Ovariální karcinom je jednou z nejzávažnějších gynekologických malignit na světě. Jeho vysoká mortalita je způsobena především vznikem mnohočetné lékové rezistence na kon- venční léčbu zahrnující taxany. Současný výzkum se proto zaměřuje na studium nových derivátů na bázi taxanů, které by mohly obcházet doposud známé mechanismy lékové rezistence. Existuje několik slibných derivátů taxanů z řady tzv. Stony Brook taxanů (SB-T taxanů), jejichž účinnost, toxicita a mechanismus působení jsou sledovány. Tato bakalářská práce se zabývá studiem změn exprese genů účastnících se vzniku rezistence v tkáních a nádorech získaných z in vivo modelů po podání derivátu SB-T-121605 v porovnání s konvenčním taxanem paklitaxelem a s neléčenými modely. Byly studovány vybrané geny ABC transporterů (ABCB1, ABCC1 a ABCG2) a enzymů metabolizujících taxany (CYP3A11, CYP3A13 a CYP2C29) a jejich myších ortologů ve vzorcích jater, střev a nádorové tkáně odebraných z in vivo modelů myších xenograftů. Nádorová tkáň byla odvozena od vysoce rezistentní buněčné linie NCI/ADR-RES ovari- álního karcinomu. Stanovení genové exprese bylo provedeno metodou kvantitativní po- lymerázové řetězové reakce v reálném čase. Podání derivátu SB-T-121605 vedlo k několika významným změnám genové ex- prese. V nádorové i střevní tkáni došlo po podání... Ovarian carcinoma is one of the most serious gynecological malignancies in the world. High mortality of this cancer is mainly due to the development of multiple drug resistance to conventional treatments involving taxanes. Therefore, current research is focused on the study of new taxane-based derivatives capable to overcome known mechanisms of drug resistance. At present, there are several promising Stony Brook taxane derivatives (SB-T-taxanes), investigated in frame of efficacy, toxicity and mechanism of action. The aim of this bachelor thesis was to study changes in the expression of genes involved in the development of resistance in tissues and tumor obtained from in vivo mo- dels after the administration of derivatives SB-T-121605 compared to conventional taxane paclitaxel and non-treated models. Expression of selected ABC transporter genes (ABCB1, ABCC1 and ABCG2), taxane metabolizing enzyme genes (CYP3A11, CYP3A13 and CYP2C29) and their mouse orthologs was estimated in liver, intestinal and tumor tissue samples taken from in vivo mouse xenograft models. Tumor tissue was derivated from the highly resistant NCI/ADR-RES ovarian cancer cell line. Gene expression was determined by real-time quantitative polymerase chain reaction.
