cholinesterase inhibitory activity as the alkaloids galanthamine (IC50 AChE = 1.71± 0.065 µM, IC50 BuChE =42.30 ± 1.30 µM) and huperzine A (IC50 AChE = 0.033 ± 0.001 µM, IC50 BuChE = >1000 µM) or rivastigmine (IC50 AChE = 0.037 ± 0.001 µM, IC50 BuChE = 3.3 ± 0.3 nM)....

One of the main roles of Human resource managers is the knowledge of each job position and its relevant role within the company, its creation, its current state and the future employees' character for the job fulfilment. The recruitment process is directly linked to the number of vacant positions as well as to its characteristics. Recruiting policies follow a set pattern, described in the literature, as a key process to allocate, approach and finally recruit the best that the human resource market offers. Motivation and education have proven as a necessary component of employee satisfaction and it enhances the unique abilities of each person. However, the research and in-house surveys show, that lack of communication in all levels of hierarchy is a major obstacle in self-development and keeping to the company's targets.
