Does the pre-mRNA splicing occur in S. cerevisiae co- or post-transcriptionally?

Until recently, the splicing and transcription were seen as almost independent processes. However, today a lot of studies provide plenty of evidence about their connection, even in the yeast Saccharomyces cerevisiae. The connection of these processes is particularly mediated by C-terminal domain of RNA polymerase II, which is consisted of tandemly repeated heptapeptide sequence - YSPTSPS. Amino acid residues of this heptapeptide sequence are specifically phosphorylated during transcription, which regulates transcription process and also the binding of specific factors. These factors are necessary for processing of the nascent transcript. Modifications of the primary transcript occur especially cotranscriptionally in higher eukaryotes, thus before the transcription is terminated and also before the functional mRNA is released. Opinion on cotranscriptional splicing in S. cerevisiae were significantly changed in the last years. However, nowadays the splicing of pre-mRNA of most genes in S. cerevisiae is seen as cotranscriptional process. RNA polymerase II pauses within the terminal exons and this pausing event provides sufficient time for each spliceosomal component to assemble on the pre-mRNA and also for catalysis of splicing before the transcription termination. Methods using preference in pain research

The thesis deals with neuropathic pain measurement in laboratory mice and rats. Neuropathic pain is characterized besides evoked allodynia and hyperalgesia also by spontaneous pain and anhedonia, further motivational-affective part of pain is highlighted. Traditional pain threshold testing is not appropriate method to measure these aspects of pain. Therefore, the aim of the thesis is to describe preference methods in pain research. We focused on three methods - conditioned place preference, sucrose preference test and thermal place preference. The first mentioned method creates an association between pain relief and a specific place, which afterwards the animal prefers. The conditioned place preference can be used to detect spontaneous pain. Sucrose preference test monitors decrease in consumption of sugar water after exposure to chronic pain, which can be interpreted as a sign of depression, so thanks to this method we have the opportunity to explore the interaction between pain and depression. The thermal place preference method studies changes of preferences between the cold and hot plate after induction of pain or analgesics. The findings based on preference methods help us to examine all aspects of neuropathic pain and develop more options for pain treatment.
Cartographic and geoinformatic evaluation of current bathymetric maps.

Cartographic and geoinformatic evaluation of current bathymetric maps Abstract This thesis is focusing on methods used in the process of creating bathymetric maps of lakes. In the first part of the thesis are described methods used for shoreline mapping and possibilities of depth measurements. In the theoretical part are also described interpolation methods which could be applied on point datasets. At first is given general description of these methods and after that follows also the detailed description of these methods in ArcGIS. Next part of the thesis describes cartographic methods used in bathymetry and in this part are also evaluated current bathymetric maps. The main discussion in this part is regarding cartographic and geoinformatic methods used in the process of creation these maps. In the last part is suggested a new attitude in this process and there are also created bathymetric maps using new combination of cartographic methods. Fire sport and its training

Objectives: The main objective of this work is to characterized fire sport in details and create a list of particular exercises aim to help in training in two fire sport disciplines. This sport is continually developing and is suitable for people of almost any age. The complex of exercises designated for the 100 m sprint with hurdles and for the climb to training tower's 4th floor with hook shaped ladder represents also means of training the fitness condition needed for other fire sport disciplines. Methodology: Collecting data from literature, coaches, sportsmen and its subsequent theoretical processing. Results: The result of this work is fire sport overall history and development. Introduction of particular disciplines and material equipment. Demonstration of training sessions in various part of yearly training cycle, formation of exercise collection with theirs description and usage in the training cycle. Exercise collection contains 11 exercises for running the 100m hurdles and 18 exercises for output to 4 floor training tower displayed on kinograms. A 22 modified exercises for the 100m hurdles and 18 for output to the tower. Key words: fire sport, special exercises, fitness training

Modelling of subterranean features in GIS: case study of Amerika quarries Abstract Goal of this bachelor thesis is to create description of modelling methodology of subterranean features by using geoinformation systems. Modeling concerns locations of quarries called Amerika in Český kras. There is system of mine drifts and adits, which is relict of limestone mining in this area. Main task in the work was to creat 3D model of underground drifts and tunnels. During the creation were collected data about this location, those were described in detail, and evaluated for suitability in model creation. Created model was used in another output: Thematic map, which is describing state of surface and subsurface at the same time. V Prahe, dňa 17. 8. 2014 Bakalárska práca sa venuje tématu umeleckého archívu s dôrazom na archív Júliusa Kollera. Zaoberá sa obecnými diskurzívnymi rámcami poňatia archívu a špecifickou podobou, ktorú nadobúda v umeleckej produkcii. Ukazuje Kollerovu archívnu činnosť ako celok s jeho konceptuálnou tvorbou a v kontexte seba-historizačných praktík, typických pre situáciu zemí bývalého východného bloku. Popisuje podobné stratégie v činnosti skupiny IRWIN, Museum of American Art v Berlíne, Lia Perjovschi, J. H. Kocman, KwieKulik, Artpool Art Research Center, Tamás St. Auby a Ilja Kabakov. Large-scale rock portrayal using both hand drawing and digital cartography Abstract This paper describes creation of the larg-scale topographic map for Svatý Jan pod Skalou in the Czech karst. Detailed informations about rock formations had been obtained and the rocks were processed into hachures. Rock hachures had been made manually and then transformed to digital form. Operational objectiv is to get to know various styles of rock drawing (including žebříčková manýra, swiss style) in order to desribe practical aspects of drawing. These and other foundation materials (including airborne laser scanning data) are used for construction of detailed map of research location. Moral criticism of art in the concept of Noël Carroll and Berys Gaut

In this thesis I focus on the moral criticism of art in the conception of two american aestheticians, who came to the conclusion that it is a legitimate approach in the evaluation of art. Namely, I address the conception of Noël Carroll and his concept of moderate moralism and the conception of Berys Gaut and his concept of ethicism. I present these against the background of other approaches with which it is possible to evaluate a work of art. Moral criticism is explained through the relation of aesthetic and moral values, and the relationship of art and its reception. The work focuses on the views of Carroll and Gaut on cognitive gain from art and how it is connected with considerations of morality.
Reflection of Realism in Serbian drama

The aim of thesis is the most precisely analyze and map the Serbian dramatic enviroment during the 19th century, mainly the activity of realism period. The thesis tries to show on the beginnings, the key realistic elements and the most important features of this activity. The thesis is also focused on the main representatives and works of Branislav Nušić and Petar Kočić,. The basic of thesis is also grasp and find the main themes of plays or clear up where and under which circumstances dramatics were looking for inspiration for their work and also with the regard on European context.
