Enzymatic system of anaerobic rumen fungi
Novotná, Zuzana
Anaerobic fungi inhabiting the digestive tract of large herbivores secrete a variety of hydrolytic enzymes, including cellulases, xylanases, mannases, esterases, glucosidases, and glucanases, which effectively hydrolyze plant biomass consisting mainly of cellulose and hemicellulose, the most abundant polysaccharides in the biosphere. The immense progress has been achieved in investigation of Neocallimastix, Piromyces, however other genera are less studied. This work therefore concentrated the effort to analyze the hydrolytic system of genera Anaeromyces, Orpinomyces, Caecomyces and fungus KF9 described as new type of fungus belonging to the genus Piromyces. The studied anaerobic fungi grown in batch culture on M10 medium with rumen fluid and microcrystalline cellulose as carbon source produced a broad range of enzymes requisite for degradation of plant structural and storage carbohydrates including cellulase, endoglucanase, xylanase, α-xylosidase, β-xylosidase, α-glucosidase, β-glucosidase, β-galactosidase, mannosidase, cellobiohydrolase, amylase, laminarinase, pectinase and pectate lyase. These enzymes were detected in both the intra- and extra-cellular fractions, but production into the medium was prevalent with exception for intracellular β-xylosidase, chitinases, N-acetylglucosaminidase, and...

Mechanismy regulace aktivity vynašečů auxinu
Křeček, Pavel ; Zažímalová, Eva (advisor) ; Novotná, Zuzana (referee) ; Reinöhl, Vilém (referee)
The morphology of plant body is shaped by genetically coded developmental programme together with environmental factors. The influence of environmental factors on the morphology is much more important in plants that in other multicellular organisms. The developmental programme provides a general scheme that is modified by environmental signals. Phytohormone auxin is a regulator of plant morphogenesis and its distribution in the plant body is an important mechanism controling the growth and development of plants and coordinating the developmental programme with environmentally-induced changes. This thesis investigates factors important for transport of auxin from cells, second part is dedicated to bioinformatic analysis of the transporters from the PIN protein family. For investigation of signals involved in regulation of activity of auxin efflux transporters I have selected (based on published information) physiological signals influencing auxin transport. These signals were changed by treatment with chemicals and resulting changes in auxin transport were measured (on the cellular level) with the intention to discover signals that can rapidly (within minutes) change the activity of auxin efflux transporters. The signals, which satisfy these requirements were subjected to further investigation. He detailed...
Integration of elements of Physical Education and Health Physical Education into general knowledge improving subjects
Novotná, Zuzana ; Kuhnová, Věra (referee) ; Hronzová, Marie (advisor)
The aim of my master thesis is an incorporation of elements of Physical Education and Health Physical Education into regular subjects. It includes utilization of different areas of a school building during teaching. The purpose of a theoretical part is to explain concepts related to movement, children of elementary school age, Physical Education and Health Physical Education. Therefore in my research I focused besides healthy children on also children with movement disorder, and children with study problems. In attachment you can find a methodical stack of different exercises for children during teaching and during breaks as well as with usage of different areas and tools. Moreover it includes pictures of children doing exercises and pictures of different exercisers which can be placed inside or outside of a school building. Last part is an evaluation of these exercises by children themselves.
Strategie rozvoje problémových oblastí městské části Brno - Židenice
Novotná, Zuzana
This diploma thesis deals with marketing strategic development in problem areas of the municipal district Brno-Židenice. These key areas were determined based on analytical processing of outputs obtained from the questionnaire survey and SWOT analysis. The main aim of the diploma thesis is to propose concrete measures that will contribute to the future development of the municipal district Brno-Židenice. Planned development assets are proposed with regard to the competences conferred upon the Municipal city of Brno. These activities are complemented by the recommended funding options.
Proposal of the Motivation System in the Company
Novotná, Zuzana ; Kaletová, Kateřina (referee) ; Novák, Petr (advisor)
This thesis focuses on the proposal of the motivation system in newly formed company. The current state of this company is analyzed using interviews with employees. Based on the results of analysis the new motivation system of the company is designed. This motivation system should increase employee motivation and lead to their total satisfaction.
