Abstrakt: [cze][eng] Fakultativní vnitrobuněčné patogeny si vyvinuly schopnost růst a přežít mimo i uvnitř buňky, což jim dává selektivní výhodu, lepší přístup k živinám, ochranu před extracelulárními složkami imunitního systému a nestálými podmínkami mimo buňku. Bakterie Francisella tularensis a Listeria monocytogenes se umí ubránit prvotním obranným reakcím imunitního systému, vstoupit dovnitř makrofágů i neprofesionálních fagocytů jako jsou například hepatocyty či endotel, předejít baktericidním procesům ve fagosomu, rozmnožit se v cytosolu a poté vzdorovat procesům buněčné smrti dokud samy nejsou připraveny infikovanou buňku opustit a napadnout další buňky. Toto přemístění mezi buňkami používá Listeria monocytogenes G-aktin hostitelské buňky, zatímco u bakterie Francisella tularensis není mechanismus zcela jasný. Popis mechanismu těchto procesů u patogenů Francisella tularensis a Listeria monocytogenes je předmětem této bakalářské práce. This is giving them a selective advatage and also better access to nutrients, protection from extracellular components of immune system and variable conditions outside of the cell. Bacteria Francisella tularensis and Listeria monocytogenes are able to resist the defence mechanisms of immune system, enter the macrophages and non-profesional phagocytes for example hepatocytes or endothelium, prevent bactericidal processes in phagosome, replicate in cytosol and resist cell death processes until they are ready to leave the infected cell and spread to another cell. For this displacement between cells is Listeria monocytogenes using actin, whereas in Francisella tularensis the mechanism is not entirely clear. The description of the mechanism of these processes in pathogens Francisella tularensis and Listeria monocytogenes is the subject of this bachelor thesis.
