Cílem této práce bylo porovnání separačních možností různých typů chromatografických kolon pro stanovení neopterinu, kynureninu, kreatininu a kyseliny močové ve vzorku moči s metodou, která je v současné době využívána v laboratoři Gerontologické a metabolické kliniky Fakultní nemocnice v Hradci Králové pro stanovení požadovaných analytů. Základní, již vyvinutou a validovanou metodou , byla metoda vysokoúčinné kapalinové chromatografie (HPLC) na reverzní fázi s kolonou Gemini 5 um C18 110Ǻ, 150 x 3 mm, Phenomenex (Torrance, USA). Jako mobilní fáze byl použit fosforečnanový pufr o koncentraci 15 mmol/l a pH 6,4 s průtokem 0,8 ml/ min. Pro kreatinin, kynurenin a kyselinu močovou byla použita detekce při vlnových délkách 235 nm pro kreatinin, 230 nm pro kynurenin a 290 nm pro kyselinu močovou. Neopterin byl detekován fluorescenčně za použití 353 nm excitační vlnové délky a 438 nm emisní vlnové délky. Všechny chromatografické systémy byly porovnány na základě testu vhodnosti systému (SST - System Suitability Test), byla zhodnocena jejich separační účinnost, rozlišení píků, asymetrie a ostatní parametry. Kolony s nevhodnějšími parametry byly vybrány pro další optimalizaci a validaci, jejímž výsledkem by měla být nová moderní HPLC metoda pro klinické využití. The aim of this diploma thesis was to compare the separative options of different types of chromatographic columns for the determination of neopterin, kynurenine, creatinine and uric acid in urine samples with the method that is currently used in the Research laboratory of the Department of Methabolic Care and Gerontology, Teaching Hospital in Hradec Kralove. Basic, already developed and validated method was High Performance Liquid Chromatography (HPLC) on reversed-phase column Gemini 5 μm C18 110Ǻ, 150 x 3 mm, Phenomenex (Torrance, USA). Phosphate buffer with concentration of 15 mmol/l, pH 6,4 and flow rate 0,8 ml/min was used as a mobile phase. Diode array detector was used for detection of creatinine (235 nm ) , kynurenine (230 nm) and uric acid (290 nm) Neopterin was monitored using fluorescent detection at 353 nm excitation and 438 nm emission wavelength. All chromatographic systems were compared on the basis of the SST- System Suitability Test and were evaluated by the separation efficiency, peak resolution, asymmetry and other parameters. Columns with the best parameters were selected for further optimization and validation. The resulting new modern HPLC method should be suitable for clinical use.
