The role of TGFß and study of prognostic factors of patients with MDS and AML

We did not find mutation in coding areas of genes for components of TGFbeta1 signaling pathway but we detected decreased or undetectable expression of these analysed genes.The decreased expression is probably caused by epigenetic changes, so by hypermethylation and deacetylation of promoter regionsof these genes.Antiproliferative and apoptotic effect of TGF1 was analysed in AML cell lines (ML1, ML2, CTV1 and Kasumi1). ML2 cells rezistence to inhibition of DNA synthesis by TGFβ1 is not caused by mutations of genes for components of TGFβ1 signaling pathway. We found that increased SnoN (Ski-like novel gene) expression on the level of coresponding mRNA and protein is probably accountable for this rezistence. Kasumi1 and M2 cells were sensitive to induction of apoptózis caused by TGFβ1 treatment but in less extent than by proteazome inhibitor bortezomib. The difference of AML cells of different lines answers shows a great heterogeneity AML in AML patients. Prognostic factors analysis in AML with normal karyotype confirmed that CEBPA (CCAAT/enhancer binding protein alpha) mutations predict favourable prognosis but the elevated EVI1 ("Ecotropic Virus Integration Site 1") and ERG ("ETS-related gene") expression are connected with unfavourable prognosis. Functional safety of pressure transmitters of BD SENSORS company

This master thesis examines the functional safety of the pressure sensor XMP i which is produced by BD SENSORS Company. The aim of this thesis is the demonstration of compliance of the pressure sensor XMP I with functional safety integrity level SIL3 requirements. The thesis is divided into three parts. The first part deals with the concept of functional safety, defines the basic concepts of functional safety, compares the approaches of functional safety according to selected standards and provides a general procedure for the functional safety determination. The second part deal with technical systems reliability defines reliability indicators and describes selected types of reliability analysis used in practice, especially the FMEA/FMECA analysis. Third part deals with the reliability analysis of the sensor XMP i and with evaluation of results.

Emergency lighting control

The aim of this thesis is to design a system for monitoring and control battery charge of emergency lighting. Discuss and choose components with respect to cost and accessibility. Design full circuit scheme and PCB with chosen components is provided. Designed system is compiled and tested in order. The item of discussion is measured discharging characteristics and conditions of accumulator.

Drivers of soil moisture trends in the Czech Republic between 1961 and 2012

Soil moisture dynamics and their temporal trends in the Czech Republic are forced by various drivers. Our analysis of temporal trends indicates that shifts in drought severity between 1961 and 2012 and especially in the April, May, and June period, which displayed such results as a 50% increase in drought probability during 1961–1980 in comparison to 2001–2012. We found that increased global radiation and air temperature together with decreased relative humidity (all statistically significant at p < 0.05) led to increases in the reference evapotranspiration in all months of the growing season; this trend was particularly evident in April, May, and August, when more than 80% of the territory displayed an increased demand for soil water. These changes, in combination with the earlier end of snow cover and the earlier start of the growing season (up to 20 days in some regions), led to increased actual evapotranspiration at the start of the growing season that tended to deplete the soil moisture earlier, leaving the soil more exposed to the impacts of rainfall variability. These results support concerns related to the potentially increased severity of drought events in Central Europe. The reported trend patterns are of particular importance with respect to expected climate change, given the robustness and consistency of the trends shown and the fact that they can be aligned with the existing climate model projections. E-learning in public service

In the era of post-industrial society/world knowledge becomes ever more important source of wealth. This process is being reflected by public service. Part of the changes in the public service is also the term 'e-learning', alias education supported by web technologies. This new method is currently expanding also into the area of civil servants education and therefore I have decided to write my diploma thesis on "E-learning in public service". The aim of the thesis is above all to analyse current civil servants' in the public service education, to explain the term of e-learning, and last but not least to controvert or confirm the hypothesis whether the e-learning way of education of public servants has a chance to spread or even make it to the top of preferred forms of education.
