Tato diplomová práce se zabývá studiem difuzních vlastností vezikulárních systémů tvořených ze dvou opačně nabitých tenzidů SDS a CTAB s přídavkem dvouřetězcového tenzidu DODAC a cholesterolu. Cílem práce bylo vybrat vhodné fluorescenční sondy a prozkoumat jejich využití při zobrazování těchto systémů pomocí metody dynamického rozptylu světla a fluorescenční korelační spektroskopie. K připraveným systémům také byly přidány různé koncentrace hyaluronanu. Jako fluorescenční sondy byly vybrány dva zástupci karbokyaninových (DiI, DiO) a dva xanthenových (F16, RBOE) barviv. U všech sond, s výjimkou RBOE, došlo k ověření vhodnosti jejich použití. Zároveň došlo k porovnání naměřených výsledků z obou metod. Bylo zjištěno, že spolu data nekorelují, z důvodu značné složitosti analyzovaných systémů a dostupné matematické modely k prokládání FCS křivek jsou tak nedostačující This master's thesis deals with the study of the diffusion properties of vesicular systems composed of two oppositely charged tensides SDS and CTAB with the addition of the double-chain tenside DODAC and cholesterol. The aim of this work was to select suitable fluorescent probes and investigate their use in imaging these systems using dynamic light scattering and fluorescence correlation spectroscopy. Different concentrations of hyaluronan were also added to the prepared systems. Two representatives of carbocyanine (DiI, DiO) and two xanthene (F16, RBOE) dyes were selected as fluorescent probes. The suitability of their use was verified for all probes except RBOE. The measurement results from the two methods were also compared. It was found that the data do not correlate with each other, due to the considerable complexity of the analyzed systems and the available mathematical models to fit the FCS curves are therefore insufficient.
