Development of the Construction of Residential Property in Brno-Židenice

The diploma thesis deals with the development of the construction of residential properties in the city district of Brno - Židenice. The thesis is divided into two parts. The first part contains general information about the city district, the development of population, houses and flats and summarizes the historical development since the first settlement. The second part describes the development of the construction of residential properties in individual time periods, including the type of construction, urban development of the city district, from the oldest construction to the analysis of the development of the city district in future years. Attached to the work is a well - arranged map that graphically illustrates the settlement of the city district in individual periods.
Electrophotocatalytic wather disinfection

This work focuses on the photoelectrocatalytic water disinfection by applying electrical voltage onto an electrode system. Working electrode consist of electrical conductor covered by thin film TiO2, because of its very important photoactivity. Disinfection effect was monitored on Gram-negative bacteria E. coli under different conditions. Inactivation of bacterial cells in contamined aqueous solution, first experiment was observed only with presence of oxygen, later reactor was irradiated with UV-A compact blacklight with intensity of 4 mW• m-2 or electrical voltage of 1V was applied to electrode. By adding both effects at the same time I was verifying photoelectrocatalysic water disinfection on chosen microorganism.
Rosehip – the source of natural compounds

The goal of this bachelor thesis was to study the antimicrobial activity of various extracts of rosehip tea. As well as to determinate concentration of biologically active compounds, polyfenols and flavonoids, and antioxidant activity in these teas. Teoretical part describes basic information about Rosa canina, general classification of tea, summary of biologically active compounds and basic methods of extraction. Practical part is focused on determination of anioxidant acivity, comcentration of polyfenols and flavonoids in tea extracts and detection of animicrobial activity of these extracts against bacterial strains Serratia marcescens and Micrococcus luteus. The results of this theasis show that aqueous extracts of rosehips tea are animicrobialy active. It was also found, that these extracts have hight contents of polyphenols and flavonoids.
Production of biogenic amines in double moulded cheese

For production of double moulded chesses are used lactic acid bacteria, which can be present in a form of non-starter lactic acid bacteria or as starter or adjunct culture. Genera Lactobacillus spp. and Enterococcus spp. are prevalent microorganisms present in these cultures. Of course, these microorganisms are for us interesting because of their possibility of coagulation, proteolytic possibility, probiotic function and antibiotic resistance, but especially because of their decarboxylation abilities. Bacteria contain decarboxylation enzymes, which are able to decarboxylized free amino acid, which rising at proteolysis during process of manufacturing and cheese ripening. Biogenic amines are the result of proteolytic activity. Biogenic amines occur practically in all foodstuffs as a common product of metabolic processes. BA are mainly presented in fermented food (cheeses), where rice just microbial action. Typical representatives of biogenic amines, which occurs in double moulded cheeses (Sedlčanský Vltavín, Bresse bleu) and in blue cheeses (Bleu des Causses, Bleu d'Auvergne) are cadaverine, putrescine, tyramine a 2 fenylethylamine and in much smaller amount histamine, spermidine and spermine too. On assessment concentration of BA is used high pressure liquid chromatography with reverse phase (RP HPLC) with utilizing simple direct derivatization with dansyl chloride and detection by UV VIS detector.
Microbial contamination of photographic materials

This diploma work is focused on a study of microbial contamination of photographic materials. In the first part of this work the information about the archive and museum microbial contaminations were collected. The most frequent contaminants of photographic collections – Penicillium chrysogenum and Aspergillus niger were studied. Their activity on photographic papers Fomaspeed and Fomabrom was evaluated. It was found that at the given experimental conditions the image layers of Fomaspeed paper are resistant to fungi mycelium growth. On the other hand, the Fomabrom paper image layer was irrecoverably damaged by action of fungi mentioned above. The degree image layer deterioration was evaluated with the aid of sensitometric curves.
Jaroslavice – place in the landscape

South Moravia has a good quality soil. Changing climatic conditions and an unhealthy human approach result in drought and soil erosion. Soil, like our nutritionist, is very rare. Therefore, in my work I deal with the proposal of a research institute for soil protection, which I place on the site of a former manor court in Jaroslavice. The Institute consists of three buildings that define a common yard. The location of the complex complies with the direct connection to the adjacent experimental field. Jaroslavice, as a municipality in the warmest and driest area in the Czech Republic, is ideal for research.
The effect of silver on yeasts photokilling process efficiency

In this diploma thesis, photocatalytic effect of titanium dioxide with addition of silver was described and demonstrated on inactivation of the yeast Candida vini. Titanium dioxide was prepared by sol-gel process and deposited by printing metod on soda-lime glasses. On the deposit of titanium dioxide was added silver in various concentrations which increased effect of TiO2. Inactivation of yeasts was examined by effectiveness of UV light with intensity 170 and 100 W/m2.
Antimicrobial activity of rosehip teas

The aim of this bachelor theses was to determine the concentration of biologically active substances such as polyphenols and flavonoids and also total antioxidant activity of tea extract of rosehip and also research antimicrobial activity and the effective concentration of the extracts against selected bacteria. Theoretical part is focuses on the description of natural antioxidants and active effect against free radicals, description and distribution of tea, botanical and chemical description od rosehip and basic information about bacteria. There is section describe selected methods for the determination of polyphenols, flavonoids, total antioxidant activity of these extracts, and verification of antimicrobial activity against grampositive and gramnegative bacteria in experimental section. From the results is flow, that the analyzed extracts have no inhibition activity against bacteria. Also was found that aqueous extracts has high proportion flavonoids and polyphenols.
Study of natural compounds of elderberry flowers

The aim of this bachelor thesis was to determine the concentration of bioactive substances such as polyphenols and flavonoids and also the total antioxidant capacity in extracts from flowers of elder. The second part was the subject of an experiment to determine the antimicrobial activity. The theoretical part deals with the description of individual bioactive substances, the description of the elder and the bacteria used. In the experimental part, aqueous extracts were prepared, in which total polyphenols, flavonoids and oxidizing activity were determined. The results of the determination of polyphenols and flavonoids show that in order to achieve a higher content of flavonoids and polyphenols, it is more appropriate to use the highest possible initial temperature for leaching the base flower. No significant differences were found when comparing the antioxidant activity of solutions from the average values. The extracts did not show significant antimicrobial activity.
Aronia – the source of bioactive compounds

The aim of this bachelor thesis is to research the possible antimicrobial activity of aqueous and ethanolic extracts of aronia berries and tea mixture containing aronia. Also, the antioxidant activity and the concentration of the biologically active substances - polyphenols, flavonoids and anthocyanins was determined. Theoretical part contains a summary of basic information about Aronia melanocarpa, chemical composition, use and health effects of substances contained in its fruits. The experimental part focuses on the antimicrobial activity of the extracts on microorganisms Bacillus subtilis, Serratia marcescens and Micrococcus luteus. Farther there is spectrophotometrical determination of antioxidant activity, polyphenolic compounds, flavonoids and anthocyanins. From the experiment results flow that the extracts have almost none antimicrobial ability except tea mixture extracted with hot water. This extract has inhibitory effect on M. luteus and growth supportive effect on B. subtilis. It has been found that aronia fruits and tea mixture are rich in polyphenols and flavonoids.
