PharmDr. Přemysl Mladěnka, Ph.D. Název diplomové práce: Příprava metodiky pro zjištění míry chelatace iontů vápníku a hořčíku Vápník a hořčík jsou biogenní prvky patřící do druhé skupiny periodické tabulky prvků. Vápník je pátým nejzastoupenějším prvkem v lidském těle. Kromě své důležité role v homeostáze kostí se podílí na řadě procesů jako srážení krve, svalové kontrakci a regulaci nervové excitability. Hořčík je kofaktorem mnoha enzymatických reakcí nezbytných pro řadu fyziologických funkcí. Cílem této práce bylo navrhnout novou screeningovou metodu pro stanovení míry chelatace iontů vápníku a hořčíku. V prvním kroku byla změřena UV-VIS spektra vybraného indikátoru o-kresolftalein komplexonu a jeho komplexů s vápníkem a hořčíkem. Následovalo ověření linearity a stanovení citlivosti metody. Současně byla měřena stabilita indikátoru a jeho komplexu s oběma ionty. V posledním kroku byla ověřena vhodnost metody na standardním chelátoru kyselině ethylendiamintetraoctové, (EDTA). Optimální vlnová délka pro komplex s vápníkem byla 572,5 nm a pro komplex s hořčíkem 568 nm. U těchto vlnových délek byla za použití indikátoru rozpuštěného v pufru o pH 7,5 linearita mezi... Calcium is the fifth most abundant element in the human body. In addition to its important role in bone homeostasis, calcium is involved in other processes such as blood clotting, muscle contraction and regulation of nerve excitability Magnesium is the cofactor of many enzymatic reactions essential for a row of physiological functions. The aim of this work was to propose a novel screening method for determination of the degree of calcium and magnesium chelation. In the first step the UV-vis spectra of the selected indicator o-cresolphthalein complexone and its complexes with calcium and magnesium were measured. This was followed by verification of the linearity and the determination of the sensitivity of the method. Concurrently the stability of the indicator and its complexes with both ions was measured.
