Léčivé přípravky analyzované v této práci byly extrakt ze senny starý více než 200 let, mast "Naso-Merfen" stará 75 let a mast "Sulfathiazol" stará 42 let. Účinnými látkami v léčivých přípravcích byly sennosidy A a B (extrakt ze senny), efedrin a menthol (mast "Naso-Merfen") a sulfathiazol (mast "Sulfathiazol"). Extrakt ze senny byl analyzován metodami RP-HPLC a HPLC-MS. Podmínky separace byly optimalizovány zejména pro separaci enantiomerů sennosidu A a sennosidu B. Účinné látky nebyly ve vzorku detegovány. Identifikován byl jejich degradační produkt a několik sloučenin typických pro sennu a byl porovnán jejich výskyt v historickém a novodobém vzorku. Navrženo bylo detailní schéma ESI− -MSn mechanismu fragmentace sennosidu A a B. Extrakt vzorku masti "Naso-Merfen" byl analyzován metodami HILIC-UV, HPLC-MS, GC-MS a AAS. Podmínky separace byly optimalizovány. Účinné látky byly kvantifikovány. Nebyly identifikovány žádné degradační produkty účinných látek. Extrat vzorku masti "Sulfathiazol" byl analyzován metodami RP-HPLC a HPLC-MS. Podmínky separace byly optimalizovány. Účinná látka byla kvantifikována. Nebyly nalezené žádné degradační produkty účinné látky. Byla potvrzena autenticita všech analyzovaných vzorků. Historical pharmaceutical preparations analyzed in this thesis were a senna extract more than 200 years old, an ointment "Naso-Merfen" 75 years old, and an ointment "Sulfathiazol" 42 years old. The active substances in the analyzed samples were sennosides A and B (senna extract), ephedrine and menthol (the ointment "Naso-Merfen"), and sulfathiazole (the ointment "Sulfathiazol"). The senna extract was analyzed by RP-HPLC and HPLC-MS. Separation conditions were optimized, especially for separation of the sennoside A and B enantiomers. The active substances were not detected in the sample. One degradation product and substances characteristic for senna were identified. Their presence in the historical and contemporary sample was compared. Detailed ESI− -MSn fragmentation mechanisms of sennoside A and B have been proposed. The sample of the ointment "Naso-Merfen" was analyzed by HILIC-UV, HPLC-MS, GC-MS, and AAS. Separation conditions were optimized. The active substances were quantified.
