Effect of equilibration and freezing parameters on bull sperm motility after thawing
Beránková, Monika ; Stádník, Luděk (advisor) ; Zuzana, Zuzana (referee)
The hypothesis assume the existence differences in bulls spermatozoa viability after thawing, characterized by higher values of motility and higher proportion of live spermatozoa, in case of prolonged equilibration and lower temperature freezing gradient. The aim of this thesis was to find the influence of different equilibration and freezing curve conditions to on spermatozoa viability after thawing. The theoretical part of the thesis summarizes available information about the bulls semen parameters, methods for its evaluation and especially the processing and long-term preservation. Within the case study part of the thesis, during the years 2013-2015 the semen of the pre-selected group of 5 breeding bulls was being collected at the Sires insemination center. The group was uniform as to age, breed and housing and treatment conditions. After initial assessment the collected semen was divided into parts, which were then processed in different wals. A part of straws was processed by standard protocol, the other part was processed using different length of equilibration and freezing curve. First part of the straws was equilibrated in a standard protocol for 120 minutes; subsequently half of the equilibrated straws was standardly frozen by the Direct Freezing method based on 3-phase freezing curve while the other half was frozen using the different 2-phase freezing curve. Second part of the straws was initially equilibrated for 240 minutes with the subsequent freezing, similarly as in the previous case, used 3-phase and 2-phase freezing curve. Frozen straws were then stored in liquid nitrogen container at -196 °C. Spermatozoa motility was evaluated using CASA. The semen was further evaluated on the proportion of live and dead spermatozoa, using coloration of Eosin and Nigrosine, and on the sensitivity of the spermatozoa membranes, using HOS test, which detects the reaction of spermatozoa tail membrane to hypoosmotic conditions. The obtained results indicated higher progressive spermatozoa motility (+5.57 %) and higher percentage of live spermatozoa (+4.47 %) following the application of the prolonged equilibration (240 min). When applying the length of equilibration 240 minutes, the values of total spermatozoa motility and HOS test were higher as well, though the differences were not statistically significant. When evaluate the effect of using different freezing curves on the parameters of the spermatozoa after thawing, higher values (average + 1.6 %) at almost all of these parameters using 3-phase freezing curve were found, the diferrences were not statistically significant. Statistically significant differences among the bulls (P < 0.01) were found in total as well as progressive spermatozoa motility, in the proportion of living spermatozoa and in value of the HOS test - the individuality among the bulls is therefore evident. While evaluating the characteristics of spermatozoa movement by CASA, statistically significant differences among the bulls were confirmed at VCL, VSL, VAP, LIN, STR, WOB (P < 0.01), also between the diferrent length of equilibration (P < 0.01), but were not statistically significant when using diferrent freezing curves.
Relations between production capabilities and economy of Charolais cattle breeding on selected farm
Rozsévač, Ondřej ; Stádník, Luděk (advisor) ; Zuzana, Zuzana (referee)
The aim of the dissertation is evaluation of relation between utility properties and economics of breeding cattle - breed Charolais at the chosen farm. Especially breeding conditions and zootechnical measures with their effects on the economic results in years 2009 - 2014. There is in the literatury searches described the principle of breeding cattle and its economic conditions in the Czech Republic. Further history, characteristic of the breed and its breeding standard. There is chracterized the current state and expansion of the breed in the Czech Republic and France, the country of origin. The way of the checking the utility and the monitoring of decisive utility properties is written here. The work was done in the farm Komensko. The farm is focused on crop production with 696ha and breeding cows without a market production of milk with 102ha meadows and pastures. There is currently bred 76 pieces in the basic herd. The herd is before completion of the transmission crossbreeding owing to repeatedly mating of bulls Charolais. The achieved results of the reproduction are good thanks to replacing insemination natural breeding. Reserves were found in growth capabilities of calves, when monitoring weights on the 120th, 210th and 365th day were lagging behind the average in the Czech Republic and France. The achieved sales display an increasing trend, but are constrained by the expansion of basic herd and accelerated eliminate of cows with participation milk breed. The farm shows a profit during the monitoring period. The farm had the highest profit in 2014 and it was 431 823.- CZK. By contrast the farm had the lowest profit in 2010 with the result 18 035.- CZK. Generally we can say the farm had the highest profits in the last free years. That happened because of the increasing revenues from the sold animals and higher income from the subsidies. Promise for the future is to finish the transitional process and to allow selling the breeding material instead of the cattle for a much better price.

Stejskal, Martin ; Zímová, Kateřina (advisor) ; Zuzana, Zuzana (referee)
Institute of landscape character was anchored in Czech legislation in 1992 as part of Act No. 114/1992 Coll., On nature and landscape protection. The concept of landscape character under that law includes not only natural, cultural and historical values that create a unique landscape character, but and visual aspects of landscape scenes that show the visual attractiveness. Thus it becomes a complex and unique tool for conservation landscape. At present, while is protecting the landscape often discussed topic, as a result of increasing building pressures on Czech countryside. This work deals with the case (current) assessment landscape. On the basis of the methodical process of assessment of proposed construction, activity or land use changes on the landscape character (Vorel et al. 2004) within the meaning of the Act no. 114/1992 Coll., On nature and landscape protection, I tried to expert approach determine the impact of potential construction lookout tower on the hill of Oškobrh on the landscape character.

Factors Affecting the Distribution of Nitrification's Final Products during Biological Pre-Treatment of Fugate
Kyselová, Aneta ; Švehla, Pavel (advisor) ; Zuzana, Zuzana (referee)
In this diploma thesis, the influence of chemical composition of fugate, pH value, temperature and other factors on representation of final products of nitrification during biological pretreatment of fugate produced in agricultural biogas stations was evaluated. During the experiment, fugate from biogas station Nový Dvůr with N-amon concentration in the range of 1300 - 3200 mg/l at input was processed. Results from the time period beginning on 21. September 2015 and ending on 22. December 2015 (day 0 - 93) were assessed. Nitrification reactor of five liter operational volume was placed in a laboratory at temperatures ranging from 20.4 to 25.2 degrees Celsius. During the experiment, volumetric loading of N-amon reactor ranged from 0,18 to 0,44 kg/(m3.d) and efficiency of N-amon removal from 68,6 to 96,9 percent. Hydraulic period of water delay was being decreased from 12,3 days to approximately 4,2 days during the experiment. pH value was regulated by NaOH to be approximately 7. The experiment proved that it was possible to attain the process of nitrification in the aggressive medium of fugate. It was ascertained that pH value, which determined current concentration of Free Nitrous Acid (FNA), had significant influence on representation of final products of nitrification. Concentrations of N-NO2 in the reflux from the reactor were being increased from 130 up to 1840 mg/l during the process on the grounds of pH value fluctuations and increasing FNA concentration which inhibited activity of nitrite oxidizing bacteria (NOB). The representation of N-NO2 from day 0 to day 93 of the experiment gradually increased from 19 percent up to 95 percent. Concentrations of N-NO3 from day 0 to day 3 increased from 570 to 1056 mg/l and conversely from day 3 to day 93 they gradually decreased from 570 mg/l to approximately 100 mg/l. Representation of N-NO3 decreased from 80 percent to 5 percent from day 0 to day 93 and it was not possible to retain them as dominant products of nitrification on the ground of sensitivity of NOB.

Quality of turf on golf field Mystěves and recommendation of management
Nová, Michaela ; Svobodová, Miluše (advisor) ; Zuzana, Zuzana (referee)
For this bachelor thesis was selected eighteen hole golf course - golf complex Myštěves. Golf Course was founded in 2001 commissioning znormovaného public course. In 2003 opened the first nine holes Club Course and in 2010 opened the second club nine, so that efforts to complete a good eighteen-hole course. Visitor statistics were monitored throughout the season 2012. This monitoring was carried out using an Internet portal TEE-TIME. This observation was statistically evaluated. During the 2012 season was visited golf complex Myštěves 6054 players. Most players have been recorded in the month of August and that 1375 players. In contrast, the least-visited month was November 2012, when the total number of players was only 156th. Speed measurements using two different greens "Stimpmetru" showed that in most measurements, the moving speed of greens at optimal levels, which is very favorable. Soil reaction on the greens, the whole course is at optimal levels, thus not need to have to modify it. This is due to soil conditions and fertilizers used. For monitoring pest was found to occur in a given location in a small extent. In contrast, algae, magical circularity grass, dry grass and small ohniskovitosti plísňovitosti lawn was highest in the summer months when this increase was supported by changeable weather with alternating rain and hot days. The highest incidence was on the greens, where arrangements were made for their removal. As proposals for action was carried out in-depth aeration of greens with applications topdresingu. This operation should improve intake of water, fertilizers and support microbial life in the upper layer of the sward. After topdresingu hearty salad dressing was performed and after 14 days on the greens combined nitrogen fertilizer applied the late summer and autumn managed to eliminate the incidence of diseases. The biggest problem of this golf course is outdated equipment maintenance lawns. Specifically mowers Ransomes (produced 1985-1997), the mower Jacobsen (the year 1994), tractor Zetor 6711 (the year 1978), bunkrovačka Toro (1997) and others. The problem with the club trying to solve, but the main obstacle is the lack of funds for the purchase of a new fleet. There are efforts to modernize, but in a very limited scale.
