Authenticity of natural plant component in cosmetics products

The purpose of this thesis was to study the authenticity of selected natural ingredients in cosmetic products. These were specifically cosmetic products that contained citruses, mint or lavender. Commercially available isolation kits were used for DNA isolation. The presence of plant origin DNA was verified by PCR method using primers specific for the ITS2 region of plants. The presence of limonene, a significant allergen contained in said plants, was determined in the samples by PCR method using primers for limonene synthase. At the same time, its presence was verified by HPLC method. In addition, two primers were tested for lavender and monitored for their efficacy, with no significant difference in the usage.

V súčastnej dobe sa veľká pozornosť zameriava na biotechnologický priemysel. V rámci biotechnologického priemyslu sa pomocou mikroorganizmov produkujú rôzne užitočné látky, akými sú napríkald chlorofyl a karotenoidy, ktoré sú významnými antioxidačnými látkami, ale aj mastné kyseliny, steroly a ubichinón. Táto diplomová práca je zameraná na kultiváciu vybraných kmeňov mikrorias rodu Scenedesmus a Desmodesmus. V prvej časti práce sú opísané kmeňe rias a študované metabolity. V druhej, experimentálnej časti, boli na vybrané kmene aplikované rôzne druhy alkoholov, ako stresové podmienky, s cieľom zvýšenia produkcie intracelulárnych metabolitov. Ďalej bola prevedená fermentorová kultivácia, pri ktorej bola aplikovaná kombinácia dvoch rôznych alkoholov. Prevedená bola tiež kultivácia pomocou flat – panel fotobioreaktoru, pri ktorej bolo použité svetlo s rôznymi vlnovými dĺžkami. Pre analýzu boli použité viaceré chromatografické techniky, výsledky analýz sú uvedené v experimentálnej časti.
