[cze][eng] Souhrn Úvod: Fodynamická antimikrobní terapie (PACT) představuje potenciální léčebnou metodu k terapii povrchových infekcí. Vzrůstající rezistence mikrobů k antibiotikům vede k hledání nových alternativních eradikačních postupů, jako je PACT. Zatímco grampozitivní bakterie jsou k fotoinaktivaci obvykle citlivé, gramnegativní bakterie a mikroskopické houby bývají odolnější. Výzkum na poli PACT vede k hledání účinných antimikrobních preparátů s širokým spektrem postihujícím bakterie i mykotická agens. Ftalocyaniny představují slibnou skupinu fotosenzitivních látek, které by se mohly vykazovat signifikantní antimikrobní fotoinaktivační efekt. Cílem této práce bylo otestovat antimikrobiální efekt 15 derivátů ftalocyaninů (Pc) při fotoinaktivaci. Metoda: fotoinaktivační schopnost 15 Pc byla testována na kmenech Staphylococcus aureus, Escherichia coli a Candida albicans. Po 30 - minutové inkubaci s Pc v koncentraci: 1 mg/l, 2 mg/l, 4 mg/l, 8 mg/l, byly kultury ozářeny světlem z low-power laseru (o vlnové délce 670 nm) v celkové dávce 20 J/cm2 a 40 J/cm2 ). Účinnost při fotoinaktivaci byla hodnocena velikostí řádového poklesu viabilních bakterií v poměru ke kontrolnímu vzorku bez ozáření a Pc. Výsledky: Osm z testovaných ftalocyaninů bylo efektivních vůči S. aureus, ale jen čtyři deriváty působily vůči E. coli a...Photodynamic antimicrobial therapy Background: Photodynamic antimicrobial therapy is proposed as a potential topical, noninvasive approach suitable for treatment of locally occuring infection. The fact that bacteria are becoming increasingly resistant to antibiotics and antiseptics has lead to an increased interest in the field of developing new alternative eradication methods, such as PACT. Whereas gram-positive bacteria are generally sensitive to photoinactivation, gram-negative bacteria and fungi are usually less susceptible. Research of photosensitizers (PS) as well as development has been going on with the aim to find effective antimicrobial substances, which would have a broad-spectrum potency. The aim of this paper is to evaluate the antibacterial and antifungal effect of fifteen Pc derivatives. Methods: Fifteen different Pc were investigated. Their photokilling activity was tested on Staphylococcus aureus, Escherichia coli and Candida albicans. After treating of microbial cells with Pc in the concentrations: 1 mg/l, 2 mg/l, 4 mg/l, 8 mg/l for 30 minutes, the cultures were irradiated with low-power laser light wavelength of 670 nm (20 J/cm2 , 40 J/cm2 ).
