Černý kašel neboli pertuse je nakažlivé bakteriální onemocnění dýchacích cest způsobené bakterií B. pertussis. Navzdory velké proočkovanosti populace ve vyspělých zemích, patří černý kašel k nejrozšířenějším onemocněním, kterému lze předcházet očkováním. Bakterie B. pertussis produkuje celou řadu toxinů, které usnadňují osídlení a pomnožení bakteriálních buněk na hostitelském řasinkovém epitelu horních cest dýchacích. Jedním z těchto toxinů je adenylát cyklázový toxin, který je z bakteriálních buněk uvolňován pomocí sekrečního aparátu typu I a po jeho vápníkem-indukovaném sbalení je schopen dopravit přes cytoplasmatickou membránu hostitelských buněk svoji N-koncovou adenylát cyklázovou doménu, která po interakci s cytozolickým kalmodulinem katalyzuje přeměnu adenosintrifosfátu (ATP) na cyklický adenosinmonofosfát (cAMP). Vysoké hladiny cAMP vedou následně k poruchám buněčné signalizace a k zásadnímu omezení baktericidních funkcí fagocytujících buněk. Cílem této bakalářské práce bylo připravit a charakterizovat 'hybridní' proteinové konstrukty, které se skládají z C-koncové části acylační domény a N-koncové části RTX domény (v pozicích 881-1038 a 881-1047) napojené na C-koncovou část RTX domény (v pozici 1562-1681) nesoucí strukturní informaci pro inicializaci správného sbalení proteinové molekuly.... Whooping cough or pertussis is a contagious bacterial airways diasease caused by B. pertussis. Despite the high vaccination coverage of the population in developed countries, whooping cough is one of the most common diseases that can be prevented by vaccination. Bacteria B. pertussis produces a variety of toxins that facilitate the colonization and proliferation of bacterial cells in the host ciliary epithelium of the upper respiratory tract. One of these toxins is adenylate cyclase toxin, which is released from bacterial cells by a type I secretory apparatus and, after it's calcium-induced packaging, is able to transport its N-terminal adenylate cyclase domain across the cytoplasmic membrane of host cells. This domain able to catalyse conversion of adenosine triphosphate (ATP) to a cyclic adenosine monophosphate (cAMP) after the interaction with cytoplasmatic calmodulin. High levels of cAMP subsequently lead to impaired cell signaling and to a fundamental reduction in the bactericidal functions of phagocytic cells.
