Managerial Decision-Making on Sustainable Transport Development in a chosen Region

The thesis is focused on sustainable transport development of the Třeboň Region (Třeboňsko). This region is specific because of its low and decreasing population density, significant seasonal tourism and highly protected nature and landscape. Several different methods methodical approaches are used in this thesis for decision-making and designing/proposing provisions which together create the whole concept of sustainable transport development in the examined area. This study has proposed several important particular provisions in the infrastructure (expanding of infrastructure for non-motorized transport, increasing road safety and throughput and others) as well as in the public transport (increase of economic efficiency, improvement of transport services). Generally conceived provisions may be implemented in territories with similar characteristics.
Management kvality ve vysokoškolském vzdělávání

The thesis deals with quality management theory as an important part of management science. The primary objective of this work is an identification, formulation and analysis of such managerial issues in quality of higher education, which either are not known, or whose resolution is not considered fully sufficient. The thesis contains a bibliography of more than 200 related scientific works and presents selected issues of quality management in higher education, such as quality perception or its sustainability. Furthermore, the work suggests new methodologies for quality evaluation and demonstrates the use of computer simulations and modelling as a tool for quality management in higher education.
The Analysis of Passengers‘ Preferences on the route Zliv-Ceske Budejovice

The aim of this research study is to analyse the demand for public transport on the route Zliv-Ceske Budejovice. Its main aim is to figure out whether comuters prefer public transport to personal transport and what effect does that make on the environment. The analysis was based on findings both prior and posterior demand. The basis for the processing methodology and study of scientific literature is held with a focus on transport and related topics. An integral part of the methodology process is quality data obtained from residents and those moving to session Zliv-Ceske Budejovice. The method used, allows us to analyse the differences between real and desired transport demands. The results of this work will help us manage public transport efficiently, in order to increase the attractiveness of public transport in the session and as a consequence, the negative environmental impacts of other modes of transport.
Managerial decision making about external relationships of high school

Diploma thesis acquants reader about decision problems at Střední odborné škola informatika a spojů a Střední odborné učiliště Kolín. Chosen problem are desribed and analysed, but also variants of solution are created, so tehy can be used as an support to decision making for school manager. These problems are about getting new students, usage of unused rooms of dorm and school cafeteria. For creating variant of solutions Cobb-Douglas utility function is used, moreover the thesis teaches reader how to work with it, so a manager of school can use it for his own decision making proces. Diploma thesis was created by request of a principal Ing. Miloš Hölzl as he is awaiting the results.
Applying methods of Competitive Inteligencie for educational companies

This thesis deals with the design of Competitive Intelligence for specific training company. The main objective of this work is to characterize and help choosing the person responsible for carrying out activities related to the CI in the company, to identify the most appropriate information sources to obtain information about the company and software tools for their monitoring. The theoretical part is drawn on the literature and is dedicated to the description of competitive intelligence and characteristics of different phases of the CI cycle. Other sections are devoted to typologies of information resources and software tools for their monitoring. Following parts are dealing with the characteristics of the category of small and medium-sized enterprises; the investigated company and education organizations fit in this category. The practical part of this thesis deals with the possible proposal of Competitive Intelligence for specific training company. As the first step we realized the analysis of the business environment and its major limitations. In the following part we deal with the CI system design, consisting of a selection of variant realization, along with the characteristics of the responsible person, the definition of information resources and recommending software tools for regular monitoring. The practical benefit of this work is to introduce CI system design, based on monitoring of information sources, obtaining information and their subsequent evaluation in relation to the competitive environment of the company. Another important asset of the work is a sample of implementation of such a system in the company of categories of small and medium-sized enterprises, which respects their limitations.
Using web pages audit for Competitive Intelligence

This thesis is focused on thorough description of Competitive Intelligence term and its fol-lows usage for webpages audit. The practical part of this thesis is primarily created for the Bisnode company. However, it can be used everyone who wants to perform on-line webpages analysis of their competitors. In thesis are also used professional tools of Majestic company and SEOPowerSuite company. I got these for limited time after my e-mail corre-spondence with company´s support team. At the beginning of practical section the Bisnode company is introduced to readers, so that they might become familiar with it. After that, the analysis was performed and divided into 4 thematic areas based on Bisnode´s wish. I ana-lyzed strengths and weaknesses of monitored sites in each of this section so that the company can learn not only from their mistakes but also from mistakes of its rivals. I tried to analyze Competitive Intelligence term in detail in the theoretical part of thesis. In addition, this term is viewed from multiple angles (its history, reflection of its Czech trans-lation, ethic associated with the CI term or description CI as process). So, this thesis can be also a beneficial for readers that know this term but want get deeper knowledges.

Ph.D. Thesis: Spatiotemporal regulation of Lck aktivity in the TCR signalling Ondřej Ballek Abstract Arguably, the most studied cell types of immune system are T-cells. They are key players of adaptive immunity responsible for targeted action against pathogens or other danger signals. Due to their central importance, any alteration in the regulation of their activity leads often to immunopathology. Thus, the knowledge how to harness their bio-destructive effector functions is of critical importance. Up today, there is only limited consensus on the nature of molecular mechanisms controlling the initiation of T-cell activation. When T-cell receptor (TCR) recognizes its cognate antigen presented on antigen presenting cell (APC), the activation signal is transmitted through the plasma membrane and subsequent phosphorylation of cytoplasmic chains of TCR complex ensues. This is commonly considered as the first biochemical sign of T-cell activation, the process called TCR triggering. How the activation signal gets into the cell and which molecular mechanisms control TCR triggering are two fundamental, yet still unanswered questions. Mechanism of action of adenylate cyclase toxin on immune function of dendritic cells

The adenylate cyclase toxin (CyaA) is a key virulence factor of the whooping cough agent Bordetella pertussis. CyaA primarily penetrates CR3-expressing myeloid phagocytes and subverts cellular signaling by a rapid conversion of ATP to cAMP. In parallel, CyaA can form cation-selective pores within cellular membrane, provoking massive potassium efflux from cell cytosol. An enzymatically inactive adenylate cyclase toxoid (CyaA-AC- ) has then been abundantly used as an efficient antigen delivery tool over the past 20 years. This work focused mainly on the mechanism of action of CyaA toxin and of its toxoid on dendritic cells. We studied the potency of the CyaA toxoid to act as adjuvant, its penetration capacity and its potential use in delivery of influenza epitopes. We show that the pore-forming activity and the activation of MAP kinases JNK and p38 were crucial for the adjuvant effects of the CyaA-AC- , which provokes maturation of dendritic cells (DC) independently of Toll-like receptor (TLR) or inflammasome signaling. Furthermore, such CyaA-AC- -stimulated DC acquired the ability to induce CD8+ and CD4+ T cells responses, as was determined both in vitro and in vivo. Maternal-fetal cellular trafficking: clinical implications and consequences

Microchimerism is the presence of small population of cells with a different genetic information within the organism, which can result from bidirectional transfer of the cells between the mother and fetus during pregnancy. It is very studied phenomenon whose biological role is not clear yet. The presence of fetal cells in mother's body is associated with both positive and negative effects on maternal health. Microchimerism plays a role in cancer or autoimmune disease and it is implicated in development of tolerance mechanisms during pregnancy. Microchimerism could be used in prenatal diagnostics for aneuploidies or in prediction of complications during pregnancy.
Molecular mechanisms of regulation of FcɛRI signaling in mast cells

Mast cells are critical component of the immune system. In pathological situations, they are activated and are responsible for allergic reaction. Therefore, detail understanding of mast cell activation at molecular level is important for design of new therapies of allergic diseases. Principal transmembrane receptor of mast cells is the high-affinity Fc receptor for IgE (FcεRI). FcεRI anchors IgE on mast cell surface and upon cross-linking with multivalent antigen it becomes phosphorylated at its intracellular immunoreceptor tyrosine-based activation motifs (ITAMs). This triggers signaling cascade leading to cell degranulation and cytokine production. The antigen- mediated signaling through the FcεRI is critically dependent on interplay with intracellular protein- tyrosine kinases that phosphorylate the ITAM motifs and many other components of the signaling pathway. This study was focused on better understanding of signaling events leading to mast cell activation; emphasis was put on early activation events. First, we examined the role of protein- tyrosine phosphatases (PTP) in FcεRI phosphorylation.
