Probing natural molecules with PPAR- to reveal potent agonist against Cancer

Práca sa zaoberá hľadaním prírodnej molekuly s potenciálnymi agonistickými vlastnosťami proti rakovine. Cieľom práce bolo predovšetkým pochopiť súvislosť medzi receptorom PPAR- a rakovinou. Následne boli molekuly zo šiestich databáz virtuálne skrínované a dokované k nukleárnemu receptoru PPAR- za využitia počítačom podporovaného dizajnu liečiv. Najlepšie molekuly podstúpili ďalšie preskúmanie, vrátane dynamickej simulácie a štúdie viazania voľnej energie. Overenie bezpečnosti najlepších molekúl bolo vykonané predikčnými modelmi ADMET. Dôležitou súčasťou práce bolo porovnanie novoobjavených molekúl disponujúcich agonistickými vlastnosťami proti rakovine s už známymi na trhu. Konkrétne Troglitazón a Rosiglitazón slúžili ako štandardy, ktoré sa vyznačujú vysokou afinitou k receptoru PPAR- a prejavili antineoplastické účinky v rôznych ľudských malígnych nádoroch, vrátane rakoviny prsníka, hrubého čreva a pankreasu. Na druhej strane, tieto dve syntetické molekuly čelia varovaniu zo strany FDA (Úrad pre potraviny a liečivá) v dôsledku ich nepriaznivých vedľajších účinkov. V rámci práce sa podarilo objaviť prírodné molekuly, ktoré vykázali lepšie vlastnosti ako aktuálne štandardy. Tieto sľubné výsledky si zaslúžia ďalšiu pozornosť. Jedna z molekúl by mohla potenciálne viesť k vývoju nového lieku.
Analysis of extensions for mounting a harvester head on an excavator

The introduction of the thesis deals with a complete research of individual components that need to be mounted on an excavator to make it possible to use it for forest harvest. An integral part of the research is a detailed market research, thanks to which various designs of manufactured attachments were analyzed. The research part is followed by a description of the selected components of the assembly, which form an excavator attachment for its use in forest harvest. The following chapter already describes the author's own designs of the extensions design in three variants. The key part of the thesis is dedicated to dynamic simulation of the harvesting process and strength simulations of the designed extensions. Using dynamic simulation, the course of the force effects acting on the attachment pin, which arise during the process of felling a tree, was determined. The maximum force effects were determined from the results of the dynamic simulation, which were then applied in the strength analysis. The finite element method was chosen for the calculation of the strength analysis. The final form of the individual design variants of the extensions arose from the performed strength analyses. The conclusion of the thesis evaluates the achieved results and a detailed comparison of the created design variants of the extensions.
Dynamic simulations of linear displacement system of CNC milling machine

This thesis deals with dynamic simulations of a linear feed system of a CNC milling machine in Adams View. Dynamic simulations enable better and faster decision making when designing new machines than prototyping. The aim of this work is to compare the torque waveforms on ball screws at working feed and at quick feed. The next task is to measure the forces acting on the load body. The values were obtained by simulation in Adams View and measurement on a CNC milling machine MCV Quick 754. The results were exported to Matlab. The resulting torque waveforms were compared, where they are relatively identical during working feed, with large differences during rapid feed.
Dynamic model of a heat pump with plate heat exchangers and ecological refrigerant

For the optimization of a heat pump, there is a need to possess its dynamic model. One of the freely available tools for creating such a model is the language Modelica and its open-source compiler OpenModelica. The thesis aims to research options for creating dynamic models of refrigeration circuits with the ecological refrigerant R290 and its components in Modelica, to create the circuit and subsequently validate it using previously measured data. Models of basic components of heat pumps - compressor, condenser, evaporator and expansion valve are created in a way described in the thesis. A simplified model of a refrigerant loop is also created
Examining the Impact of Demand-Side Management Through Dynamic Simulation: Integrating Smart-Grid-Ready Control of Residential Heat Pumps

This paper investigates the suitability of air-to-water heat pumps for smart grids, with a particular focus on load shifting. The research uses dynamic building energy simulations to show that the utilisation of these features effectively addresses the energy consumption challenges of heat pumps while ensuring that heating and domestic hot water demands are met. The implemented control system successfully shifts the morning and evening peaks to the daytime period, resulting in a modest 2% increase in total energy demand. These results not only demonstrate the balancing of heat pump energy consumption, but also provide insights for the development of objective functions. Such functions can play a key role in supporting the smooth integration of renewables into the wider energy framework.
Dynamically balanced resonance adhesive tester

This diploma thesis is dedicated to the development of mobile resonance-adhesion tester TriTec. The aim of this thesis is to improve the parameters of the tester in comparison to the previous version of tester, especially to minimize the vibrations of the tester while running without load, and to the make long-distance transportation of tester easier. The first part of thesis was aimed on history of development of TriTec tester, causes of it´s problems and on analysis of vibration minimization possibilities. As a best solution is considered a rotational balance mechanism, which was designed in next part of thesis. After that a new front wheel drive and an user interface conception were designed. The modified tester reaches all the deserved parameters and is able to compete static resonance-adhesion testers by it´s lower price and high mobility.

This master’s thesis deals with a strength analysis of the main frame and loading arms of a skip loader. For a calculation of load states is used a dynamic simulation and its results are used as a boundary conditions for the strength analysis using finite element method (FEM). The results of the analysis is evaluated and on theirs basis are designed a construction improvements. The part of the thesis is drawings documentation of the construction improvements.
The trailer for a chipper

The goal of this thesis is structural design of the trailer for a LASKI chippers and to create a construction drawings. Important part is information about legislation rules, which this trailer has to fulfill. The aim of this work is to do a dynamic simulation using multi-body system. Results of this simulation are used for a stress analysis witch using a fine element method.
Trailer frame VARIANT 200 for transport cabel spools

The aim of this thesis is an analysis of the construction of a cable trailer VARIANT 200. For the calculation of load states was used a multi body system in program MSC.ADAMS. The results from these calculations were used as lifting conditions for strength control with finite element method (FEM). All acquired results were analysed and according to them changes were made to the frame construction. Part of this thesis contains drawings documenting the above mentioned changes.
The study of the behavior of track chassis using dynamic simulation

This thesis deals with the study of the undercarriage, their components and organization. Further, this work is focused on selected types of machines with tracked chassis moving in forestry or for operation in forest ground. The second part of the thesis deals with the design of the tracked chassis for the forwarder and the subsequent verification of the functionalities by dynamic simulation.
