[cze][eng] Práce se věnuje stanovení metodiky pro analýzu dermální permeace a absorpce kofeinu in vitro. Zajištění standardních postupů při provádění experimentu je zásadní pro správné vyhodnocení a interpretaci výsledků. V první části se práce věnuje možnostem a způsobu transportu látek přes kožní bariéru. S narůstajícími výhodami, které poskytuje transdermální podání léčiv, se navyšuje počet studií zabývajících se možnostmi podání léčiv právě touto cestou. Na druhé straně lze také sledovat nebo předvídat riziko intoxikace po vystavení jedince různým druhům chemikálií. K získání reprodukovatelných výsledků je zapotřebí stanovit standardní metodiku. V této práci byl zvolen za modelovou hydrofilní sloučeninu kofein. Kožní bariéru představovala dermatomovaná kůže prasete o dvou rozdílných tloušťkách z oblasti břicha a ucha. Za použití Franzových cel bylo provedeno celkem 8 pokusů s různými typy kůže a to ve variantách finite a infinite dávkování. Experimentální část práce se zaměřuje na vlastní stanovení prostupu kofeinu. Měření se provádělo jak ve vlastní receptorové tekutině, tak v jednotlivých vrstvách kůže a na jejím povrchu. Ke stanovení bylo použito metod spektrofotometrie a HPLC. Byla potvrzena schopnost kofeinu permeovat všemi vrstvami kůže a získané výsledky jsou interpretovány jako množství...This thesis aims to the assessment of methods used in in vitro analysis of dermal permeation and absorption of caffeine. Guarantee of standard processes during an experiment is essential for the rightness of results evaluation and interpretation. First part is focused on alternatives and different transport ways of substances through the skin barrier. As transdermal drug delivery offers an increasing number of advantages, the number of studies performing transdermal experiments with drug delivery grows up. On the other hand we are also able to observe or predict the risk of intoxication of individuals after exposure to different kinds of chemicals. The standard methods assessment is of great importance to obtain reproducible results. In this thesis, caffeine was chosen as a model hydrophilic compound. Skin barrier was simulated by dermatomed pig skin of two different thicknesses from ear and abdomen. Eight experimets were carried out with different types of skin using Franz cells in two types of dosing - finite and infinite. Experimental part of the thesis aims on actual determination of caffeine permeation . Measurements were taken in receptor fluid, in particular layers of the skin and on its surface.
