Compact equipment for mechanical durability testing air circuit breakers
Dušek, Pavel ; Šedivá, Soňa (referee) ; Štohl, Radek (advisor)
This master's thesis is focused on the description of mechanical durability testing of low voltage circuit breakers, description of circuit breakers Sentron 3WL and 3WA and the design of own solution for mechanical durability tests of Sentron 3WL and 3WA air circuit breakers. The solution itself consists of selecting components, designing the electrical schema, creating block diagrams of the program structure, creating the necessary software and verifying the functionality of the equipment.
Testing equipment for function test of automatic standby unit
Dušek, Pavel ; Pásek, Jan (referee) ; Štohl, Radek (advisor)
This bachelor’s thesis is focused on the description of automatic transfer switch and designing a tester for automatic transfer switch MODI ZA made by OEZ s.r.o. The solution itself consists of creating the state diagrams of the function test steps, programming software for Simatic S7-1S200 PLC and Simatic KTP 400 control panel and creating a technical documentation containing the tester wiring diagram, and layout of the insides of the switchboard. It also contains the diagram explaining how to connect the automatic transfer switch MODI ZA to the tester.
Functional and Pathophysiological-morphological Correlates of Neurodegenerative Diseases
Dušek, Pavel ; Roth, Jan (advisor) ; Baláž, Marek (referee) ; Menšíková, Kateřina (referee)
This doctoral thesis pictures neurodegenerative diseases as a multilevel process, describes various correlates on each pathophysiological level, and presents selected correlates in Huntington's disease and mitochondrial membrane protein-associated neurodegeneration (MPAN). It uses various methodological approaches such as basic laboratory research, clinical work, imaging, database formation, and database summary. Changes in the amount of respiratory chain complex I and respiratory chain complex IV in buccal ep- ithelial cells of Huntington's disease patients are described. The insufficient power of optical coherence tomography as a biomarker in Huntington's disease is demonstrated. Various phenotypes of MPAN are summarized, and an association between C19orf12 mutation and visual impairment is confirmed. A phenotype of a well-documented case of MPAN is presented. Keywords: C19orf12 mutation; color discrimination; contrast sensitivity; huntingtin; Huntington's disease; iron accumulation; mitochondrial membrane-protein associated neurodegeneration; multilevel process; neurodegeneration; optical coherence tomogra- phy; parkinsonism; respiratory chain complex; retinal nerve fiber layer thickness

Activity of the cathepsin B in patient with the bladder cancer
Dušek, Pavel ; Babjuk, Marek (advisor) ; Soukup, Viktor (referee) ; Broďák, Miloš (referee)
Bladder cancer is the most common malignant neoplasm of the urogenital tract. Of non- invasive diagnostic methods in clinical practice normally only is used cytology to its limits. While there are several commercially available tests that have the character of a tumor marker. None of them, however, has not yet been recommended for the standard diagnosis or monitoring of patients with bladder cancer. Cathepsin B is a thiol - dependent protease found in normal circumstances in the lysosomes of mammalian cells. Cathepsin B are lysosomes involved in the degradation of proteins and has various functions in maintaining cell metabolism in normal conditions. The incidence of cathepsin B or its proteins and their high activity is described in a number of neoplastic diseases. About its importance for urological tumors has so far been only very few reports. Activity of the cathepsin B in patients with invasive bladder cancer were examined by determining its expression in tumor tissue and determining the concentrations of cathepsin B and pro-cathepsin B in their serum urine. Our results confirm that the diagnostic efficiency of the pro-cathepsin B urine urine is high. All so far published studies are focused on the investigation of biologically active proteins. Pro- cathepsin B is the biologically inactive...
Porphyrine conjugates for MRI
Dušek, Pavel ; Vojtíšek, Pavel (advisor) ; Kotková, Zuzana (referee)
The theoretical part of this bachelor's thesis focuses on clinical applications of MRI methods. It also presents a list of gadolinium-based contrast agents used in clini- cal practice. A new conjugate of 1,4,7,10-tetraazacyclododecane-4,7,10-triacetate-1- {methyl-[(4-aminophenyl)methyl]phosphinic} acid and 5,10,15,20-tetrakis(4-amino- phenyl)porphyrin potentially active as a contrast agent is designed on the basis of this review. Due to unique properties of porphyrin unit, this compound may act as a bimodal contrast agent or contrast agent for functional imaging. The target compound and its ytterbium-complex were synthesized and partially characterized.
