Janoušek, Jiří ; Kubíček, Vladimír (advisor) ; Drastík, Martin (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Jiří Janoušek Supervisor: Ing. Vladimír Kubíček, CSc. Title of diploma thesis: Use of SPE in analysis of benzimidazoles I Anthelmintic resistance in parasitic worms is an up-to-date topic. In order to combat this resistance we need to find proper analytic methods of its monitoring. The goal of this thesis is to find a suitable method of benzimidazole extraction, using a solid-phase extraction. During the experiments I used SPE tubes Supelco DSC-C8 (1ml, 100mg). In my thesis I tested the influence of pH and composition of elution agents on recovery of extraction. I performed the extraction in acid, neutral and basic environment. Elution agents consisted of a mixture of acetonitrile and polar phase, with four different mixing ratios. The best results were obtained during extraction in basic environment. For SPE tubes DSC-C8 the most suitable elution agent consisted of 4 parts of acetonitrile and 6 parts of 10% ammonia solution. Keywords: benzimidazoles, flubendazole, mebendazole, solid-phase extraction, resistance, anthelmintics
Study of the effect of the amount of chelating agent addition on the result of monoclonal antibody conjugation
Janečková, Adriana ; Bárta, Pavel (advisor) ; Janoušek, Jiří (referee)
v anglickém jazyce Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Student: Mgr. Adriana Janečková Supervisor: Mgr. Pavel Bárta, Ph.D. Title of rigorosum thesis: Study of the effect of the amount of chelating agent addition on the result of monoclonal antibody conjugation Monoclonal antibodies have become the important tool not only for the therapy, but also for the diagnosis of the infectious, autoimmune or cancer diseases in the last few decades. In addition to their own therapeutic effect, monoclonal antibodies are used as vectors for biological substances with therapeutical and diagnostic importance. Besides bacterial and plant toxins, monoclonal antibodies are also effective in their capability to deliver radionuclides to the site of their specific binding, i.e. the antigen against which they were prepared. The so called radioimmunoconjugates are then successfully used in radiodiagnostic and radiotherapeutical field. Radionuclides can be bound to antibody molecule either directly in the case of halogenation or indirectly. The latter method is the most common one due to a greater stability and suitability for more radionuclides, which can be transported by antibodies. Substances that allow the radionuclide biding to the monoclonal antibody...

Vision-Based Autonomous Navigation Of An Unmanned Aerial Vehicle
Janoušek, Jiří
This article is focused on the real-time detection and recognition of the objects by using acamera located on Unmanned Aerial Vehicle (UAV). The detected objects in the image carry informationabout the next points of the flight plan according to which the drone performs an autonomousflight. The drone can recognize the information encoded in the patterns on the ground by the field ofview of the camera, therefore, it can perform flight manoeuvres consequently. The detection andinformation recognition is performed by a single-board computer and it is based on comparing imageswith a predefined Haar cascade. The single-board computer on the drone recognizes a size andspacing of the individual contrast points which determine the partial information about the next flightpath. Afterward, single-board computer transmits the detected information to the Pixhawk controlunit by using the Mavlink protocol in real-time. The aircraft control unit ensures the accurate positioningof the UAV by sequential execution of the commands.
Production of pressure filter casing
Janoušek, Jiří ; Forejt, Milan (referee) ; Podaný, Kamil (advisor)
The project presents a proposal for the technology of production of the pressure filter casing used in hydraulic systems, which is made of non-alloy carbon steel 12 020. With the help of a literature study of cold extrusion and calculations, the production of the component was designed, which will take place in two operations. Before each operation, the surface treatment is performed by phosphating and soaping and inter-operational annealing must take place. Tool sets were designed for both operations and a press was selected from the company's machine park with a nominal force of 25,000 kN. Finally, a technical and economic evaluation was proved, which shows that the price of production of one piece is CZK 187. The turning point was set at 3 497 pieces per year.
