This bachelor thesis deals with the situation of milk commodity on the Czech market in selected years. The first part of this thesis describes basic theoretical issues concerning agrarian markets, agribusiness, milk as a source of nutrition and cattle as a source of dairy raw material. In the second part of the thesis, information on cattle numbers, yield, consumption and production of basic dairy products and prices of these dairy products is elaborated. Furthermore, information on milk production is elaborated within individual regions, the whole Czech Republic and selected countries of the European Union. The last part is devoted to foreign trade, where the issue of import and export is described in particular. The final part also graphically depicts the foreign trade of various categories of dairy products.
Case Study of Physiotherapy Treatment of Patient with the Diagnosis brain stroke

Title: Case study of physiotherapy treatment of patient with the diagnosis brain stroke Aims: This work aims to review current litreture concerning brain stroke and its treatement. Another purpose is to apply physiotherapeutic techniques to the patient after a stroke and evaluate their effect. Summary: The main topic of this bachelor thesis is physiotherapy in patient with a stroke. It is devided into two parts, where general section describes the definition of stroke, anatomy of the brain vessels anatomy, prevention andepidemiology, clinical manifestations. The key chapter concerns non-pharmacological treatment and commonly used rehabilitation aproaches in these conditions. The practical part focuses on clinical assessment of the patient after brain stroke including taking a detailed medical history, performing applied kinesiology and evaluating functional impairment. Aim: The aim of this bachelor thesis is to study and analyse theoretical knowledge about diagnosis of systemic sclerosis and to describe a case report concerned with a patient diagnosed with this diseas. Summary: This bachelor thesis deals with the diagnosis of systemic sclerosis. Theoretical part describes detailed information about systemic sclerosis and its classification into rheumatic disease classification system. Furthermore, the review describes theaetiology of the disease, symptoms and available treatment (specifically non-pharmacological treatment including physiotherapy). Use of the nanofiber scaffold for transfer of stem cells onto the injured ocular surface in mouse experimental model

Corneal damage is one of the most common causes of impaired vision or even blindness. When the injury is more extensive and the limbal region is involved, the natural regeneration of the cornea is not sufficient. Such damage can lead to the limbal stem cell deficiency (LSCD). The only option for LSCD treatment is transplantation of the limbal tissue or a transfer of limbal stem cells (LSCs) cultured from the healthy eye. The allogenic transplantation of the limbus or cultivated LSCs with a systemic administration of immunosuppressive drugs is needed in the case of bilateral LSCD. Nevertheless, the cell therapy is very promising approach for LSCD treatment. Transplantation of mesenchymal stem cells (MSCs) seeded on an appropriate scaffold turned out to be a suitable therapy of the LSCD. In our experimental model of LSCD we use nanofiber scaffold for MSC and LSC cultivation and for transplantation of these cells onto the chemically injured mouse eye. MSCs have immunosuppressive and immunomodulatory properties. We showed that MSCs have the ability to inhibit production of molecules associated with the inflammation and support epithelial regeneration in the damaged cornea. These inhibitory properties were confirmed in both in vitro and in vivo mouse model. Results thus showed beneficial effects of stem cell transplantation for murine corneal healing and for suppression of a local immune reaction which can impede the healing process. Such similarity of in vivo and in vitro results allows us further experiments to clarify mechanisms of MSC regenerative and healing properties after the transplantation onto the injured cornea.
Nanofiber scaffolds for local delivery of stem cells and immunosuppressive drugs for therapeutic purposes

Cell-based therapy of local tissue injuries or damages requires application of stem cells and inhibition of harmful inflammatory reaction which could impede the healing process. To increase the effectiveness of this therapy, a local administration of drugs can avoid their side effects associated with a systemic treatment. A local therapy requires suitable carriers, which can transfer the cells and drugs to the site of injury. As a promising carriers turned out nanofiber scaffolds prepared by electrospinning technology from various types of polymers. The main advantage of this technology is a possibility to define properties of nanofiber scaffolds, optimal for the growth and transfer of stem cells, and which could incorporate various types of immunosuppressive drugs. Here we describe the formation and use of nanofiber scaffolds prepared by needleless electrospinning technology from poly (L-lactic acid) (PLA) which are loaded with immunosuppressive drug Cyclosporine A (CsA). We show that CsA-loaded nanofibers effectively and selectively inhibit proliferation of activated T cells and suppress the production of T cell cytokines in vitro. Simultaneously, these nanofiber scaffolds enable growth of mesenchymal stem cells (MSCs) and thus can serve as stem cell carriers. Moreover, using an experimental mouse model of skin transplantation, we showed that covering skin allografts with MSC-seeded and CsA-loaded nanofibers significantly inhibited the local production of pro-inflammatory cytokines IL-2, IL-17 and IFN-gamma, and supported healing. Thus, nanofiber scaffolds seeded with stem cells and loaded with CsA can serve as carriers of cells and drugs for a local cell therapy and for simultaneous effective immunosuppression.
Immunomodulatory mechanisms of stem cells and their use for therapy of ocular disorders

Stem cell-based therapy represents a perspective approach for the treatment of many so far incurable diseases. Mesenchymal stem cells (MSC) are currently the most studied stem cells. They are able to differentiate into different cell types, to produce growth and trophic factors and can suppress the functions of cells of the immune system. During the study of the immunomodulatory properties of MSC, we focused on their effect on B cells. The mechanism of impact of interferon-γ (IFN-γ) on MSC and their effect on the production of interleukin 10 (IL-10) by B cells was analysed. We have demonstrated that MSC-treated with IFN-γ inhibit production of IL-10 by activated B cells via the cyclooxygenase-2 involving pathway. Due to their regenerative and immunomodulatory properties, MSC can be for treatment of many diseases. In this study we focused on the disease and damage of the eye. The limbal stem cells (LSC) are used for the treatment of damaged ocular surface, however their isolation is difficult and they can not be used in all cases of damage. Objectives: The aim of this thesis is an introduction and summary of theoretical and practical knowledge about reverse shoulder replacement and a case study of patient with with the diagnosis. Methods: This bachelor thesis has been devided into two main parts, the general part and the special part. The general part includes theoretical knowledge related to the topic of reverse shoulder replacement and the therapeutic methods used in physiotherapy. The special part includes a case report of the patient with reverse total endoprosthesis of the shoulder joint. This part contains kinesiological examination, diagnostic and therapeutic management and the final assessment of the physiotherapeutic effect. Clinical applications of stem cells for the treatment of neurodegenerative diseases of CNS

Stem cells have a huge therapeutic potential due to their ability to differentiate in multiple tissues. They could be used for neurodegenerative diseases treatment, which are typical for loss of specific groups of neurons, progressive course and lack of effective treatment due to their complicated pathophysiology, only therapies for elongation and simplification of patients' life are available. This thesis summarizes results of completed clinical studies and informs about ongoing studies, in which stem cell treatments are used for selected neurodegenerative diseases. Stem cell therapy for multiple sclerosis and amyotrophic lateral sclerosis have a long history, some of the studies has proven therapeutic efficiency of stem cells. We don't know much about effect of stem cell treatment for patients with Alzheimer's and Parkinson's diseases, because first clinical studies were finished recently. But all clinical trials have proven safety of stem cell treatment. Replacement of damaged neurons haven't been reached yet, just protection of remaining neurons by neurotrophic and immunomodulatory factors secreted by stem cells. Sexual dysfunction and pelvic floor dysfunction in patients with systemic rheumatic diseases

Title: Sexual Dysfunction and Pelvic Floor Dysfunction in Patients with Systemic Rheumatic Diseases Objectives: To assess sexual functions, quality of life and pelvic floor function in female patients with Systemic Sclerosis (SSc) and Idiopathic Inflammatory Myopathies (IIM) compared to age-/sex-matched healthy controls (HC). Methods: In total, 41 women with SSc (mean age: 50.9, disease duration: 5.8 years), who fulfilled the ACR/EULAR 2013 classification criteria for SSc, 41 healthy controls (mean age: 50.9) without rheumatic diseases, 22 women with IIM [mean age: 55.1, disease duration: 7.9 years, dermatomyositis (DM, 8)/ polymyositis (PM, 10)/ necrotizing myopathy (IMNM, 3)/ inclusion body myositis (IBM, 1)], who fulfilled the Bohan/Peter 1975 diagnostic criteria for DM/PM, and 22 healthy controls (mean age: 55.1 years) filled in 12 well-established and validated questionnaires assessing sexual function/quality of life, pelvic floor function, fatigue, physical activity and depression. Results: Compared to HC, patients with SSc and IIM had significantly higher prevalence and greater severity of sexual dysfunction (FSFI, BISF-W: in all subscales as well as total scores), dysfunction of pelvic floor (PISQ-12), and worse sexual quality of life (SQoL-F).
