5 ABSTRACT Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Karolína Štěrbová Supervisor: prof. RNDr. Lenka Skálová Ph.D. Title of diploma thesis: Changes in expression of biotransformation enzymes in nematodes affected by albendazole Barber's pole worm (Haemonchus contortus) is highly pathogenic parasitic nematode, which assaults gastrointestinal system of small ruminants. Disease. Which caused is named by the agent haemonchoses. Long-term infection leads to develop of anaemia, lose weight of host and often even death. For treatment of this disease, synthetic antiparasitic drugs called anthelmintics are used. However, worldwide problem is growth of resistance against all used anthelmintics. Cause of this problem is probably common and unreasonable using of anthelmintics and the exposure of the parasites to sub-lethal doses. This thesis is focused on study of the mechanisms of resistance of barber's pole worm to albendazole, anthelmintic belonging to benzimidazoles group. The increased expression of biotransformation enzymes could contribute to resistance development. In our study, the influence of albendazole on expression of selected genes in adults of H. contortus from the ISE (Inbread Susceptible Edinburgh) strain was studied. Worms...
Induction and inhibition of acenaphthenol dehydrogenase

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Title, Name, Surname of candidate: Klára Štěrbová Title, Name, Surname of tutor: Doc. RNDr. Lenka Skálová, Ph.D. Title of a diploma work: Induction and inhibition of acenaphthenol dehydrogenase Breast cancer belongs to hormone-dependent tumours. Apart from lifetime exposure to estrogens, the volume of female active sex hormones in breast tissue also plays a big part in the development of this disease. One of mechanisms regulating concentration of estrogens in the target tissue at the pre-receptor level is their interconversion from inactive estron to its active form, 17β-estradiol enzymes from the aldo-ketoreduktas 1C (AKR1C) subfamily. On the basis of specific substrates, inductive and inhibitive studies, enzymes involved in the conversion of individual hormones can be characterized. Relatively specific substrate for AKR1C is acenaphthenol (AcNOH). However, due to the fact that in a multi-enzyme model system the involvement of other enzymes in the reaction cannot be excluded, the enzymes which take part in conversion of acenaphthenol labeled "acenaphtenol dehydrogenase" (AND). The Aim of my diploma paper is to spectrofluorimetrically determine the activity of AND in cytosole taken from breast...
The Czech project The Cover of the book: a description of the project and its perspectives
Štěrbová, Kateřina ; Římanová, Radka (advisor) ; Drobíková, Barbora (referee)
The bachelor's thesis Book covers: the project description and its perspectives deals with the description of the project from its beginning up to the present. We especially emphasize the Research Library of South Bohemia, which is together with the Moravian Library, the administrator of the whole project. The main goal of this thesis is to describe in detail the whole project Book covers and closely capture its development. The introductory theoretical part sums up the project history as well as the description of the integration of the Research Library of South Bohemia into the project. Other chapters deal with current development of Book covers and we briefly mention the very scanning and subsequent processing of covers, title pages and contents illustrated by the example of the Research Library of South Bohemia. The bachelor' s thesis also contains a practical part which introduces the methodology and setup of the research. The analysis was realized by the qualitative methodology, using semi structured dialogues. The summery contains compilation and evaluation of the gathered facts
