Occurrence of β-rutinosidase in eukaryotic microorganisms
Adamcová, Kateřina ; Weignerová, Lenka (advisor) ; Králová, Blanka (referee)
Rutinosides are very common glycosidic aroma precursors. The glycosidic moiety influences wine aroma, flavour and taste of juices, so its cleavage has many consequences. These interesting insights led us to a diglycosidase - the extracellular β-rutinosidase from Aspergillus niger. The purified β- rutinosidase was partly analyzed by MALDI-TOF/TOF. The insert encoding for β-rutinosidase was ligated into the expression vector pPICZα A. Pichia pastoris KM71H was used as an expression system. It was find out, that β rutinosidase gene consists of a 1137 bp, encoding protein with 379 amino acids. The enzyme was determined to have relative molecule mass 60 kDa by sodium dodecylsulfate polyacrylamide gel electrophoresis. The pH and temperature optima of the enzyme were found to be 3,0 and 50 řC, respectively. p-Nitrophenyl-β-rutinoside was used as a substrate Powered by TCPDF (www.tcpdf.org)
Johann Georg de Hamilton. Life and Work.
Ourodová, Ludmila ; Prahl, Roman (advisor) ; Adamcová, Kateřina (referee) ; Jandlová Sošková, Martina (referee)
The content of this dissertation is the life and œuvre of Johann Georg de Hamilton, a relatively obscure painter of hunts, portraits of horses, hunting still-lifes and hunting scenes. Johann Georg de Hamilton (1672-1737), a painter belonging to a famous Scottish family, was influenced in his creative work considerably by the 17th -century Flemish painters of still-lifes and hunting scenes. He was active predominantly in Vienna and in South Bohemia, in service of Adam František, Prince of Schwarzenberg, as well as Karl VI of House Habsburg. He created hunting-themed paintings and portraits of horses to members of both the secular and the ecclesiastic aristocracy of the lands of the Austrian Empire, such as the Houses of Liechtenstein, Serényi, Althan and others. This dissertation is the very first attempt at a monographic analysis of the life and œuvre of this painter. In addition to new bibliographic data, it offers an in-depth insight into the relationship between the person who commissioned his work, Adam František, Prince of Schwarzenberg, and the painter Johann Georg de Hamilton on the basis of extant correspondence, and also attempts to present the painter's œuvre in a cultural-historical and artistic context. The dissertation mentions the first exhibition of a collection of Hamilton's work,...
The communication strategy of the Knižní klub company
Adamcová, Kateřina ; Halada, Jan (advisor) ; Čeňková, Jana (referee)
The diploma thesis deals with the communication strategy of Knižní klub publisher since its establishment till the end of the year 2013. Knižní klub belongs to publishers that mostly in 90s' formed Czech literary culture by its business model. Until recently it was a part of the world media group Bertelsmann, which has operated literary clubs all over the world since 50s' of the 20th century. It entered the Czech Republic shortly after the revolution and brought many forbidden titles. The policy of edition represented the mix of the quality world literature and also the whole range of saleable books which copied actual readers' trends. On this basis, I focus on historical background and its influence of the policy of edition. Then I deal with the development of the literary club after the year 1989. With The Knižní klub publisher, which has gone through several changes from distribution company to appropriate publisher, I try to target the communicational activities in socio-cultural context. Communication strategy was observed through club catalogues and media. Media image in social-cultural supplements Víkend Mladé fronty Dnes, Salon Práva and critical monthly journal Literární noviny is also part of the text and helps to understand the strategy of the publisher.

Dörflová, Jana ; Macek, Petr (advisor) ; Adamcová, Kateřina (referee)
The subject of interest of this work is the collection of statues ordered by Count František Antonín Špork, placed in 1735 in the castle garden at Lysá nad Labem. This collection is made up of 32 statues and is made up of primarily allegorical subjects (allegories of the Seasons, the Continents, Day and Night, the Elements, and the Twelve months). It is supplemented by Pairs of lionesses, lions, and sphinxes. Originally the collection was attributed to the workshop of M. B. Braun, more recently it has been credited to "Sculptors from Benátky", which is most commonly identified in literature with František Adámek from Benátky nad Jizerou. In the first part of this written work, the person of F. A. Špork will be briefly introduced as the one who commissioned the works of art. Another part of the work deals with the history and the sculptural decoration of the castle gardens. The work is focused on the formal analysis of the sculptures of the collection, to identify their characteristics and comparison with the works of M. B. Braun. The final part of the work deals with authorship of the collection. Existing knowledge from various sources...
The history of Kruhˇs publisher and policy (critique)
Adamcová, Kateřina ; Halada, Jan (advisor) ; Čeňková, Jana (referee)
The bachelor thesis History of Kruh's publisher and edition policy (critique) is a description of Kruh publishing house and it also critically analyzes editorial policies. The thesis includes Kruh's predecessors Východočeské nakladatelství and Jiří Chvojka's publishing house in Havlíčkův Brod. Kruh is associated with Jiří Chvojka's publishing house only remotely, but it is an interesting object for comparison of regional publishing houses. There had existed two publishing houses in the east of the Czech Republic, which published the regional literature. Their production extended the complexity of the national literature. This thesis outlines historical figure of Jiří Chvojka and edition policy of his publishing house. Furthermore, I also wrote about Krajské nakladatelství in Hradec Králové, which had been operated from 1950 till 1951. Even at that time it was possible to find an attempt to publishing regional literature. For each publishing house is outlined their origin, historical context and part of edition policy. Attention is focused on regionalism and its application to the literature of the Eastern Bohemia. I analyzed the publishing house Kruh in relation with former circumstances. I used empirical-analytic approach in this thesis, which creates a critical study of this publishing house. Keywords...
