Study of Reversible Adsorption of Nucleic Acids on Solid Surfaces

Magnetically driven separation techniques using magnetic solid carriers are one of modern methods to speed up and facilitate the previously used separation and purification procedures. The use of magnetic particles in biology imposes strict requirements on physical, and chemical properties of the particles, including low toxicity, biocompatibility and non-interference with the chemical environment in diagnostics. The aim of this study was to evaluate carboxyl-functionalised magnetic non-porous P(HEMA-co-GMA), P(HEMA-co-EDMA), PGMA, silica-coated lanthanum manganese peroskvite La0.75Sr0.25MnO3 and thermosensitive poly(N-isopropylacrylamide) microspheres – P(NIPAAm) for DNA isolation from different types of complex food and environmental samples containing PCR inhibitors. The solid-phase reversible immobilisation (SPRI) of nucleid acids on microsphere surface and the release of adsorbed DNA were optimised. DNA from real samples (milk products and probiotic food suplements, mouse faeces) was apparently adsorbed on solid particles from the aqueous phase system composed of 16% PEG 6000 and 2M NaCl. The conditions of the subsequent release absorbed DNA to the elution buffer (pH of elution buffer, temperature and time of elution) were optimized. The quality of eluted DNA and the presence of target DNA were examined by PCR and q-PCR using domain-specific Bacteria and genus-specific Lactobacillus primer set. Real-time PCR was used for an estimation of the PCR interference by comparing the amplification efficiencies of purified DNA containing solid nanoparticles with the DNA standards free of any nanoparticles

This paper describes the design of control, measurement and power electronics department for dynamic loading of electronically commutated motors with electric brakes. After comparing the advantages and disadvantages of possible conceptions the proposal focuses on electric brake operating in generator mode and the value of load is controlled by the current dissipated in a dummy load.

Analysis of Czech return migration and social remittances - A case study of Ireland Abstract The thesis connects two relatively young concepts - social remittance and return migration, both happening on the background of transnationalism. This Master's Thesis is a case study of Czech migration to Ireland. The thesis aims to identify the return motives and the so-called breaking point - when and under what circumstances the decision to return is made, whether there is a potential for the transfer of social remittances to the non-migrant population in the country of origin and how migration strategies, acquisition and the spread of social remittances differ for low-skilled and high-skilled migrants. The research is based on a qualitative approach. It allows a deeper and closer understanding of the return motives, transnational relations, the acquisition, transfer and impact of social remittances, their character and mutual connections among the selected group of informants. For that reason, the research method uses semi- structured interviews with two groups of informants - the Czechs living in Ireland and the Czechs who already returned from Ireland to the Czech Republic. This diploma paper deals with the migration problemacy and it primarily focuses on one of the most important elements, which affect migration - i.e. migration policy. We may polemize about the actual influence of migration policies on the final size and structure of migration flows, about the effectivity in fulfilling of their objectives; however we must agree on the fact, that migration policies represent an important factor, which affects migration movements. In this work we therefor aim to explain how migration policy arises, what influences its formation and shape and what kind of instruments we may subsequently use to value its fruitfulness. Seeing that in current time the labourmigration is the most common type of the total migration, we focused on the actual situation in the labour-migration policy and we were trying to find out which kind of instruments are used in this area as well as what types of programmes are offered. We pay attention to this problemacy not just from the view of Czech republic, but furthermore from the position of Great Britain, because we presume, that because of its history Britain has multiyear experience in this area, so thus it can be a great source of inspiration for Czech republic. The role of Frontex agency in the migration crisis and attitudes towards its development within selected European Union institutions

This diploma thesis deals with the role of Frontex in protecting the EU's external borders during the migration crisis and its attitude towards its activities within the selected bodies of the European Union. In particular, it examines the period since the beginning of the migration crisis in the spring of 2015, until September 2016, when Frontex was reformed and the new European Border Guard and Coast Guard continued to be called Frontex. The aim of the thesis is to find out what attitudes to the role of Frontex were among the groups of the European Parliament. With regard to Schengen area without internal borders within the EU, an integrated border management system has been created to effectively manage the crossing of external borders and to address migration challenges. For controls at the EU's external borders, various instruments and agencies have been set up, such as Frontex. The theoretical part of the thesis therefore deals in more detail with the Schengen area and the functioning of Frontex, including its specific competences. The phenomen of environmental migration in the context of the floods: case study of rural areas in the South Bohemian Region

The aim of thesis is the environmental migration in rural areas of the South Bohemian Region. The thesis focuses on the migration because of the floods which took place there in 2002 and 2013. These floods are considered one of the most destructive floods in the last century. The aim of this work is based on mixed research in order to determine whether there is the environmental migration in selected municipalities and furthermore, how people react to these events. It also analyzes the impact of external factors such as geographic location, awareness and role of the municipal village on the migration. The research is based on qualitative and quantitative methods. Based on the research it could be said that the environmental migration does not exist in the four selected villages. In conclusion, the above selected external factors do not affect the migration of people due to the floods. In other words, they do not affect human decision whether to stay in the village or leave it. Ongoing expansion of digital images requires new methods for sorting, browsing, and sear- ching through huge image databases. This is a domain of Content-Based Image Retrieval (CBIR) systems, which are database search engines for images. A user typically submit a query image or series of images and the CBIR system tries to find and to retrieve the most similar images from the database. Optimally, the retrieved images should not be sensitive to circumstances during their acquisition. Unfortunately, the appearance of natural objects and materials is highly illumination and viewpoint dependent. This work focuses on representation and retrieval of homogeneous images, called textu- res, under the circumstances with variable illumination and texture rotation. We propose a novel illumination invariant textural features based on Markovian modelling of spatial tex- ture relations. The texture is modelled by Causal Autoregressive Random field (CAR) or Gaussian Markov Random Field (GMRF) models, which allow a very efficient estimation of its parameters, without the demanding Monte Carlo minimisation. 1 Abstract International migration has an impact on the economy in both the receiving and the sending countries due to the fact that foreign workers will 'reinvest' the financial remittances in their home country and support the domestic economy in the source state while fulling the demand for work in the receiving country and improving the financial climate in the host state. Work agencies and the client system are some of the most important details of this system that allows migrants to get a job and often facilitates participation and provides additional services to organize their stay in the host country. The theoretical basis of this thesis is the migration network theory and also the neoinstitutional theory with its concept of limited human rationality as well as a concept of institutional "limitations" in the context of created institutional rules. In the theoretical part of this work the agency and the so-called client system, which performs the same duties on an illegal level, - were explored as institutional units in their historical, real, social dimensions. Impact of the Ukrainian conflict on population mobility

Since spring 2014, an armed conflict has been taking place in the eastern part of Ukraine between the Pro-Russian separatists and the Ukrainian state. As a result of this armed conflict, there was a stream of forced migration. The statistics speak up to about 1.7 million people internally displaced and about a hundred other hundreds of people seeking international protection abroad. Of those seeking international protection, more than 200,000 have been granted temporary asylum in Russia. This thesis examines the impact of the Ukrainian armed conflict on the mobility of these displaced persons. The first way to study the impact of a selected conflict on population mobility is quantitative data analysis. In this analysis are compared the data on conflict intensity with data on internal migration in Ukraine and international migration to the states Russia, Poland and Czechia. The second way this diploma thesis examines the impact of the conflict on population mobility is based on the research of ten Ukrainians who left for Czechia from conflict areas and whose primary reason was the military conflict. The results show that there are links between the intensity of the military conflict and the migration strategy of the people of Eastern Ukraine.
