Cytochromy P450 jsou významné enzymy účastnící se metabolismu endogenních i exogenních látek. Jejich substrátem jsou také karcinogeny, které mohou aktivovat, čímž může docházet ke vzniku rakoviny. Pokud jsou tedy cytochromy P450, zodpovědné za aktivaci karcinogenů indukovány, riziko rakoviny se zvyšuje. Jedním z induktorů těchto enzymů jsou i chemopreventivní látky, které jsou v současnosti velmi oblíbené a doporučované. Proto by měl být sledován vliv chemopreventivních látek na cytochromy P450 a vznik rakoviny. Cytochromy P450 se nejčastěji vyskytují v játrech. Nicméně existují také formy, které nebyly dosud prokázány v žádné zdravé lidské tkáni, ale jejich zvýšená exprese byla zjištěna v nádorech. Z tohoto důvodu by mohly sloužit k diagnostice a prognóze rakoviny. Mezi tyto cytochromy patří CYP2S1 a 2W1, které mohou být prognostickými markery kolorektální rakoviny. Proto by bylo vhodné mít nástroje k detekci těchto enzymů. Jednou z možností je imunodetekce cytochromů P450 technikou "Western blot" pomocí specifických protilátek. Nyní jsou nejčastěji využívány savčí protilátky (IgG), avšak protilátky izolované z vaječného žloutku (IgY) získávají na popularitě zejména kvůli velkému množství nesporných výhod. Pro přípravu peptidických imunogenů byly nejprve z primární struktury CYP2S1 a 2W1 vybrány... The cytochromes P450 are enzymes participating in metabolism of endogenous and exogenous compounds. Their substrates include also carcinogens which may initiate carcinogenesis after activation by CYP450. Inductors of these enzymes are also chemopreventive compounds which are very popular and recommended in current time. Thus, studying of the effect of the chemopreventive compounds on cytochromes P450 induction and cancer development is of a high clinical importance. The CYPs are most commonly found in the liver. However, there are forms that have not been detected in any human healthy tissue but their overexpression was observed in tumors. For this reason, they could serve for diagnosis and prognosis of cancer. Among these cytochromes are CYP2S1 and 2W1 which can be prognostic markers of colorectal cancer. Therefore, it would be opportune to have some tools for these enzyme detection. One option is immunodetection of cytochromes P450 by Western blot using the specific antibodies. Today mammalian antibodies (IgG) are the most widely used but antibodies isolated from egg yolk (IgY) become popular mainly due to the large number of undisputed advantages.
