Cílem této práce bylo připravit syntetickou sekvenci 5'UTR sestřihové varianty Var 14 glutamátkarboxypeptidasy II (GCPII). K tomu byla použita metoda dvoukrokové PCA. Sekvence byla rozdělena do 8 překrývajících se oligonukleotidů a spojena do jedné dsDNA pomocí dvou po sobě jdoucích PCR. Produkt syntézy byl zaklonován do pomocného vektoru pUC19. Po jeho osekvenování byly opraveny nalezené mutace. Produkt byl subklonován do cílového vektoru pcDNA4 His Var 14 již obsahujícího sekvenci vlastního genu GCPII. Tento konstrukt byl následně použit k sestrojení kalibrační křivky, která bude i nadále sloužit jako standard pro RT-PCR pro zjištění kvantity této varianty GCP II u pacientů s rakovinou prostaty. Konstrukt bude také používán jako expresní vektor pro produkci Var 14 varianty GCPII v eukaryontním baculovirovém expresním systému. Klíčová slova: 5'UTR, dvoukroková PCA, pUC19, RT-PCR, GCPII The aim of this work was to prepare the synthetic 5'UTR sequence of splicing variant Var 14 of glutamate carboxypeptidase II (GCPII). A method of two-step PCA was used to this purpose. The sequence was divided into 8 overlapping oligonucleotides that were combined into a single dsDNA by two consecutive PCR. Product of the synthesis was cloned into the auxilliary cloning vector pUC19. After the sequencing analysis detected mutations were corrected. The product was subcloned into the target vector pcDNA4 His Var 14 which already contained the sequence GCPII gene. This construct was then used for the construction of the calibration curve, which will serve as a standard for RT-PCR for quantitative detection of this variant of GCP II in patients with prostate cancer.
