Physiological functioning of preptin and its analogs

Preptin is a 34 amino acid long peptide that is derived from pro-IGF2, where it is cleaved from its E-domain and produced together with the matured IGF2. In my thesis we prepared two full-length preptins (mouse and human) and 8 preptin analogues (fragments with or without olefin metathesis). The full-length preptins as well as the preptin fragments were synthesized by solid-phase peptide synthesis. Mouse or human preptin served as our reference molecule for comparison with the results of bioassays of preptin analogues. All substances were tested for different biological activities to further elucidate the effect of preptin as a possible biologically active substance. We tested the effect on insulin secretion in pancreatic β-cells, indirectly in- vestigated the effect on bone cell proliferation and tried to approach the discovery of a possible receptor for preptin. From all our results we found that although preptin or some of its analogues have a higher effect than controls, the results are not consistent and the effect is not significantly higher. We were unsuccessful in our search for a receptor, despite having done a number of saturation experiments with radiolabelled preptin in many cell types. Defence of the obliged party in the execution (deferral and discontinuance)

The topic of this rigorous thesis is the defense of the debtor in execution with the focus on institutes deferral and discontinuance of execution. The thesis concerns execution carried out by the judicial officers according to Act No. 120/2001 Coll., on court executors and execution activities (Execution Procedure Code). The aim of this thesis is to describe and evaluate the possibilities of debtor's defense, to analyze application problems and to propose a modification de lege ferenda. I refer to the conclusions of the judicial decisions to individual institutes, which is an important source for the interpretation of legal rules. This rigorous thesis consists of four chapters, which are further separated into individual subchapters. The first chapter is devoted general interpretation of term and purpose of execution, sources of law and main principles of execution proceeding. This chapter also includes a historical excursion to the development of execution law in our country and an example of a foreign procedure in the exaction of claims. The main principles of execution proceeding are explained in detail, including examples of specific provisions of a statute in which these principles are expressed. IGF2 and preptin, biologically active peptides derived from pro-IGF2

Pro-IGF2 is a 156 amino acid long protein and it is a prohormone from which two bio- logically active peptides are formed, IGF2 and preptin. IGF2 is an insulin-like growth factor 2 that, together with IGF1 and insulin, shares a similar 3-D structure and similar biological effects. It is a peptide whose sequence corre- sponds to amino acids 1-67 of pro-IGF2. It is produced mainly by the liver, but it is formed in most cells of our body. It is involved in the regulation of growth, differentiation, embryogenesis and in recent years the interest in IGF2 has been increasing mainly due to its function in the central nervous system. Preptin is a newly isolated 34 amino acid long peptide whose sequence corresponds to amino acids 69-102 (E-domain) of pro-IGF2. Preptin affects glucose metabolism as well as bone homeostasis. Disorders in glucose metabolism usually lead to diabetes mellitus, and disorders in bone homeostasis most often lead to osteoporosis. Preptin has been shown to increase the second phase of insulin secretion and to have a positive effect on the proliferation and differentiation of osteoblasts, bone cells promoting bone growth. The animal experiments seem to be current theme of these days. This thesis is focused on using animals, especially rodents, in the Czech Republic for the last ten years. There is also emphasis on the legal principles of animal protection and observance of three "R" principles. In this study there were used data from available sources about using laboratory animals in the Czech Republic. Also there was prepared a questionnaire research in different scientific institutions which use animal models in their researches. There were found out from available sources that the amount of experimental animals is on the same level (about 230 000) during the years 2006-2015. There were no deflection. Also this fact is the same for using rodents (about 120 000) as biomodels. Further there was found out that the laboratory mouse (Mus musculus var. alba) is the most used experimental model. 66,7% of questioned respondents considered the laboratory mouse the most common used laboratory animal and 60% of questioned respondents supposed that the number of used laboratoty animals was decreasing.
Evaluation of selected postural stability parameters of diabetic patients

Title: Evaluation of selected postural stability parameters of diabetic patients Objectives: The main objective of this thesis is to describe the level of postural stability using the selected parameters in diabetes mellitus type 2 and to try to find a relationship between the severity of disease. We also want to compare the level of postural stability for such an illness with respect to the values of selected parameters of the healthy population. Methods: It is a descriptive study. The theoretical part was the basis of the processing problems of diabetes mellitus, with a focus on neuropathy. After that we evaluate data of selected parameters of postural stability in diabetes mellitus type 2. The second part involves the experiment, which refers to the evaluation of this issue in terms of posturographic analysis. A group of 30 type 2 diabetic patients were divided according to their diabetologist disease severity into 4 groups. A control group of healthy subjects included a sample of 7 probands. The patient group was composed of 18 men and 12 women, whose average age was 62.63 ± 15.55 years, average weight 91.17 ± 19.97 kg, who were subjected to measurement using a pressure plate FootScan (RScan International, Belgium). The thesis deals with theoretical knowledge process of training employees in the organization and their personal development, as well as a practical approach to the process of educating city workers Rakovník working at the Municipal Office in Rakovník, which is the authority of a municipality with extended powers in this range ensures the execution of state administration. Review of literature in the first part characterizes human resource management, human personality and describes in detail the process of lifelong learning and personal development, motivation and corporate training. The second part is defined by the statutory procedure for training of local government officials. The practical part consists of a description of the organization Rakovník Municipal Office, its activities and scope, as well as familiarity with the organizational structure. Based on the study of documents, observation and questionnaire survey, which solves view officials on the issue of their further education, an assessment of the level of this process in the organization. In conclusion processed suggestions for possible improvements.
Educational materials for geological exposition of Nature Science Museum "Help for nature" in Tyn nad Vltavou

The purpose of the presented bachelor thesis is to develop a tutorial programme which would acquaint students with the topic of rock cycle. The final versin of the programme and students'worksheet was based on test-students knowledge and skills. Using analyzes of the educational program framework for primary education, textbooks and didactic materials from the Nature Science Museum of Semenec in Tyn nad Vltavou was created worksheet which have become the foundation for tutorial program of the rock cycle. The worksheet was tested in four classes and modified into the final form. This worksheet will be used in the Nature Science Museum of Semenec. Part of this work is to create a methodological sheet for teachers. The benefit of this work is the development of new educational program with worksheet on the theme of rock cycle.

Physiotherapy and treatment of elbow enthesopathy

The topic of my work with the name ?Physiotherapy and Treatment of Enthesopathy in the Ulnar Joint? is very current at present. A large part of the population suffers from it, in the consequence of excessive loading. Very often the first symptoms, which could prevent the problems, are not taken into account. It often starts after single hard work (computer typing, wood chopping, etc.), and that acute form very often changes into a chronic form. The treatment of such problems is usually very protracted. The choice of the treatment is difficult as well and no method of a correct procedure of therapy has been described yet, because each individual reacts in a different way. It is not possible to estimate how long it will take, before the everyday activities could be resumed. As the aim in the theoretical part of my work I chose to study and apply the current available information from various resources and thus create an educational material about the application of suitable treatment. The basic anatomy of ulnar joint, kinesiology, the most often factors and reasons of the origin are described there. Further there were special methods of examination and a clinical picture, therapy and prevention against the problems. For the practical part I chose a method of a qualitative research. Three cases of patients, who were treated by MUDr. Kunovsky in Tabor and who underwent physiotherapy there, have been studied and dealt with. The research was carried out by means of observation, anamnesis and discussions during the treatment, the documentation of the patients and special examination methods. The treatment methods I have focused on were mobilization, technique of soft tissues, pro-prioceptive neuromuscular facilitation, physiotherapy and taping technique. Some forms of applications had a positive effect on the patients and brought improvement, the others showed no changes. In my opinion the individual elements should be combined and applied for a longer period of time and the anamnesis of the patient should be thoroughly discussed, to find out the main reasons of the origin. The patients learned a procedure during the therapy how to face correctly the origin of that problem and how to apply efficiently the preventive measures to avoid the recurrence of the problems. My work could be used for the purpose of studies of future physiotherapists, for the patients, or a source of information for public.

In my diploma thesis I have dealt with motivation and satisfaction of employees in the company PRAGA Louny CZ a.s. The company PRAGA Louny CZ a.s. faces high level of workers' fluctuation. The aims of my diploma thesis were to find out if the employees think about the leave of the company, what are the factors that cause their un-satisfaction, what could make their satisfaction better, what factors positively affect their motivation and in the end to propose possible steps that could make the present situation in the company better.
