City within City
Menšíková, Kateřina ; Žalmanová, Petra (referee) ; Nový, Vítězslav (advisor)
The bachelor thesis deals with the design of a set of residential buildings based on the cohousing principle. The proposed buildings consist of three apartment buildings intended for community housing of three social groups - seniors, students and young families. The design works with varying degrees of privacy and is based on a classic urban block typical for the area. It leaves the public parter as the street character, creating a platform. On the higher floors above the platform, the area is open and allows the greenery to pervade from the opposite side to the courtyard. The opening of the area creates better conditions for housing and also a semi-private space at the level of common floors of community housing. This space is a continuation of a semi-public park, serving as a background for cohousing and kindergarten.
Reservoirs - Brno, Zluty kopec (Yellow hill)
Menšíková, Kateřina ; Horňasová, Tereza (referee) ; Nový, Vítězslav (advisor)
Master’s thesis focuses on area of historic water reservoirs on Yellow Hill, situated between streets of Tomešova and Tvrdého. They continued to supply Brno with non-potable water until 1997 and TIC is currently looking for a new way to make water reservoirs accesable and more interesting to the general public. Due to the existence of underground reservoirs the site has remained undeveloped and is covered with dense greenery. Thanks to that, the character of the place is in the context of the surrounding build-up area very unique. The proposal not only respects that, but seeks to support it. The whole area is undergoing through new landscape design and is completed by three single-storey pavilions, which with their various functions ensure the operation of the site. The historic buildings that were used to manage the underground reservoirs are renovated and integrated into the operation of the site. An important part of the design is also the solution of barrier-free access to reservoirs, their interconnection and installation of optical device - the Camery Obscury in the oldest of the reservoirs. The whole place becomes more accesible and attractive for visitors.
Functional and Pathophysiological-morphological Correlates of Neurodegenerative Diseases
Dušek, Pavel ; Roth, Jan (advisor) ; Baláž, Marek (referee) ; Menšíková, Kateřina (referee)
This doctoral thesis pictures neurodegenerative diseases as a multilevel process, describes various correlates on each pathophysiological level, and presents selected correlates in Huntington's disease and mitochondrial membrane protein-associated neurodegeneration (MPAN). It uses various methodological approaches such as basic laboratory research, clinical work, imaging, database formation, and database summary. Changes in the amount of respiratory chain complex I and respiratory chain complex IV in buccal ep- ithelial cells of Huntington's disease patients are described. The insufficient power of optical coherence tomography as a biomarker in Huntington's disease is demonstrated. Various phenotypes of MPAN are summarized, and an association between C19orf12 mutation and visual impairment is confirmed. A phenotype of a well-documented case of MPAN is presented. Keywords: C19orf12 mutation; color discrimination; contrast sensitivity; huntingtin; Huntington's disease; iron accumulation; mitochondrial membrane-protein associated neurodegeneration; multilevel process; neurodegeneration; optical coherence tomogra- phy; parkinsonism; respiratory chain complex; retinal nerve fiber layer thickness
Molecular genetic aspects of neurodegenerative diseases
Parobková, Eva ; Matěj, Radoslav (advisor) ; Kalinová, Markéta (referee) ; Menšíková, Kateřina (referee)
Genetics plays a crucial role in translational research, which ultimately aims to develop new therapies that modify neurodegenerative disorders. We anticipate that individual genetic profiling will become increasingly important in a clinical context with implications for patient care in line with the proposed ideal of personalized medicine. Although whole exome sequencing (WES) is now widely used in gene identification studies, there is no doubt that whole genome sequencing (WGS) will soon replace WES as the standard method for gene discovery. Hopefully, in the near future, comparable initiatives will be developed to generate global, fully genomic, publicly available data for various forms of neurodegenerative diseases such as FTD. In the first part of this work we focused on a group of patients diagnosed with "pure" sCJD and sCJD and PART or early phase AD and sCJD with more advanced AD. We compared and analyzed a new generation sequencing (NGS) method of 15 genes that may be involved in the development of the disease. We identified a p.E318G polymorphism in the PSEN1 gene, which was found in three patients (10.3%), one of whom had pure sCJD and two were sCJD + AD. No relevant variants were observed for the other AD APP and PSEN2 genes. In PRNP, insertion of p.P84_Q91Q was detected in one patient...

Relation of the Secondary Housing Users to Nature
Menšíková, Kateřina ; Bartoš, Michael (advisor) ; Kopp, Jan (referee)
This diploma thesis analyses second homes in a specific region of the Lužnice river middle course (the cadastral town of Soběslav and the cadastral parcels of the Roudná, Klenovice and Skalice villages). The applied research part is based on a theoretical background research that defines basic terms, outlines appropriate topics with a connection to second homes (rural tourism, history of second homes, naturbanisation) as well as describes the analysed region. The aim of this thesis is to discover what motivates second home dwellers to use these dwellings for individual recreational activities and to identify the current situation of second home dwelling in this specific region. Another goal I have set for this thesis is to analyse how satisfied are second home dwellers with public services and facilities and whether they are thinking about converting their second (holiday) homes into permanent homes. The methodology of this thesis is based on a questionnaire analysing the second home settlements in the region specified above. The data have been evaluated using Microsoft Excel a PSPP software. The evaluation of foals jumping abilities.
MENŠÍKOVÁ, Kateřina
The aim of this work was pass judgment the jumping abilities of foals measured at freestyle jumping during one's year. By the help of videorecording and subsequently computer analyses was assessed 427 jumps in free scope performed by 6 mares and 6 stallions in year-class 2002, 2003, 2004. Video records were made out by digital video kamera CANON type DM {--} XM 2 E. Processing of video records is done through the use of a programme for work with digital video records Pinnacle Studio 9. Movement mechanics analysis is practised in a programme Lucia 32 G Version 4.11. Following parameters were observed during a jump: jump bow characterized by 4 angles formed deciding physical parts, maximum flexion of the forelimbs {--} carpal joint and fore fetlock joint, rise of the forelimbs, flexion of the hindlimbs above a jump - knee-joint, hock joint, hind fetlock joint, location of the maximum carpal flexion. To objective assessment was made evaluation by statistical packet R version 2. 2.4.1 (2006) with usage procedures of Linear Models and there was verified demonstrativeness of differences among particular horses. It is evident from the findings, that jumping abilities of horses during the horse rearing are approximately same, but some of the qualities may change by training. . The objectively values found out during judging of a jump by way of video analysis make it possible to gain more accurate pieces of information about jump abilities of a horse and these pieces of information are usable in breeding of horses for a show-jumping.
