Detached house with a car repair shop

The bachelor thesis deals with design house with a car repair shop and elaboration of design documentation of the building. It is a slab-brick building, which is divided into a two- story living area and single storey part of the car repair shop. The building will be covered with flat and gablet roof. The main text contains the message type A, B, D. The content is guided by Annex 6 of Decree No. 62/2013. Attachments are inserted into the preparatory work, the drawings and calculations. Annex consists of 7 components.
Woman´s Clothing Factory

The women´s clothing factory is situated in the city part Brno-Židenice in the area of one so called „brownfields“. Currently there are rusty storages and old garages standing in the building ground. The site is bordered by several streets – it is Rokytova street, Komprdova street and Údolíček street – and the fourth side is closed by boundaries of a cemetery. The area can be entered from the north-west street Údolíček. According to the ground plan, its location is temporary, so we changed it a little to better match the needs of the company. The entrance for pedestrians is situated at the north-west too. And there are also parking places and other necessary manipulation space situated on the grounds. The clothing factory is aimed at production of Giorgio Armani´s women confection so the architectural solution is based on the basic philosophy of the company. Their models are famous for their simple design with a patterned fabric that make them special. So the basic mass of the object is a simple block, which is sensitively placed at the surrounding landscape and which is made special by the parts of ovals that are embedded or comming out of the basic mass. These parts of ovals create both – resting and functional spaces. The strong horizontal character is broken by a cylindrical body stucked into the basic shape that surmounts the block by one floor. A big part of the production hall is glassed-in so we can get enough light into the interier and the employees can be in touch with the surrounding. There is a facade net strunged in front of the glass facade. The net limits overheating and also plays the role of the unifying element. The building integrates a producion hall, storages, sanitary rooms, administrative space, canteen with affilated spaces and energy traffic necessary for running the factory. The wing cell is a steel skeleton combined with reinforced concrete circumferential wall and tie brick walls.
The Development and Contemporary Seniors Houses in the Czech Republic, Focus to South Bohemia Region

The thesis deals with the theory of social service types with regard to the situation in České Budějovice. It includes types of social service, the development and current situation in the Czech Republic. The situation abroad is discussed as well. The Act no.108/2006 Coll., on social service is described and the part dealing with social services related to senior citizens is highlighted. The research part is aimed at assessing the quality of social service by its users and social facilities workers. The research was made by questionnaires completed with clients of retirement homes (Máj, Hvízdal and Dobrá Voda) in České Budějovice. The research revealed that clients of social services are satisfied with the quality. Their relatives were satisfied to a great extent.
Using the Interactive Whiteboard in English Language Classes at Middle School Level

This diploma thesis is focused on using the interactive whiteboard in English language classes at middle school level. The theoretical part is focused on describing the interactive whiteboard, development and description of particular language skills (listening, speaking, reading, writing) and language subskills (grammar, pronunciation). The practical part deals with a preparation of some English classes. From observation of the classes, from the interviews with teachers and the questionnaire among pupils I will make a conclusion for training which language skills and subskills it is good to use the interactive whiteboard.
Quest for protein biomarkers of neural stem cells differentiation

Understanding neurogenesis and neural cell differentiation presents a unique challenge for treatment of nervous system disorders. To gain more insight about molecular mechanism of differentiation of neural cells, we applied different proteomic approaches using classical 2-DE followed by MS and antibody microarrays. Based on 2-DE, profile of constituent proteins of neural stem cells and their differentiated progenies was estabilished at first and then the protein species that are significantly up or down regulated during the differentiation were selected. Differentiation of neural cells was accompanied by changes in the expression of proteins involved in DNA and RNA binding, mRNA processing and transport, stress responses, iron storage and redox regulation. Immunoblot verified changes of hnRNP A1, hnRNP A2/B1, RafB, heme-oxygenasy 2, GRK2 proteins and alphaB-crystallin (S45), CDK1/2 (Y15), PKC mu (S738+S742), proline-rich Akt substrate (T246) phosphorylations during differentiation.
Protein fractionation and relative quantitation using PF 2D and ITRAQ for biomarker quest

ProteomeLab PF 2D System - Protein Fractionation in 2 Dimensions (Beckman Coulter, Fullerton, CA, USA) has been developed to fractionate complex protein mixtures by chromatofocusing in the first dimension followed by high-resolution non-porous silica reversed phase chromatography (RP LC) in the second dimension. Despite the high-resolution power of ProteomeLab PF 2D, UV-based quantitation could be compromised due to possible co-elution of several proteins into one fraction. Hence, we present an optimized protocol for application of isobaric tags for relative and absolute quantitation (iTRAQ) and MALDI-TOF/TOF mass spectrometry to obtain quantitative data from peptides derived by tryptic digestions of intact proteins fractionated by ProteomeLab PF 2D technique. To demonstrate the feasibility of such an approach, protein expression patterns obtained from the ProteomeLab PF 2D fractionation of human T-lymphoblastic leukemia CEM cell line were utilised.
Proteomika CDK inhibice v nádorových buňkách

In order to improve our understanding of the biochemical basis of the anti-cancer activity of olomoucine-derived synthetic cyclin-dependent kinase inhibitors (CDKIs) and to search for novel proteins associated with these biological effects we applied complex proteomic approaches. To analyse cellular responses to the CDKI we used two cancer models: the CEM T-lymphoblastic leukemia cell line representing hematological malignancy, and the A549 lung adenocarcinoma cell line as a solid tumor model. Cancer cells of these lines were cultured in both the presence and absence (controls) of the CDKI, bohemine (BOH). Cellular proteins of both of these lines were then extracted and fractionated using conventional two-dimensional gel electrophoresis (2-DE), and for the CEM T-lymphoblastic leukemia cell line we also used a 2-D liquid phase fractionation system ProteomeLab PF 2D ( Beckman Coulter). Computer-assisted data analysis of the resulting 2-D protein expression maps was applied to determine the similarity/dissimilarity of the maps and to select characteristic protein spots or bands based on the quantitative differences between BOH-treated and control cells. Many of these differentially expressed proteins were identified by mass spectrometry, since they represent candidate biomarkers of cancer cell responses to CDK inhibition and cellular pathways that are relevant to the anti-cancer activity of the CDKIs. Subsequently, we focused directly on these proteins in confirmatory studies using various techniques (including quantitative immunoblotting, immunocytochemistry and functional activity analyses) to demonstrate the validity of the proteomic results and extend our knowledge of the CDKI effects.

A comparison of Czech and Italian export policies

The opening chapter gives a general overwiev of Czech and Italian national economies' characteristics. The second chapter treats the system of national export assistance adopted in the Czech Republic. Consequentially, the third chapter is dedicated to the export policy of Italy. A comparison of Czech and Italian export policies from various points of view is put forth in the fourth chapter.
