Coal humic acids and their applications

The dark-colored humic acids are important parts of less coalified coals. Thanks to their properties, they are used in agriculture, veterinary medicine, industry and in modifications of the soil in the environment. In my work, I included the survey of producers of the coal humic acids and their application. An extraction of these humic acids and a determination of their content in the original raw material has been made on a domestic sample.
Fluorescence spectroscopy in the research of aminoclays

Aminoclay belongs to a large group of organic modified materials based on clays. These materials could find many potential practical applications and, in particular, may be used as a matrix with the ability of the organisation and stabilisation of incorporated substances. The utilisation of aminoclay in the field of medical applications seems highly prospective, but there is the need to visualise the clay system and the processes within it. This essay deals with the issue of aminoclay visualisation by means of fluorescent spectroscopy. Through fluorescent spectroscopy, aminoclay was discovered to be autofluorescent. The autofluorescence of aminoclay is not usable in practical applications because of the low fluorescence intensity. This is the reason for the research into other possibilities for modifications of the internal structure of aminoclay by europium ions. These ions in chelated form are able to be fluorescent and modifications of the external structure are possible by electrostatic implementation of fluorescent dyes. Characterisation of the prepared complexes was provided by spectrofluorimetry and fluorescent microscopy for the purpose of evaluation of the practical application of aminoclay.
Gelation of hydrophobized hyaluronan

This diploma thesis deals with hyaluronan modified by palmitoyl and its gelation. Gels were created from palmitoyl hyaluronan with molecular weight 216 kDa and degree of substitution 11 % in concentrations 15 and 20 g dm-3 in water and concentrations 10, 15, 20 g dm-3 in NaCl and TSB. Also gel from palmitoyl hyaluronan with molecular weight 35 kDa and degree of substitution 10 % in concentrations 20, 30 g dm-3 in NaCl was created. Gels were investigated concerning medical applications. Gels were rigid and had very good properties, which was confirmed by rheology. The physical properties (pH, water content) of gels and stability were investigated. On the grounds of the MTT test, three methods of cell incorporation were suggested. Gels are nontoxic, biocompatible, and biodegradable with nontoxic degradation products and that is why they are excellent aspirants for use in biomedicine.
Study of aggregation process and physical stability of polymeric micelle by fluorescence probe method

Amphiphilic block copolymer poly(ethylene glycol)-b-poly(–caprolactone) (PEG–PCL) was synthesized from poly(ethylene glycol) (PEG) and poly(–caprolactone) (PCL) by using ring-opening polymerization. The structure and composition of this copolymer was determined by IR spectroscopy. Polymeric micelles were prepared by membrane dialysis method and direct dissolution of copolymer in water. The process of aggregation and physical stability in water solution was studied by fluorescence spectroscopy using pyrene and perylene as fluorescent probes.The results of steady-state and time resolved fluorescence measurements indicate that system of PEG–PCL forms both unimolecular and multimolecular micelles, which depends on copolymer concentration. The critical micelle concentration (CMC) was around 0,002 g/L. Measurement of micelles size by dynamic light scattering method suggested that systems with higher concentration form bigger aggregates. In addition, copolymer behavior was explored under physiological conditions. The results show that CMC of copolymer increased in 0,15 mol/L sal solution at temperature 37°C to 0,02 g/L when compared to copolymer in water.
Physical hydrogels based on biopolymers and surfactants

This doctoral thesis studies the properties and behaviour of phase-separated hydrogels. These can be prepared by interaction of polyelectrolytes with oppositely charged surfactants. Negatively charged polysaccharide hyaluronan and cationic surfactant carbethopendecinium bromide (Septonex), whose properties, behaviour and utilization are described in the first two chapters of theoretical part, were selected for this role. Hyaluronan is naturally-occuring in living organisms and is known for his specific targeting to the tumour cells. Septonex is used as antiseptic and disinfectant. Experimental part of this work is focused on examination of the structure and behaviour of these types of hydrogels especially from rheological point of view. The crucial part of this study was to establish mechanical properties and their dependence on environment. This led to design further studies. Structural analysis was held using the methods of ATR-FTIR, ionic chromatography and ICP-OES helping to measure the inner content of entry materials in the gels and supernatants respectively. The last chapter deals with antimicrobial activity. The results of this study indicate a potential usage of these substances in the field of medical applications.
Hydrogels prepared from hydrophobized hyaluronan and micelles

This Master‘s thesis is focused on the preparation of hydrogels based on a hydrophobic derivative of hyaluronic acid palmitoyl hyaluronan in presence of biosurfactant decyl glucoside’s micelles and micelles of surfactant triton X-100. For followed characterization were chosen rheological tests and infrared spectrometry. There was observed influence of preparation method, type and concentration of surfactant and presence of micelles on final features of the hydrogel. Moreover, an experimental test of swelling was performed with interesting results for future research.
Influence of selected parameters on biotechnological production of polyhydroxyalkanoates

The aim of this work is to study microbial production of polyhydroxyalkanoates (PHA). Theoretical part is focused on production of PHA using microoganisms and transgenic plants. Bacterial strain Cupriavidus necator H16 was used for laboratory production of PHA. Various waste oils were used as sole carbon and energy source. Salt of propionic adic and 1-propanol were used as intermediate for 3-hydroxyvalerate monomer (3HV) unit. Incorporation of 3HV to polymer can improve material features of PHA. The major part of experimental work was focused to study influence of aeration (concentration of dissolved oxygen) to bacterial growth, selected metabolic pathways and formation of PHA. Furthermore, influence of aeration to monomer composition of polymer was evaluated. According to experimental conclusion of this work it was approved dependance between aeration and monomer composition of PHA. Moreover, it was approved that higher concentration of oxygen supports bacterial growth and influences PHA content in cells. In addition, NADPH is one of the substrates influencing flux of acetyl-CoA throughout the metabolism; higher intracellular concentration of NADPH inhibits TCA cycle and enhances accumulation of PHA in cells. For this reason, specific enzymatic aktivity of several selected intracelular enzymes were measured, including those enzymes which can generate NADPH.
Rheology of hyaluronane solutions

Předmětem tohoto studia bylo prozkoumat reologické chování vodných roztoků vysokomolekulárního hyaluronanu. Byl studován vliv zvyšující se koncentrace biopolymeru v roztoku, která se pohybovala v rozmezí od 1 do 3 hmotnostních procent, a také vliv vzrůstající iontové síly rozpouštědla, způsobené přídavkem chloridu sodného, na viskoelasticitu a stabilitu těchto roztoků. Pro obsáhlejší popis viskoelasických vlastností roztoků byla použita, vedle běžných oscilačních měření, také metoda ceepových testů, ze které bylo možno určit důležité veličity, jako je procentuální poměr viskozní a elastické složky vzorku, rovnovážná poddajnost, viskozita při nulovém smykovém napětí a retardační čas. Ty byly následně porovnávány s výstupy z jiných typů měření, jako jsou právě oscilační a tokové křivky, nebo nesly dopňující informace důležité pro detailnější popis viskoelastických vlastností těchto roztoků. Ke studiu stability vzorků během namáhání pak byla použita metoda peak-hold, která ukázala na velmi dobou mechanickou i časovou odolnost roztoků hyaluronanu a naznačila hranice, za kterými už dochází k trvalému poškození struktury a degradaci řetězců hyaluronanu a je s němi proto potřeba při manipulaci s roztoky tohoto biopolymeru pro jejich další použití v aplikacích počítat.
Hyaluronan-Micelle Aggregates and their Potential for Nanomedicine Applications

This thesis deals with the study of preparation and characterization of hyaluronan-micelle aggregates. The theoretical part deals with drug delivery systems, characterization of used materials and methods especially fluorescence spectroscopy, dynamic light scattering and turbidimetry. Methods of determination of measured data are summarized in the experimental section. The result section is divided into two subsections dedicated to different preparation methods of hyaluronan-surfactant complexes. Induced aggregates of hyaluronan with Septonex are characterized in terms of their origin and stability, and the results are compared with previously studied surfactants CTAB. In the second part are discussed so-called decorated micelles, their formation, properties and stability.
Study of interactions of the surfactant component of Septonex with selected proteins

The study of protein and surfactant interactions is of great significance in a number of applications, such as the cosmetic, food or pharmaceutical industries and many others. However, they require further study due to their compositional complexity and the limitations of current analytical approaches. In this thesis, the cationic surfactant septonex in combination with two differently charged proteins lysozyme and bovine serum albumin under different physiological conditions (temperature, surfactant concentration, environment and others) was selected to study the interactions. Characterization of protein-surfactant interactions is a very important but challenging task, therefore it is essential to use appropriate approaches to explore the nature of these interactions. In order to unify the information to provide rational models, calorimetric methods (DSC, ITC) and dynamic light scattering were used. Isothermal titration calorimetry monitors the evidence for the formation of the system of the mentioned substances and information on aggregation behavior, differential scanning calorimetry characterizes the thermal stability of proteins and dynamic light scattering made it possible to monitor changes in particle size. Both proteins have been proven to interact with positive septonex, even if the lysozyme molecule is also positively charged. However, significant differences were found between the two proteins. From the obtained results it is evident that the identical charge of the protein with the surfactant has an effect on the intensity of the measurement, although all measured interactions showed an endothermic character.
