A design of manipulator for laser engraving device
Hanák, Jan ; Knoflíček, Radek (referee) ; Tůma, Jiří (advisor)
This thesis presents a research in the field of laser technologies used in industry and systems for laser marking of rotating parts. The output is a design of a laser cell that serves to mark rotating parts and that can be incorporated into an automated production line. The concept is developed in the form of a 3D model which is accompanied by necessary technical calculations and a justification of the choice of used components.
Study of adsorption of organic pollutants on wheat bran biochar
Hanák, Jan ; Pořízka, Jaromír (referee) ; Diviš, Pavel (advisor)
This diploma thesis explores the application of the Diffusive Gradients in Thin Films (DGT) method for determining and monitoring the concentrations of dissolved pharmaceuticals and organic dyes in water and food. The theoretical part introduces the issue of the occurrence of these organic substances in water and food, the principles of the DGT method, the characteristics of adsorption and adsorption isotherms, and the introduction of biochar as an adsorbent. The experimental section primarily focuses on optimizing and validating adsorption gels containing commercial biochar and unmodified biochar from wheat bran for the adsorption of specific substances using the DGT method. Oxytetracycline was primarily tested from a selection of antibiotics and malachite green from organic dyes, which is widely used in aquaristics. The results demonstrate a comparison of the effectiveness of adsorption using these two types of adsorbents and their characteristics, such as infrared spectroscopy analysis or images from a scanning electron microscope. From the Langmuir isotherm, the maximum adsorption capacity for oxytetracycline on commercial biochar was found to be 314,065 mgg-1 and the maximum adsorption capacity for wheat bran biochar was 89,720 mgg-1. The maximum adsorption capacity of commercial biochar for malachite green was 439,406 mgg-1. The work further emphasizes the potential of the DGT method as a tool for food analysis and investigating contaminants in water systems, which further move into the food chain.
Development of antimicrobial wound healing nanostructure biomaterials
Hanák, Jan ; Wikarská, Monika (referee) ; Márová, Ivana (advisor)
The bachelor thesis is focused on optimizing the production of nanoparticles with a bioactive component - especially on enzymes and antibiotics encapsulated into lipozomes. Nanoparticles with active substances could be used in the field of pharmacy or cosmetology. In the theoretical part the composition of the skin, skin injuries, and wound covers are described. The materials used for the preparation of nanoparticles and nanofibers are characterized as well as processes to produce such nanomaterials. Enzymes, antimicrobial drugs, and methods for determining antimicrobial activity are mentioned in this section, too. The practical part deals with the characterization and optimization of the formation of lipozome particles with encapsulated enzymes. The formed particles were divided into two groups. The first type of particles were pure lipozomes, while the second ones enriched by polyhydroxybutyrate to increase the encapsulation efficiency and improve the stability of the particles. The enzymes bromelain and lysozyme were selected for the highest encapsulation efficiency. The encapsulation of these two enzymes took place in two ways - by thin film hydration and by sonication method. Both methods showed high encapsulation efficiency, but the sonication method proved to be unsuitable due to enzyme denaturation. The proteolytic activity of the released enzymes from the particles was measured. The stability of the particles was monitored in a physiological and aquatic environment by measuring the zeta potential for up to 28 days. Moreover, the enzymes, ampicillin, and particles created by sonication were subsequently subjected to antimicrobial tests by using two test bacterial strains. Finally, alginate particles with encapsulated enzymes were also formed, then the encapsulation efficiency and proteolytic activity of the enzyme were measured. In this study also other methods of encapsulation of active substances are outlined, which could be possibly used to produce nanomaterials suitable for wound healing.
Calculation of the position of the support relative to the spindle on the machine for drilling braking holes
Hanák, Jan ; Knoflíček, Radek (referee) ; Tůma, Zdeněk (advisor)
This bachelor thesis do a research on trains wheels, axles, trains wheelsets and an injection hole which serves for nondestructive disassembly of wheels from axle. The output of this work is an evaluation of possible ways of injection hole drilling and a calculation proposal of drilling spindle tilt versus in relation to support in the course of drilling.

Bylo provedeno hodnocení kontaminace vodního prostředí na šesti vybraných lokalitách, které se nacházejí na malých tocích povodí Labe nad a pod čistírnami odpadních vod. Sledovány byly lokality Pacov, Brloh a Prachatice. Byly prováděny testy akutní toxicity a subchronické toxicity. Zpráva obsahuje pouze stručnou anotaci výsledků projektu a v příloze články, které byly k projektu publikovány.
