RNA vazebný protein Whi3p v kvasinkové buňce ovlivňuje celou řadu dějů, z nichž nejvýznamnější je regulace velikosti kvasinkové buňky při vstupu do S fáze. V nedávné době byla objevena souvislost mezi proteinem Whi3p a ploidií kvasinkové buňky. Bylo prokázáno, že v kmenech s absencí genu WHI3 (whi3Δ) dochází ke zvýšení ploidie. Protein Whi3p totiž ovlivňuje expresi genů kódujících podjednotky kohezinu a rovněž reguluje hladinu Nip100p na posttranskripční úrovni. Nip100p je důležitou součástí dynaktinového komplexu. Obě možnosti regulace úzce souvisejí se správným rozchodem chromozómů v průběhu anafáze mitotického dělení. Microarrayové a imunoprecipitační analýzy prokazují interakci Whi3p s více než 400 mRNA. Translací těchto cílových mRNA vznikají strukturní i regulační proteiny esenciální pro standardní průběh jaderného dělení. Konkrétní důsledky interakcí Whi3p s cílovými RNA nejsou známy, nicméně poukazují na dosud neznámou úlohu proteinu Whi3p při mitotickém dělení pučících kvasinek. Klíčová slova: Whi3p, RNA vazebný protein, dynaktinový komplex, regulační mechanismy v anafázi, Saccharomyces cerevisiae RNA binding protein Whi3p in the yeast cell affects a variety of processes, the most important is the regulation of the yeast cell size when entering the S phase. Recently, a link between Whi3p protein and yeast cell ploidy has been discovered. It has been shown that in strains lacking the WHI3 gene (whi3Δ) there is an increase in ploidy. The Whi3p protein affects the expression of genes coding cohesin subunits and also regulates an amount of Nip100p on the post-transcriptional level. Nip100p is an important part of the dynactin complex. Both possibilities of regulation are closely related to the correct chromosome distribution during anaphase of the mitotic division. Microarray and immunoprecipitation analyzes demonstrate Whi3p interaction with more than 400 mRNAs. By translating these target mRNAs both structural and regulatory proteins, that are essential for the standard course of nuclear division, are formed. The specific consequences of Whi3p interactions with target RNAs are not known, however, they point to the unknown role of Whi3p protein in the mitotic division of budding yeast.
