Práce se zabývala optimalizací metody pro stanovení 10 těkavých látek vyskytujících se jako minoritní složky v alkoholických produktech. Kvantifikace byla provedena metodou interního standardu, přičemž byly porovnávány výsledky s použitím klasického interního standardu 1-pentanolu a navrhovaného ethanolu. V případě ethanolu byla očekávána lepší opakovatelnost a přesnost výsledků, neboť tato metoda nevyžaduje úpravu vzorku. Optimalizovaná měřicí metoda byla pro oba interní standardy porovnána sledováním těchto parametrů - opakovatelnost, výtěžnost, linearita a limity detekce a kvantifikace. Bylo zjištěno, že, dle očekávání, poskytuje metoda navrhovaného interního standardu ethanolu správnější výsledky pro analýzu reálných vzorků alkoholických nápojů. Nicméně ne ve všech parametrech předčila metodu klasického interního standardu. Ta poskytla lepší výsledky pro opakovatelnost a linearitu sady proměřených kalibračních roztoků, a to i přes to, že musel být každý vzorek předem upravován. Dále bylo zjištěno, že pro analýzu pomocí SPME se nehodí vzorky s vysokým obsahem ethanolu, případně jiných těkavých látek, protože vlákno pak ztrácí svou schopnost sorbovat látky ve stejných poměrech a kvantitativní analýzu není možné spolehlivě provést. This bachelor thesis dealt with the optimization of a method for the determination of 10 volatile compounds occurring as minor components in alcoholic products. The quantification was performed by the internal standard method, comparing the results using 1-pentanol and ethanol as classical and proposed internal standards, respectively. In the case of ethanol usage, better repeatability and accuracy of the results were expected as this method does not require any sample pre-treatment. The optimized measurement method was monitored for both internal standards, while following these parameters - repeatability, recovery, linearity and limits of detection and quantification. It was found out that, as expected, the method of the proposed ethanol internal standard gives more accurate results for the analysis of real alcoholic beverage samples. However, it did not outperform the classical internal standard method in all parameters. That one gave better results for repeatability and linearity for the set of calibration solutions measured, despite the fact that each sample had to be pre-treated.
