PharmDr. Martin Doležal, Ph.D. Název disertační práce: Vztah mezi strukturou a biologickou aktivitou nových modulátorů acetylcholinesterasy Práce se zabývá přípravou periferně působících inhibitorů acetylcholinesterasy jako potenciálních léčiv onemocnění myasthenia gravis. Tyto látky mohou být použity také jako profylaktika před otravou ireverzibilními inhibitory AChE. Dokáží obsadit aktivní místo enzymu a zabránit tak přístupu ireverzibilního inhibitoru. Požadavky na vlastnosti těchto látek byly dostatečná inhibiční účinnost (k porovnání byly vybrány standardy používané v současné době v terapii myasthenia gravis), dostatečně selektivní působení proti AChE a nesnadná prostupnost hematoencefalickou barierou pro snížení případných nežádoucích účinků. Bylo připraveno celkem šest sérií biskvartérních sloučenin. In vitro byla stanovena jejich inhibiční účinnost proti lidské erytrocytální popř. rekombinantní AChE a lidské plazmatické BChE. V rámci každé série byla na základě získaných hodnot provedena studie vztahů mezi strukturou a účinkem. Podařilo se připravit několik sloučenin, které prokázaly nezanedbatelný inhibiční potenciál. PharmDr. Martin Doležal, Ph.D. Title of Doctoral Thesis: Relationship between structure and biological activity of new modulators of acetylcholinesterase This work focuses on the preparation of peripherally acting AChE inhibitors as a potential treatment of myasthenia gravis. These compounds could also be used as a pre- treatment of poisoning by organophosphorus AChE inhibitors. They can occupy the active site of an enzyme making it inaccessible for irreversible inhibitors. Compounds used in this indication should fulfill these requirements: inhibit efficiently AChE (standards currently used in the treatment were chosen for the comparison of the inhibitory efficacy), have selective effect on AChE and not to cross the blood-brain barrier in order to lower the possible side effects. Six series of bisquaternary compounds were prepared. Their inhibitory ability was determined in vitro against human erythrocyte or recombinant AChE and human plasmatic BChE.
