Design of steel-concrete composite structure of road box girder bridge on by-road of Bludov
Kuba, Michal ; Marván, David (referee) ; Nečesal, Petr (advisor)
The aim of this work is design and assessment of the box-girder road bridge on by-road of Bludov. Bridge is designed as composite steel concrete bridge with 5 spans with distances between supports of 31,25 m; 40,0 m; 60,0 m; 60,0 m and 40,35 m. Main bearing structure is designed as open steel box-girder with reinforced concrete deck. Box girder is braced with longitudinal and transversal bracings. Main structure is separated for each traffic direction. The bridge crosses river Morava and railway in 2nd and 3rd span, respectively. The category of road I/44 is S 21,5/100. The height of steel box girder is designed as 2,5 m. Bridge will be built by incremental launching method, with in-situ casted concrete deck afterwards. The bridge is designed according to current standards. Supporting elements are designed from steel of class S 355 and concrete of class C35/45.
Interactive web applications for teaching purposes in the field of signal processing
Kuba, Michael ; Rajmic, Pavel (referee) ; Záviška, Pavel (advisor)
This thesis deals with selected topics of signal processing. The aim of the project is the implementation of four interactive web applications in JavaScript, intended for teaching purposes. These applications deal with following topics: rational number resampling of the discrete signal, mean and root mean square signal values, discrete linear and circular convolution in 1D, and discrete cross-correlation. The thesis is divided into the theoretical and the practical part. There is a theroretical foreword of topics the applications deal with, including an introduction to signals followed by a description of functionality and the user interface of created applications.
Acquisition and evaluation of datasets from the vibrodiagnostic model
Kuba, Marek ; Marada, Tomáš (referee) ; Zuth, Daniel (advisor)
The master thesis deals with the use of artificial intelligence methods in order to classify static and dynamic imbalance. The measured data has been obtained with the help of the vibrodiagnostic model. The measurements were processed in the Matlab program environment and with the help of the classification learner app afterwards. The measured data has been processed by the common machine learning classifiers. The success rates of the particular classifiers and the measurement courses are further described in the thesis.
Genetic Algorithms - Multi-core CPU Implementation
Studnička, Vladimír ; Kuba, Martin (referee) ; Matoušek, Radomil (advisor)
his diploma thesis deals with creating the most universal library of genetic algorithms in C++, as much as possible, implemented with the certain number of universal operators, and then with testing created library on some examples. Library must support multi-core processors, implementation will be done over OpenMP. The library will be tested on three examples in all. The first two examples are mathematical functions, that are used just for genetic algorithms testing. Last problem for test is N-Queens problem. Finally we will use genetic algorithms to try find solution for Eternity II puzzle, there is declared a 2 million bounty for full solution.
Temperature regulation in technological process
Kuba, Marek ; Marada, Tomáš (referee) ; Zuth, Daniel (advisor)
My bachelor thesis deals with the concept and construction of my own equipment for temperature control in individual fermentation tanks which is commonly used in winery industry. The work is divided into two parts. The literature research contains brief description of suitable technology and temperature sensors. My practical part is focusing on the selection of individual components and evaluation of the results. It was also necessary to create a program for temperature control, design documentation and comparison of my final product with others commercial mechanism. In the end of my work I give summary of various recommendations for the practical use of my own product.
Steel structure of airport hangar
Kuba, Michal ; Balázs, Ivan (referee) ; Pešek, Ondřej (advisor)
The aim of this work is design and assessment of the steel roof structure of a airport hangar for sports aircrafts in Zlín. Ground plan dimensions are 36 x 78 m. The main supporting system is composed of truss girders. On the one side the truss girders are pinned on column, on the other side they are pinned into foundation. The truss girders are arc-shaped, with structural height 1,5 m. Distance between main structures are 6 m. Construction is designed as system without purlins. Bars of the steel structure are from rolled profiles and are designed according to current standards. Supporting elements are designed from steel of class S 355 and S 460.
Business Plan Proposal for a Company
Kingová, Jana ; Kuba, Martin (referee) ; Konečná, Zdeňka (advisor)
The content of this diploma thesis is a proposal for a business plan for the establishment and development of a company. Specifically, it is a business plan for a café called Ollie, which is also a community center, a workshop and a place to work situated in Brno Komárov, near the Komec sports complex. The first part consists of the theoretical basis of the work containing an explanation of the concepts needed to write the practical part of the thesis. The practical part contains an analysis of the current state and the actual design of the solution. Methods used are for example PESTLE or Porter’s Analysis.

Novel fluorescent nucleotides for metabolic labelling and for the construction of DNA probes
Kuba, Miroslav
The aim of the thesis was to synthesize new nucleosides, nucleotides and the corresponding DNA probes bearing various fluorescent labels, which can be used for bioanalytical applications. In the first part of the thesis, 2'-deoxycytidine and the corresponding nucleoside triphosphate bearing tryptophan-based imidazolinone fluorophore were synthesized by Sonogashira cross-coupling reaction. The fluorophore showed sensitivity to pH and viscosity. Nucleotide was used for the construction of modified oligonucleotides (ON) and DNA by primer extension (PEX) or polymerase chain reaction (PCR). Labelled ON probe was used for sensing interaction with single-strand binding protein, which resulted in increased fluorescence intensity of modified ON. Next, thymidine and thymidine triphosphate labelled by bezylidene- tetrahydroxanthylium fluorophore were synthesized by copper-catalyzed azide-alkyne cycloaddition (CuAAC). Fluorescence of the fluorophore is dependent on the polarity and viscosity of the environment. Incorporation of the modified nucleotide into DNA, by PEX or PCR, led to dramatic increase of the fluorescence presumably due to the interactions of the fluorophore in the major groove. Unfortunately, the modified nucleotide was not suitable for in cellulo imaging due to its cytotoxicity. The modified...
