Nepatrná část prakticky nekonečného sekvenčního prostoru dosáhla během evoluce obrovské funkční rozmanitosti proteinů. Jako nejpravděpodobnější předchůdci dnešních proteinů jsou díky konformační rozmanitosti a funkční promiskuitě považovány vnitřně neuspořádané proteiny (IDPs), které nezaujímají plně definovanou trojrozměrnou strukturu. Jak se však tyto proteiny dále vyvíjely do mnohdy rigidních a vysoce specializovaných proteinových struktur? Tato evoluční trajektorie nachází největší podporu v teorii indukovaného foldu, podle které byl vývoj struktury zprostředkován interakcí a koevolucí primordiálních nestrukturovaných proteinů s různými kofaktory či molekulami RNA. Ačkoli jsou některé náhodné sekvence z přírodou nevyužívaného sekvenčního prostoru také schopny tvořit funkční proteiny se sekundární a terciální strukturou, jako vhodnější kandidáti pro vývoj struktury a funkce se dnes zdají být náhodné sekvence s vysokým obsahem neuspořádanosti, které mají nižší tendenci k agregaci. Právě vybrané náhodné proteinové sekvence s vysokým obsahem neuspořádanosti byly v této práci strukturně charakterizovány pro jejich další využití při evolučních studiích. Na základě předchozí studie v naší laboratoři byly vybrány tři arteficiální proteiny z knihovny náhodných sekvencí. Ty byly v předkládané práci... An infinitesimal fraction of the practically infinite sequence space has achieved enormous functional diversity of proteins during evolution. Intrinsically disordered proteins (IDPs) which lack a fully defined three-dimensional structure are the most likely precursors to today's proteins because of their flexible conformation and functional diversity. But how have these proteins evolved into often rigid and highly specialized protein structures? This evolutionary trajectory has the greatest support in the theory of induced fold whereby the development of the structure was mediated by the interaction and coevolution of primordial unstructured proteins with different cofactors or RNA molecules. Although some random sequences from the sequence space which is not used by nature are also able to form folded proteins the more suitable candidates for evolution of structure and function appear to be random sequences with a high content of disordered which have low aggregation propensity. The selected random protein sequences with high disorder content have been structurally characterized in this work for their further use in evolutionary studies.
