Synthesis of NIR Reductive Probes to Monitor Hypoxia During Fungal Infection

Invasive fungal infections represent a significant problem, resulting in approximately 1.5 million deaths worldwide annually. Among the most common invasive fungal infections are aspergillosis and candidiasis. Both opportunistic infections have a high mortality rate (50% and 27%, respectively). The diagnosis of these infections is often complex, leading to delayed detection and an increased risk of complications. Recently, near-infrared (NIR) hypoxia probes have been investigated for the diagnosis and monitoring of fungal infections. These probes enable non-invasive measurement of tissue oxygen concentration and can contribute to early diagnosis of fungal infections. Hypoxia can also be a relevant target for the delivery of antifungal agents. For this reason, a series of hypoxia-selective probes has been developed. These probes contain N-methyl-2-nitroimidazole and a fluorophore that absorbs in the near-infrared spectrum. We anticipate that probes will function as follows: the reducible breakdown of N- methyl-2-nitroimidazole in a hypoxic environment will activate the probe, leading to the release of the fluorophore, which absorbs in the near-infrared spectrum. Pathological personality structure in the stories of clients treated for addictive substance abuse in a chosen Highlands facility Abstract This thesis deals with the issue of personality disorders, termed in the title as pathological structures of personality, of people treated for addictive substance abuse. Numerous Czech and foreign studies pointing out high prevalence of personality disorders with substance abusers served as a base. I also proceeded from the finding that little was written in theory or dealt with in practice concerning comorbidity of abovementioned issues. The theoretical part is divided into three parts. The first part develops the topic of personality disorders, chosen types of addiction and the unifying topic of psychiatric comorbidity. The aim of the thesis is, by analyzing two case studies, to describe the possible manifestations and treats of specific personality disorders, to observe the way they affect their life or the development of the illness connected to substance abuse and the part they took in their relapses or in their cooperation with the services for substance abusers. I reached the goals of this thesis by the methods of qualitative research, using a semi-structured interview that was later on transformed into a case history. Synthesis of model, self-immolative, phosphate-based linkers for delivery of oximes

This bachelor thesis explores a possibility of preparation and usage of oxime prodrugs. Thesis aim is to deliver oximes by using self-immolative phosphorous-based linkers that undergo cyclization reaction, leading to the release of oxime from the phosphorous. For this purpose, model systems containing: (1) a photolabile DMNB group, or they could contain enzymatically activable ester group, (2) self-immolative arm with the phosphate core, and (3) oxime as the leaving group, were prepared. Acetophenone oxime, cyclohexanone oxime, and griseofulvin oxime were used as the model oximes. In particular, the synthesis of target molecules and their self-immolation (i.e., controlled breakdown) have been studied. These reactions were monitored by 31 P NMR spectroscopy. THE ABSTRACT OF THE DIPLOMA THESIS The Name of the Diploma Thesis: The motivation to lose the habit of smoking in patients terated for some other kind of addiction Abstract: Starting point: The prevalence of smoking in people addicted to other narcotics is usually 2 - 3-times higher than in common population. The same percentage of people addicted to tobacco is among patients treated in some in-patient treatment for people addicted to narcotics or gamblers. Simultaneous treatment of addiction to tobacco and other kinds of addiction is possible and is recommended. It seems to be also effective and favourable for treatment of other patient's addictions - it supports the improvement of physical and mental condition. The interest in treatment of addiction to tobacco often appears in patients. But the question is how much they are motivated and informed by the staff. There is no integrated schedule focused on breaking the habit of smoking in in-patient clinics for addiction treatment. Targets of Thesis: To discover the rate of addiction to nicotine in patients treated for some other addiction.
