Kopecký, Jiří ; Dostál, Lukáš (referee) ; Veselka, František (advisor)
This work deals with the use of the light microscope in industry and its use within the automatization of the diagnostic method specialized on electrical machines with a sliding contact. The method is called the dust particles method and it is used to determine the loss of the brushes used in the machines. The current method involves a manual evaluation of an acquired data, which demands additional tests and the evaluation itself is very difficult. The objective of this work is to simplify and accelerate the method using the computer analysis. First, the usage of the light microscope, through which the samples are obtained, is showed. The method uses these samples as the input. Then, the principles of the dust particles method are demonstrated and the image analysis using the MATLAB program follows. The guide to the image analysis with the elimination of rising defects using the MATLAB program is available. Then, the volume of the intercepted dust particles is calculated using the MATLAB program. The dust particles were obtained from the LFC 55h brush using the Arsenal LPT 1513 – T light microscope.

Czech Travels to Richard Wagner
Fránek, Michal ; Kopecký, J.
The study explores the phenomenon of Czech travels to famous German composer Richard Wagner and to the places associated with his cult (Munich, Bayreuth, Zurich, etc.). It focuses on the Wagnerian journeys – accomplished and not – of Czech composers (Bedřich Smetana, Antonín Dvořák, Zdeněk Fibich, Josef Bohuslav Foerster), performers (Ema Destinnová, Karel Burian, etc.), music critics (Otakar Hostinský, V. V. Zelený), painters (Jaroslav Čermák) and writers (Julius Zeyer, Jaroslav Maria, Teréza Nováková), tracing their different motivations.
Utilization of dynamic mechanical analysis as an add-on module of oscillatory rheometry for hydrogels
Dále pak byly diskutovány výsledky z měření pomocí oscilační smykové reometrie a následné porovnání výsledků mezi dynamickou mechanickou analýzou a oscilační smykovou reometrií.
Hydraulic analysis of the Damnice water supply system
Bergerová, Marie ; Kopecký, Josef (referee) ; Ručka, Jan (advisor)
This thesis deals with hydraulic analysis of the proposed concept of connecting the Damnice group water supply systém to the Znojmo group water supply system. In firt part, the issue of drought and the related threat to the supply of drinking water are presented. The second part is focused on the basics of mathematical water supply network modelling and software program EPANET 2.2 used for modelling is described. In the practical part, the focus area is characterised and the process of building the model in EPANT 2.2 is clarified. A hydraulic analysis was processed for simulating four variants that represented the connection of different count of unicipalities to the proposed group water supply system. For individual variants, the optimal pipe dimension of the main supply pipe as well as the branching water pipes along the route were sought. In processing the results of the hydraulic analysis, pressure ratios, flow rates, water age in the water supply network and the reserve capacities of the water tanks were assessed.
Environmental drivers of microbial community composition in a temperate forest
Martinović, Tijana ; Baldrian, Petr (advisor) ; Vašutová, Martina (referee) ; Kopecký, Jan (referee)
Recently, much attention has been focused on the importance of forests to the global carbon cycle. Microorganisms are a critical component of the carbon cycle in forests, participating in important ecosystem processes and degrading various carbon compounds, from simple to highly recalcitrant compounds. However, most studies in this regard have focused on fungi. Only in recent years has it become known that bacteria are also capable of degrading complex plant polymers in soil, and that their contribution to the carbon cycle may be as important as that of fungi. Furthermore, our knowledge of the temporal dynamics of both fungal and bacterial communities is limited, as well as their response to different environmental drivers. The main objective of this dissertation was to fill these knowledge gaps by (1) quantifying the rate of temporal turnover of fungal and bacterial communities in soils of temperate forests, (2) describing the factors that shape fungal communities in spruce and beech stands during their long-term development (stand age), (3) assessing the contribution of fungi and bacteria to the utilisation of different carbon sources in forest soils, and finally (4) describing the development of bacterial communities in response to clearcutting of a spruce stand. The study of microbes...
Microbial communities of soils affected by long-term heavy metal contamination
Průchová, Pavla ; Kopecký, Jan (advisor) ; Matyska Lišková, Petra (referee)
This work is focused on microbial communities living in the soil affected by long-term exposure to heavy metals. The soil was sampled at two sites with different levels of contamination near Příbram. In the samples, respiration rate was measured in vitro after addition of carbon sources and at different levels of cadmium, one of the contaminating metals found in the soil. After the incubation with carbon sources, soil samples were collected for environmental DNA isolation. Gene coding for 16S rRNA in Actinobacteria was amplified from the environmental DNA samples and the amplicon composition was assessed by terminal restriction fragment length polymorphism analysis. The resulting profiles were used to compare actinobacterial communities in both groups of soil samples and in individual treatments. The analysis showed a clear distinction between the two sites differing in the contamination level and shifts in the community composition of various intesity depending on the added substrate.
