Cytochrom P450 je významný enzym katalyzující velké množství reakcí metabolismu endogenních i exogenních látek. Existují látky, které zvyšují expresi tohoto enzymu, což může vést k aktivaci karcinogenů a vzniku rakoviny. Mezi tyto látky patří i dnes velmi propagované chemopreventivní látky, které jsou masově užívané ve formě potravinových doplňků. Je tedy důležité mít k dispozici metody k určení indukce cytochromu P450. Jednou z takových metod je imunodetekce cytochromu P450 metodou Western blotting. K detekci proteinů na membráně slouží specifické protilátky získané většinou ze savců. V této bakalářské práci je dokázáno, že stejně dobře však mohou posloužit i protilátky získané ze slepičích vajec. Těmito vaječnými protilátkami byly detekovány cytochromy P450 1A1 a 1A2 v jaterních mikrosomech po exposici potkanů modelové chemopreventivní látce β-naftoflavonu. Cytochrome P450 is an important enzyme which catalyzes a large amount of reactions of endogenous and exogenous metabolism. There are compounds which increase expression of this enzyme and it leads up to the carcinogen activation and cancer formation. It includes much propagated chemopreventive compounds which are consumed abundantly in dietary supplements. It is therefore important to have the methods for determination of cytochrome P450 induction. One of these methods is the cytochrome P450 immunodetection by Western blotting. The detection of proteins on the membrane is done by the specific antibodies mostly gained from mammals. In this bachelor thesis there is the evidence that the antibodies from hen egg serve as well as the mammalian antibodies. The cytochromes P450 1A1 and 1A2 was detected by these egg antibodies after rat exposure to the model chemopreventive compounds β-naphthoflavone.
