Vysoké procento úmrtnosti spojené s kardiovaskulárními onemocněními podtrhuje obrovský význam studia regenerace srdeční svaloviny. Zajímavým přístupem pro regeneraci srdce může být použití buněčné kultury progenitorů Sertoliho buněk a peritubulárních myoidních buněk odvozené z varlat juvenilních samců Xenopus tropicalis, která byla označena jako XtiSC. U těchto buněk bylo zjištěno, že sdílí řadu vlastností s mezenchymálními kmenovými buňkami. Právě mezenchymální kmenové buňky jsou známé pro své imunomodulační a protizánětlivé schopnosti podporující regeneraci srdce. Cílem této práce bylo ověřit imunomodulační potenciál XtiSC v rámci regenerace srdce po amputaci apexu srdce na modelu Xenopus tropicalis za nepřítomnosti makrofágů. Důvodem deplece byl předpoklad, že XtiSC se v mnoha aspektech chovají jako makrofágy. Jako nástroj pro depleci makrofágů byl vybrán klodronát enkapsulovaný do liposomů. Bylo potvrzeno, že XtiSC mohou pozitivně ovlivňovat regeneraci srdce jak v klasickém modelu poranění srdce, tak i v modelu s depletovanými makrofágy. Bylo odhaleno, že dospělci Xenopus tropicalis s depletovanými makrofágy, kterým byly 3 dny před apikální resekcí injikovány XtiSC do kosterního svalu zadní končetiny, vykazovaly 7 dní po poškození snížené ukládání fibronektinu a zlepšení regenerace srdeční... The high percentage of mortality associated with cardiovascular diseases shows the importance of studying cardiac muscle regeneration. An interesting approach for heart regeneration may be the use of a cell culture of Sertoli progenitor cells and peritubular myoid cells derived from juvenile testes of Xenopus tropicalis that was named XtiSC. These cells were found to share a number of characteristics which are typical for mesenchymal stem cells. Mesenchymal stems cells are well-known for their immunomodulatory and anti-inflammatory abilities that promote heart regeneration. The goal of this master thesis was to test the immunomodulatory potential of Sertoli progenitor cells during heart regeneration after heart apex amputation in Xenopus tropicalis without macrophages. The reason for macrophages depletion was the assumption that XtiSC behave like macrophages in many aspects. Clodronate encapsulated in liposomes was chosen as a tool for macrophage depletion. It was confirmed that XtiSC can positively influence heart regeneration in a classic model of heart regeneration, but also in a model with depletion of macrophages. It was revealed that Xenopus tropicalis adults without macrophages injected with XtiCS into the hindlimb skeletal muscle 3 days before apical resection showed reduced fibronectin...
