UNIVERZTA KARLOVA Farmaceutická fakulta v Hradci Králové Katedra farmaceutické technologie Autorka: Simona Kodytková Název diplomové práce: Disoluce léčiva z tenkých filmů na bázi plastifikovaných PLGA derivátů Školitel: PharmDr. Eva Šnejdrová, Ph.D. Cílem předložené práce bylo formulovat kapalné soustavy pro tvorbu in situ tenkých filmů (FFS) na bázi kopolymerů kyseliny D,L-mléčné a glykolové (PLGA) a studovat vliv kombinace kyseliny salicylové a methyl-salicylátu ve filmu a vliv jejich různých koncentrací na disoluční profily. V teoretické části byly shrnuty obecné poznatky o charakteristikách, látkách používaných k formulaci FFS a metody jejich testování. V experimentální části byly připraveny kapalné formulace tvořící in situ filmy evaporací rozpouštědla, založené na lineárním nebo větveném PLGA polymeru s kyselinou salicylovou jako léčivem obtížně rozpustným ve vodě, a s methyl-salicylátem ve funkci multifunkčního plastifikátoru, a to v různých koncentracích. Byl proveden test rozpustnosti kyseliny salicylové, methyl-salicylátu a jejich směsi a disoluční testy léčiva a plastifikátoru do tlumivého fosforečnanového pufru pH 5,5 při 37 ᵒC. Analýza salicylátů byla provedena metodou HPLC. Na základě výsledků lze konstatovat, že rychlejší disoluce kyseliny salicylové a methyl-salicylátu byla... The theoretical section summarizes the general knowledge about the characteristics, substances used to formulate FFS and methods for their testing. In the experimental section, liquid systems forming in situ films by solvent evaporation were prepared, based on a linear or branched PLGA polymer containing salicylic acid as a poor water soluble drug, and methyl-salicylate as a multifuctional plasticizer, at different concentrations. The solubility test of salicylic acid, methyl salicylate and their mixtures, and dissolution tests of drug and plasticizer in phosphate buffer buffer pH 5.5 at 37 řC were performed.
