Role of the low-molecular metabolites in the development of yeast colonies

Previous research of colonies formed by yeast Saccharomyces cerevisiae growing on glycerol agar medium revealed two major cell types of U and L cells that are formed within these colonies. This colonial cell differentiation seem to be caused by communication among yeast cells as well as whole colonies and affected by changes in the environment (for example changes in nutrients). Studies of U and L cells showed that U cells are more resistant against biological, chemical and physical stresses than L cells. The aim of this thesis was to isolate U and L cell types and investigate their resistance against selected low molecular weight chemical substances produced in Ehrlich pathway. Ehrlich pathway was discovered in 1907 and was classified as amino acid catabolic pathway in yeast S. cerevisiae. The low molecular intermediates are formed in Ehrlich pathway which are called fusel (original name from German) alcohols and acids. These chemical substances are widely used in food industry and cosmetics especially because of their aroma. The effort to minimize complications in ovulation induction with gonadotropins

The effort to minimize risks of ovulation induction with gonadotropins The aim of this study was to evaluate three different starting doses of follitropin beta based on patients' response to clomiphene citrate treatment to assess the lowest effective dose for follicular development. In fact the determination of the appropriate gonadotropins' starting dose to achieve only mono-follicular development is the most important issue in ovulation induction. This is the main goal to minimize cycle's cancellation due to low or high ovarian response and to prevent multiple pregnancies or ovarian hyperstimulation syndrome. The objective of the trial was to determine the effect of different follitropin beta dose (50, 75 and 100 IU daily) on follicular development, thickness of the endometrium, total consumption of gonadotropins per cycle etc. We evaluated 90 cycles with ovarian stimulation for 90 patients with unexplained infertility, mild male factor or anovulatory disorder. We prospectively divided patients into 50, 75 and 100 IU groups based on patients' response to clomiphene citrate treatment. We performed 85 intrauterine inseminations. The basic principles and capabilities of development using Niagara Framework, on which Hawk is based, are explained. Lastly, the specific components and external database application is described.

Candida parapsilosis is an opportunistic fungal pathogen of humans causing a variety of infections. Immunocompromised individuals represent the most threatened group of patients. The increasing frequency of infections and occurrence of drug resistant strains are the main reasons for research focused on novel antimycotic compounds. Inhibition of secreted aspartic proteases (Sap) of pathogenic Candida spp. appears to be a potential target of therapeutic intervention. The genome of C. parapsilosis contains at least three genes coding for secreted aspartic proteases, denominated SAPP1-3. Protease Sapp1p has been well biochemically and structurally characterized, whereas Sapp2p and Sapp3p have been given less attention. The first part of the thesis is focused on structural analysis of Sapp1p complexes with selected peptidomimetic inhibitors binding to the active site of the enzyme. In addition, complex of the isoenzyme Sapp2p with the well-known secreted aspartate inhibitor Pepstatin A has been analyzed. The second part is related to the fact that C. parapsilosis belongs to the Candida spp. with the unique ability to translate standard leucine CUG codon mostly as serine. Differentiation within yeast populations of Saccharomyces cerevisiae: influence of cultivation conditions

Long-time research of chronologically aging yeast populations of Saccharomyces cerevisiae laboratory strains revealed that yeasts are able to differentiate into specialized cell types. Differentiation of liquid cultures growing in glucose rich medium and differentiation of colonies growing on solid glycerol medium has been previously studied. These populations create two fractions of cells with diverse morphology which adapt their metabolism and physiology to enable a long-term survival of the yeast population in environment with limited nutritional potential. In this study, yeast subpopulations isolated from colonies growing on solid glucose medium and liquid cultures cultivated in glycerol medium were characterized. Newly isolated cell types were compared with already known cell types isolated from colonies and liquid cultures. Selected metabolic processes and stress resistance were analysed in studied populations. Based on previous studies of yeast differentiation, a spectrum of GFP-labelled marker proteins was choosen and production and localization of these marker proteins was monitored within yeast populations. The topic of my diploma thesis is " Differences between the transfer of shares in a business corporation for consideration and the purchase the business establishment". The main objective of my thesis is to explain and compare two legal institutes that are frequently used in order to change financial or proprietary structure in a business corporation. In my thesis I analyse the most important aspects which have to be taken into consideration while making these transactions. These aspects are legal, accounting and tax viewpoints that are crucial for a responsible seller or purchaser to consider according to the aim he wants to achieve by this transaction before the disposition commencement with one of these institutes. In practical part of my thesis I present some examples and their references following particularly from judicature of superior law courts (legal source since 1 January 2014 ) which show consequences and their view of aspects of individual steps concerned with share or company conversion. Further the practical part contains specific accounting methods and taxes related to these transactions.

This thesis deals with searches within large-scale text data, which are enriched by semantic information, especially the highlighting of named entities. The thesis begins by analysing methods used for the indexing of partly structured text data and the methods used for searching. The main body of the thesis consists of the description and implementation of a query engine, which analyses queries and requirements on range and structure. This query is later converted into an MG4J search engine format and applies restrictions on the results. These results have been tested on time response, as well as on a number of results, which are then compared to older results obtained from English Wikipedia.

This bachelor thesis deals with the issue of metal cutting and automation of manu-facturing processes. The first part is a presentation of some of the theories and methods of cutting and non-cutting operations. The basis and principles are described. Following part focuses on cutting materials, its basic division and expected progress. Next part describes distribution of machining centres and the market presence. The attention is also focused on robotization and its use in engineering industry. The most perspective and modern technology appears to be the non-cutting operation, which is described in this thesis.

Flow Measurement of Hot Dusty Gas

The bachelor thesis deals with the measurement of hot dusty gas. The introduction outlines the problems of measurement. Another part of the work contains a theoretical introduction to the measurement of gas flow. In successive chapters the basic principles and characteristics of suitable methods for measuring the hot dusty gases are described. Finally, the function blocks are implemented in the Simatic STEP7 for calculation of volume flow rate and blocks are tested on PLC.
