Ph.D. Title of diploma thesis: Study of the cytostatic effects of sulfur mustard Sulfur mustard (HD) belongs to blistering agents used in chemical warfare. It is a bifunctional alkylating agent that covalently modifies DNA. The cytostatic effect of HD is characterized by halting cell division without cell death. The duration for which a cell can remain in this state depends on the extent of DNA damage, which can subsequently be converted into a cytotoxic effect leading to cell death. The aim of the study was to determine the concentration range of HD gas that exhibits cytostatic effects and to investigate the sensitivity of HaCaT skin keratinocyte cells synchronized in the G1, S, and G2/M phases of the cell cycle to alkylating damage induction following release from cell cycle blockage. The cytostatic effect was observed only at very low concentrations, in the range of units of µmol.l-1 . In our experiments, a concentration of 2 µmol.l-1 of HD prevented cell proliferation for 2 days. The orphan drug Epidyolex is indicated to reduce the frequency of seizures in these diseases. The aim of this work is to provide a comprehensive overview of orphan drug legislation and the potential of its reimbursement from health insurance and determine the costs of DS and LGS from a societal and governmental perspective. Methods: The data for the analysis is obtained from the questionnaire focused on the social benefit drawdown, increase in household costs, and loss of productivity of patients and their caregivers. Respondents are recruited in a cooperation with patient organisations and specialised centres. Taxation of legal persons

The bachelor thesis deals with the possibilities of optimizing the corporate tax. The thesis is divided into two main parts - theoretical and practical. The first part explains all the basic concepts related to corporate income tax. In the practical part, a specific legal entity is introduced and suggestions leading to a reduction of the entity's tax burden are described. The result is a comparison of the current situation with the proposal and a quantification of the overall savings.
Index Suitable for Similar Search in High-dimensional Spaces

In this paper, we focus on indexing and searching in high-dimensional data. To achieve the target we implemented the Metric Index, a model of the similarity search based on the metric spaces, that employs many of known principles of partitioning and filtering. The metric space is a general model of similarity, which enables the usage of implemented index for various data. With this index, stored data could be searched effectively. The internal structure of data is hidden, we just require an implementation of the function for feature extraction, which produces a vector representing data, and the metric function applicable to the given data. The Metric Index was implemented as a data cartridge, the mechanism for extending the capabilities of the Oracle server. This data cartridge enables indexing of large unstructured data in the Oracle server known as LOBs.
A tool for testing of algorithms for learning languages

Goal of this work was to develop a tool for testing algorithms of grammatical inference of context-free languages. Grammatical inference is a process of learning of grammars and languages from data. Learning could mean finding a suitable model that describes data. Due to this model we could for instance compress this data, create new data or find out which data are consistent with this model. The model in our tool is context-free grammar. We describe how to generate the context-free grammar and data from which the algorithm can try to learn the language of this grammar. The problem of context-free grammar complexity was solved in order to evaluate success achieved by an algorithm in different learning tasks. Also some alternatives of evaluating success of an algorithm are described. This tool is also useful to create data for publishing results of the algorithm.
Drop-out of the beginning teachers

The Diploma thesis focuses on the drop-out of beginning teachers. Theoretical part elaborates on the phenomenon of drop-out, it describes the reasons why the beginning teachers leave and why they are the group of teachers which is the most threatened by drop-out. Furthermore, this part describes problems that beginning teachers have to deal with and the most frequent reasons for leaving the profession, their rate and consequences of drop-out. The end of the theoretical part presents options and suggestions that would help solve this problem. The empirical part of the thesis is realized by an on-line questionnaire and in-depth interviews. The research focuses on four main aspects: 1 - the motivation to study the teaching and to practise it, 2 - the main reasons for new teachers to leave, 3 - the possibilities to prevent drop-out and 4 - whether the respondents continue to work with children or not. Last but not least, the research focuses on the possibilities that would help respondents to stay in the profession. The results of the research confirm the reasons for quiting as described by literature. The qualitative description of the reasons for drop-outs provides a deeper understanding of how they interact in an institutional, interactive and cultural level.
