The diploma thesis deals with the motivation and barriers in education of adult students at a college. The issue of motivation and barriers of selected economically active students in higher education is discussed. The thesis is divided into a theoretical and an empirical part. The theoretical part describes the basic concepts related to the topic of the diploma thesis - motivation, motivation to learn, lifelong learning, adult education, higher education, barriers in education and more. The aim of the thesis is to find out and describe what motivated and continues to motivate selected economically active adults to study at a college, to identify the barriers they feel in this education and to find out how they deal with these barriers. The thesis identifies, analyzes and describes the motivation and barriers of selected adult students in education at a selected college. As part of the empirical part of the thesis, based on its aim, research questions are set, to which answers are subsequently sought through investigation. Motivation of teaching staff from a selected kindergarten for further education and professional development

The thesis focuses on the motivation of teaching staff for further education, the use of their potential and the professional development of individuals. The thesis is divided into theoretical and empirical parts. The theoretical part is mainly focused on motivation, motivational program and the teaching staff itself. Attention is also paid to further education, lifelong learning and curriculum and strategy documents. Within the research part, four research questions have been set based on the aim of the thesis. The empirical part is based on the research design of the case study. Within the case study, both qualitative and quantitative data collection methods are used to obtain data. For the teaching staff, a questionnaire survey method is used, while data is collected from the school management through a semi-structured interview. From the data collected, certain barriers are identified that make it difficult for the teaching staff to further their education and development. Each research question contains graphs that were obtained through the questionnaire survey, which are then described in detail. Comparison of selected educational platforms

The rigorous thesis is an extension of the diploma thesis of the same name and deals with the comparison of selected educational platforms. Its aim is to analyze the selected online platforms and their functions with regard to the possibility of incorporating modern teaching methods in adult education. The theoretical part is based on the literature search and other available sources and defines the concepts related to the issue, which are andragogy, adult education methods and online education. It also presents current trends and goals of education in the Czech Republic and the European Union. The theoretical part concludes with a summary of the important theoretical background from which the research part of the thesis draws. In view of the need for a detailed examination of the selected platforms, a qualitative research strategy and a content analysis method were chosen. An interview method was additionally chosen. The online learning platforms explored were Knowspread, Thinkific, Miro and BigBlueButton. For the purpose of the rigorous analysis, their course design features were characterized and presented. The bachelor thesis "Communication and its influence on the resolution of the conflict between teachers and parents" deals with the position of parents in the educational process and the importance of proper and quality communication between parents and school. The thesis focuses on the description of conflict situations between the school, its management and teachers, and parents. The theoretical part describes and defines the basic knowledgein the field of conflict and communication. There are presented types of conflicts, main stages of conflicts, their causes and potential triggers, possible solutions and obstacles that may occur in these solutions. The work also deals with prevention and the principles of good communication, and examples of good practice are presented as well. The practical part focuses on the description and analysis of specific conflict situations in the selected private primary school. The chosen method of case study deals with the examination of conflicts that first-grade teachers in the institution deal with. Charles University, Faculty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Title of Diploma Thesis: Effect of endoglin blockage on inflammation-induced endothelial dysfunction in human aortic endothelial cells Author: Andrea Škubová Supervisor of Diploma Thesis: PharmDr. Katarína Tripská, Ph.D. Aim: The aim of this thesis was to describe the effect of TNF-α and LPS-induced inflammation on the expression of endoglin and its transcription factors, the expression of biomarkers of endothelial dysfunction, the levels of soluble endoglin, and the adhesion and transmigration of monocytes through human aortic endothelial cells. We further investigated the effect of endoglin blockage by the monoclonal antibody TRC105 (carotuximab) on these processes. Methods: In this thesis, human aortic endothelial cells (HAEC) were used, and they were exposed for 16 hours to TNF-α (10 ng/ml) and LPS (100 ng/ml) and for 12 hours to TRC105 (300 µg/ml). The mRNA expression of endoglin and its transcription factors (KLF6, RelA, NR1H3), mRNA expression of inflammatory adhesion molecules (ICAM-1, VCAM-1, SELE) and mRNA expression of MMP-14 enzyme were measured by real time qRT-PCR. Aptamers application in research and diagnostics

The bachelor thesis deals with affinity molecules that are able to specifically bind to the target molecule. The first chapter is devoted to antibodies, i.e., affinity molecules of protein nature. It discusses their structure, development, production, and use. The main focus of the thesis is on aptamers, or short stretches of single-stranded DNA or RNA, which are often referred to as synthetic alternatives to antibodies. They have similar functions to antibodies but differ in structure, development, and production. Because of their similarity to antibodies, aptamers are used in various detection technologies to diagnose diseases. Aptamers are still in the research phase, but the first commercially available aptamer diagnostic kits are slowly starting to appear on the market. The final chapter compares the pros and cons of the two biggest rivals in the field of affinity molecules - antibodies and aptamers. Motivation of non-teaching staff for education

The thesis deals with the issue of further education of non-teaching staff which falls within the field of employee care. Today's society places ever higher demands on workers and organizations in the field of education, so it is necessary for schools to focus on the continuing education not only for teaching staff but also for other workers, so-called non-teaching staff. These workers also need to maintain a certain status in society which they will only be able to do if they have the opportunity to develop and further their education. The theoretical part is divided into three parts. The first chapter introduces the field of lifelong learning, formal, non- formal, informal education and andragogy. The next chapter focuses on the specificities of adult education. It also describes the needs of the motivation of adult individuals to learn. The last section then introduces the barriers that greatly affect adult education. The last chapter defines the concepts of education, development and learning. Well-being of secondary school teachers in Prague

This thesis deals with the subjective perception of well-being among secondary school teachers in Prague. The theoretical part of the thesis first defines the basic concepts related to the topic, namely the definition of the concept of well-being itself, which is viewed from many sources, as it is a very broad term. At the same time, the history of the term, its inclusion in andragogical reality and the topicality of the topic are described. Subsequently, all the areas that fall under this term are described, the ways in which well-being can be measured, but also possible obstacles to well-being are described. Then the theoretical part of the thesis is devoted to the teaching profession and the problematic areas that fall within this profession - demandingness, salary, prestige and others. The practical part focuses on qualitative research, in which data and opinions of individual teachers on their subjective perception of well-being, their current state, possible obstacles they perceive and what they perceive as key to their own well-being are collected through semi-structured interviews; at the same time, some additional questions are answered to reveal the relationships between different areas and well-being itself.
