Construction Technology project microbrewery Highlands

The aim of thediploma thesis is to make the detailed building-technological project of microbrewery Vysočina. It is concentrated on the principles of building organization what makes construction facility, the calculation of electric power and water supply, calculation of the costs of operating the construction site and its graphic representation for chosen construction phases. I also make the project of the optimal machine composition. After that I compile the budget and costing of the microbrewery, the time and financial plan and the evaluation of main sources. I technologically solve the phase of upper shell construction made of reinforced pre-cast construction for building object of the microbrewery Vysočina. I apply the other ways of assembly of upper shell construction and show their evaluation in terms of price and time construction. I make the inspection and test plan, environmental aspects, the source register of working risk, heat-engineering review of building case and fire safety solutions.
Evaluation of the Financial Situation of a Company and Proposals for its Improvement

My master‘s thesis deals with the evaluation of the financial situation of the selected company during the years 2007-2011 which is accomplished by methods of the financial analysis. On the basis of established results, there are formed suggestions which will lead to better financial environment of the company in the following term.

The aim of the bachelor thesis is to solve the technological phase of foundation for the object of a microbrewery “Vysočina”, to establish technological orders about earthworks and foundations. It applies in alternative way the two others methods of foundation and does their price and term evaluation. The other aim is to elaborate the documents of principles of organization of construction includes the graphical illustration of construction facilities and the movements of chosen composition of machines.
Magnetic Beads-Based Electrochemical Techniques for DNA-Protein Interaction Monitoring

Electrochemical techniques, in connection with separation of nucleoprotein complexes at magnetic beads, are suitable for the monitoring of DNA-protein interactions. For the detection of complexes captured at the beads it is possible to utilize intrinsic electrochemical activity of the protein, intrinsic structure-selective signals of the DNA, or indicator DNA substrates tail-labeled with electroactive moieties.
Using Anthraquinone Labeled Nucleotide Triphosphates for Electrochemical Analysis of DNA

Base-modified anthraquinone-nucleotide triphosphates were used for DNA labeling. Anthraquinone modified nucleotides were incorporated into DNA by primer extension with KOD polymerase. In cyclic and square wave voltammetry at HMDE or PGE antraquinone gave a well developed characteristic signal suitable for differentiation of the labeled DNA from unlabeled one.
Electrochemical analysis of DNA using switchable redox moieties

Labelling of DNA with electrochemically active moieties proved to be a convenient way to the development of electrochemical techniques for the sequence-specific DNA sensing. Through combinations of various labels differing in redox potentials, independent redox coding of different DNA sequences or individual nucleobases can be attained. Applications possibilities of electrochemistry in analysis of modified DNAs are further extended by facile monitoring of chemical conversion of reactive groups on DNA during post-labelling with ultimate redox labels. In addition, controlled in situ electrochemical conversions of specific intrinsic and extrinsic DNA components can be utilized to switch their electrochemical signals and improve signal resolution.
Tail labelled oligonucleotide probes for the detection of DNA-protein interactions

DNA-protein interactions can be monitored via different ways. We introduce novel, fast and simple approaches in DNA-protein interaction detection based on electrochemical measurements of DNA alone (structure-sensitive DNA sensing), or DNA modified with osmium tetroxide bearing nitrogenous ligands, or measurements of redox-active moieties enzymatically attached to the end of a DNA substrate thus forming a labeled tail (by terminal transferase).
Redox labelling of nucleic acids for analyzing nucleotide sequences and monitoring DNA-protein interactions

Nucleobase labelling of DNA for electrochemical sensing was attained through chemical modification of thymine bases with osmium tetroxide in the presence of nitrogenous ligands, or via enzymatic incorporation of nucleotide conjugates with redox-active moieties using labelled deoxynucleoside triphosphates. DNA hybridization, primer extension and PCR techniques were used for sequence-specific DNA assays. Tail-labelled DNA substrates were applied to monitor DNA binding by tumour suppressor p53 protein.

The following dissertation is presented in two parts. The main part is focused on the collection of twelve jewellery pieces, which I designed and produced myself. In case of decorative objects I tried to find a balance of shapes, structures and materials. The jewellery is made from organic and natural resources which I often find charming and fascinating. The greatest inspiration for me had to be the world`s natural civilisations, which don{\crq}t regard the jewellery pieces just as decoration but for them represent also magical and spiritual meaning. In the theoretic part of my dissertation I focused on the comparison of jewellery in natural civilisations with the primate jewellery of mankind. In this part of my work I searched for the roots of magic and art in general and I discovered the connections with todays remaining natural civilisations {--} magical meaning of jewellery, use of material and meaningful ornaments. Here I also mentioned the use and representation of jewellery. Attempting to clarify the concept of amullet, talisman and fetish. All written text is supported by picture documentation. In the last chapter I summarised the main focus of my dissertation and reflected on the creative process of my work. The last part of my dissertation contains all photo documentation of the creative process.

We employed single step aqueous phase Suzuki-Miyaura cross-coupling reactions of halogenated 2'-deoxynucleoside 5'-triphosphates with 3-aminophenyl and 3-nitrophenylboronic acid for synthesis of modified nucleoside triphosphates (dNTPs). These dNTPs were then enzymatically incorporated into DNA in Primer extension experiment (PEX). Electrochemical detection using square-wave voltammetry of single-strand modified oligonucleotides (ONs) bearing 3-aminophenyl and 3-nitrophenyl tag demonstated excellent utilization in labeling of DNA.
