Monitoring of the fatty acid profile of pates

This thesis provides information on the nutritional values of different types of meat pates. It is primarily focused on the determination of fat and fatty acids content using gas chromatography with a flame ionization detector. The theoretical part focuses on the description of meat pates, primarily their composition. This part contains characterization pf fatty acids, methods of their extraction and analysis. The last chapter in the theoretical part is dedicated to the description of the methods which are used to determine the content of carbohydrates, crude protein and water. In the experimental part, a standard, manufactured pâté and 20 pates that were purchased were analyzed. All of them contained pork and different types of liver. In the first step, the heating time of the standard pate was optimized for the safe inactivation of Listeria monocytogenes. For the extraction of lipids was chosen method with the SOXTHERM extractor and hexane as solvent. The yeld was compared with traditional Soxhlet extraction. Following, the yield of extraction with the flexIKA extraction and petroleum ether solvent was also compared. The extracted fat was analyzed by gas chromatography. With the help of this analysis, we obtained information about the qualitative and quantitative representation of fatty acids in the samples. All samples were compared with a standard pate made by us. Carbohydrate analysis was performed spectrophotometrically according to Dubois. The results were compared with the crude protein content, which was determined by the Kjeldahl method.
Proteom rostlin v reakci na abiotický stres

Plants as a sessile organisms have evolved complex mechanisms to cope with unfavorable environmental conditions. This diploma thesis summarizes current state of knowledge and focuses on light, temperature and phytohormones in plant abiotic stress responses. The practical part is devoted to proteomics and completes and extends the project started during my Bachelor program. Transgenic Arabidopsis seedlings carrying inducible barley cytokinin oxidase/dehydrogenase (CaMV35S> GR> HvCKX2) were profiled to elucidate proteome-wide responses to down-regulation of cytokinin levels under heat stress at standard (80 umol.m-2.s-1) and low light intensity (20 umol.m-2.s-1). In total, 2-DE analysis and mass spectrometry analysis revealed 63 differentially abundant proteins involved in diverse metabolic processes. These data provide evidence of a link between temperature, light and cytokinin signaling in Arabidopsis and will be used in the modeling of cytokinin-light-temperature interactions.

This thesis includes literature data on the topic Sources of Protein in Human Nutrition. In the first part, the protein itself is described in detail, as a chemical structure in human physiology, structural units of protein at the level of biochemistry of amino acids and their brief description in human physiology, recommendations on the use of daily protein intake for various social groups, detailed information on animal protein sources and plant origin. The second part of the work reflects the problems connected with the difficult situation of individual territorial groups under the threat of "protein hunger". This work provides examples that serve as a barrier in the sphere of food production.
The effect of 14-3-3 protein on intradomain interaction of ubiquitin ligase Nedd4-2

EN The human ubiquitin ligase Nedd4-2 (NEDD4L) ubiquitinates a wide range of membrane proteins and receptors, playing a key role in maintaining homeostasis. This enzyme is regulated by phosphorylation and subsequent interaction with 14-3-3 proteins, which primarily affects its ability to interact with various substrates. However, very little is known about the molecular basis of this protein-protein interaction. In this work, we focused on biophysical characterization of the role of individual phosphorylation sites and also on mapping the structural changes in the Nedd4- 2 protein induced by 14-3-3 protein binding. Our experiments using analytical ultracentrifugation methods revealed that two phosphorylation sites Ser342 and Ser448 are primarily required for stable binding of Nedd4-2 to 14-3-3 proteins. The crystal structure of the 14-3-3ηΔC:Nedd4-2335-455 T367A complex than revealed the simultaneous binding of both phosphorylated residues to the binding groove of 14-3-3 protein. Subsequent modeling based on small-angle X-ray scattering and chemical cross-linking data combined with mass spectrometry indicated extensive structural changes in the individual domains of the Nedd4-2 protein. My bachelor thesis deals with the analysis of eating habits of health education students and physical education students at the Faculty of Education of the University of South Bohemia. The theoretical part is focused on the basic components of nutrition, energy intake and expenditure, nutritional directions, physical constitution and adolescence. In the practical part the data provided by the students who record the composition of the diet into the application kaloricketabulky.cz. The focus of the thesis is on the intake of proteins, carbohydrates, fats and energy intake.
Algorithmic prediction of functional changes within protein sequence

The Following thesis provides a theoretical background to the biology of a naked mole-rat (Heterocephalus glaber), highlighting its unique characteristics that could potentially be explained by altered epigenetic mechanisms. In order to identify potentially functional evolutionarily changed sites in epigenetic modifiers, we developed a novel approach to protein scanning through the utilization of multiple-sequence alignments and protein annotations available in public databases. The key result is computer software and analysis of the obtained results, identifying candidate proteins with potentially altered functions.
Detection of iron-induced proteins and their effect on anthracycline production in submerged culture of Streptomyces coeruleorubidus

In this bachelor's thesis, proteins from the group of enzymes - esterases and their possible induction after the addition of iron to the culture medium were analyzed. Furthermore, the effect of the addition of Fe2+ cations on daunomycin production under submersion cultivation conditions of Streptomyces coeruleorubidus culture was evaluated. The work is divided into two parts. The first part introduced the genus actinobacteria called Streptomyces, their occurrence, properties and possible benefits due to the production of secondary metabolites. In the second, experimental part, the use of the Streptomyces coeruleorubidus strain was described. The effect of iron addition was evaluated by HPLC analysis and native polyacrylamide electrophoresis (native PAGE). HPLC analysis showed that the addition of iron in the form of FeSO4 to the medium increased daunomycin production by approximately 21.5-fold. During native PAGE and after staining, proteins, i.e. esterases, most likely from the groups of and esterases, were detected. Those were more or to a greater extent produced by the Streptomyces coeruleorubidus if the medium contained iron.
