Architectural and urban developement of Liberec 1352-2006
Zeman, Jaroslav ; Macek, Petr (advisor) ; Horyna, Martin (referee)
Liberec a relatively young city is compared to with the majority of urban settlements in our country but its development has been equally interesting and complicated. The onset of the industrial revolution meant a major breaking point in particular because the following gründer period has marked it most notably. The characteristic of the city is, beside a large urban fragmentation and complex disorder influenced by the local field configuration, especially the phenomenon of garden cities. Liberec with Prague, Usti nad Labem, Brno and Opava accounted for one of the centers of the second stream of culture, represented by German-speaking architects settled on our territory, which formed a natural counterpart to the architecture of the Czech majority.

Intra-articular Calcaneal Fractures - Osteosynthesis with Calcaneal Locking Compression Plate - Contribution to Fill Defect in Body of Calcaneus
Zeman, Jaroslav ; Koudela, Karel (advisor) ; Plánka, Ladislav (referee) ; Skála-Rosenbaum, Jiří (referee)
Calcaneal fractures are common injuries that prevent patients from getting on with their normal life for a long time. They are usually caused by axial forces leading to impaction of the talus into the calcaneus. Falls and jumps from heights are the most common causes. These fractures also occur bilaterally. We used open reduction and internal fixation with a calcaneal LCP for the treatment of 98 patients with 114 calcaneal fractures in our department from August 2005 till December 2011. Bilateral fractures of the calcaneus occurred in 16 patients - 2 women and 14 men. On the day of injury plain radiographs of the calcaneus in lateral and axial projection were taken. Computed tomography was performed in all operated patients in sagittal, transversal and coronal planes. On the basis of CT findings the fractures were classified as Sanders types I - IV. Patients with type II and III fractures were indicated for surgical ORIF treatment. The results were evaluated using the Rowe score. The aim of the experimental work was to assess the biomechanical strength of the calcaneus without fracture and to compare it with the strength of the calcaneus after the osteosynthesis with an angular stable plate alone or in combination with filling of the calcaneal defect with injectable self-hardening hydroxyapatite...
Creating the learning programme for facilitating the higher education school choice and verifying its benefits
Zeman, Jan ; Valenta, Josef (advisor) ; Valášková Vincejová, Eva (referee)
The thesis aims at improving the process of choosing a university by designing an experimental educational course composed of activities focused on self-knowledge and working with information. The theoretical part includes an overview of the existing research about higher education school choice. To create suitable activities, it was necessary to know the importance of choice factors, prospective students' needs and the facts about current sources of information. The thesis also presents the current state of career consulting and courses focused on a similar subject. The empirical part describes the content of the online educational workshop and its implementation including students' evaluation. The verification of the course's benefits was secured by interviews with the students and by evaluation of their learning outcomes from the activities. Furthermore, the thesis contains a description of the research limits caused by the Covid-19 pandemic and a recommendation for future improvements and methodical use. Key words: Higher education choice, career consulting, prospective student, decision-making, designing methodology

Title of the bachelor thesis: Use of regenerative procedures by students of UK FTVS Department: Department of Health Physical Education and Sport Medicine Supervisor: MUDr. Simona Majorová Goals: The main aim of the thesis was to find out which regeneration procedures UK FTVS students use the most, identify the main reason for use and the factors determining the selection of the regenerations procedure. The secondary objective was to summarize and characterize regeneration forms and resources on the basis of porfessional literature dealing with regeneration in sport. Than on the basis of the survey results with regards of professional literature make regeneration recommendation for students in practice. Metohods: The necessary informations was collected by using a non - invasive method of online survey. A total of 60 UK FTVS students from domain TVS participated in the research, across all grades regardless of gender. Results: On the basis of online survey was found that the most used thermal regeneration procedure among students is sauna, water procedure is swimming pool - swimming, the most used type of massage is sports massage and light procedures are not used by the vast majority of students. For electroprocedures, we chose only a questions about the experience with them, because of the need...
Biochemical and molecular studies of cytochrome c oxidase and ATP synthase deficiencies
Fornůsková, Daniela ; Zeman, Jiří (advisor) ; Hyánek, Josef (referee) ; Stiborová, Marie (referee)
Mgr. Daniela Fornuskova PhD thesis Biochemical and molecular studies of cytochrome c oxidase and ATP synthase deficiencies ABSTRACT The mammalian organism fully depends on the oxidative phosphorylation system (OXPHOS) as the major energy (ATP) producer of the cell. Disturbances of OXPHOS may be caused by mutations in either mitochondrial DNA (mtDNA) or nuclear DNA (nDNA). One part of the thesis is focused on the role of early and late assembled nuclear-encoded structural subunits of cytochrome c oxidase (CcO) as well as Oxa1l, the human homologue of the yeast mitochondrial Oxa1 translocase, in the biogenesis and function of the human CcO complex using stable RNA interference of COX4, COX5A, COX6A1 and OXA1L, as well as expression of epitope-tagged Cox6a, Cox7a and Cox7b, in HEK (human embryonic kidney)- 293 cells. Our results indicate that, whereas nuclear- encoded CcO subunits Cox4 and Cox5a are required for the assembly of the functional CcO complex, the Cox6a subunit is required for the overall stability of the holoenzyme. In OXA1L knockdown HEK-293 cells, intriguingly, CcO activity and holoenzyme content were unaffected, although the inactivation of OXA1 in yeast was shown to cause complete absence of CcO activity. In addition, we compared OXPHOS protein deficiency patterns in mitochondria from skeletal...
The impact of IGF1 and selected IGF1 gene polymorphisms on postnatal growth in children SGA/IUGR and extremely preterm newborns.
Kytnarová, Jitka ; Zeman, Jiří (advisor) ; Baxová, Alice (referee) ; Houšťková, Hana (referee)
Long-term outcome of extremely preterm neonates depends on many endogenous and exogenous factors. Long-term follow-up of extremely preterm neonates during childhood and analyses of IGF1 gene polymorphisms may help to better understand the problems connected with delayed postnatal growth and the progression of cardiovascular diseases and diabetes mellitus type 2 in adulthood. The aim was the long-term follow-up of anthropometric parameters in children born at 22−25th and 26−27th week of gestation and to study the association between postnatal growth of extremely preterm children, children small for gestational age (SGA) and children born at term with appropriate birth weight/length (AGA) and IGF1 gene polymorphisms: (CA)10-24 repetitive polymorphism in promoter, microsatellite marker D12S318 and 185 bp in 3'UTR, (CT)n polymorphism (CA)n polymorphism 216 bp in the intron 2. Methods. 242 infants born at 22-27+6 weeks were enrolled. Anthropometric parameters were measured at the ages of 2 and 5 years in 72 children born at 22-25+6 week (group I) and 85 children born at 26-27+6 week (group II). Polymorphisms of IGF1 were analysed in 51 extremely preterm, 208 AGA and 59 SGA children using fragment analyses. The data of postnatal growth data in AGA children were obtained at 18 months, in SGA and extremely...
Význam laktátu v diagnostice mitochondriálních onemocnění u dětí
Magner, Martin ; Zeman, Jiří (advisor) ; Baxová, Alice (referee) ; Procházková, Dagmar (referee)
The lactate level assesment in various body fluids plays an important role in the diagnostics of mitochondrial disorders in children. However, the interpretation of lactate level is often difficult due to its unspecificity and variability even in particular mitochondrial disorders. Three specific aims have been stated in this PhD Thesis: 1. To analyse the role of lactate examination in the differential diagnosis between children with mitochondrial disorders and children with other diseases. 2. To study the lactate level differences in various mitochondrial syndromes. 3. To characterise the clinical and laboratory data of neonates with mitochondrial disorders and to suggest new diagnostic algorhytms. Clinical and laboratory data from patients hospitalized in the Department of Pediatrics were collected. Laboratory methods were provided in the cooperation with the Mitochondrial laboratory of the Department of Pediatrics and Institute of Inherited Metabolic Disorders. The study with lactate levels in 107 patients documented that brief seizures lasting less than 2 minutes did not increase lactate concentration in the CSF. CSF-lactate was a relialable marker in differential diagnosis in the children with mitochondrial disorders against children with epilepsy. 2. The severity of particular phenotype is more...
Binding of eIF3 in complex with eIF5 and eIF1 to the 40S ribosomal subunit is accompanied by dramatic structural changes
Zeman, Jakub
In eukaryotic translation, eukaryotic initiation factors (eIFs) are at least as important as the ribosome itself. Some of these factors play different roles throughout the entire process to ensure proper assembly of the preinitiation complex on mRNA, accurate selection of the initiation codon, errorless production of the encoded polypeptide and its proper termination. Perhaps, the most important factor integrating signals from others and coordinating their functions on the ribosome is eIF3. In Saccharomyces cerevisiae, eIF3 is formed by five subunits. All these subunits contain structural motifs responsible for contact with ribosomal proteins and RNAs. In addition to these highly structured parts, the rest of eIF3 is unstructured and very flexible. Therefore, despite the recent progress thanks to the use of a cryo-electron microscopy, a precise structure and position of eIF3 on the 40S ribosomal subunit are still not known. Also, the presence of eIF3 on 80S during early elongation and its role in reinitiation and readthrough are not fully understood. In order to crack mysteries of yeast eIF3, we used x-ray crystallography, chemical cross- linking coupled to mass spectrometry, and various biochemical and genetic assays. We demonstrated that eIF3 is very compactly packed when free in solution. This...
