Tato bakalá ská práce je úvodem do projektu zabývajícího se vlivem evoluce genetického kódu, tedy i v ase se m nícího repertoáru aminokyselin v proteinech, na vývoj struktury a funkce bílkovin. Prvotní aminokyselinová abeceda byla z ejm pouze polovi ní velikosti abecedy sou asné a zahrnovala podle dostupných analýz okolo 10 prebioticky p ítomných aminokyselin, pocházejících z exogenních i endogenních zdroj . Obsahem této práce je mRNA transkripce a bezbun ná exprese p ti randomizovaných proteinových knihoven (obsahujících zhruba 1011 variant). Uvedené knihovny se liší svým aminokyselinovým složením a odráží tak jednotlivé p edpokládané fáze evoluce genetického kódu. Hlavním cílem tedy je p evést knihovny z DNA formy do formy protein a poskytnout tak zdroj pro purifikaci knihoven a jejich následnou biofyzikální charakterizaci (výskyt sekundární a terciární struktury) a analýzy výskytu funkce v takto navržených knihovnách. Celý výzkum by tak teoreticky mohl p isp t k zodpov zení základních otázek nejen z oboru proteinové evoluce, ale i syntetické biologie i proteinového inženýrství. Klí ová slova: Bezbun ná exprese, proteiny, evoluce genetického kódu This bachelor thesis is an introduction to a project dealing with the influence of genetic code evolution (and the consequentamino acid repertoire in proteins) on the evolution of protein structure and function. The initial amino acid alphabet was apparently only half the size of the present alphabet and according to available metaanalyses it included only about 10 prebiotically present amino acids derived from potential exogenous or endogenous sources. This work includes mRNA transcription and cell-free expression of five randomized protein libraries (containing approximately 1011 variants). These libraries differ in their amino acid composition and reflect individual predicted stages of genetic code evolution. The main goal is to convert libraries from DNA to protein form to provide a source for library purification and their subsequent biophysical (occurrence of secondary and tertiary structure) and functional analyses. The whole research could theoretically contribute to a few basic questions not only in the fields of protein evolution but also in areas of synthetic biology or protein engineering.
